Unveiling the role of miRNAs from MSC-EVs in neuroinflammation and behavioral impairments induced by chronic alcohol consumption

<p><strong>Part of the project: "Unveiling the role of miRNAs from MSC-EVs in neuroinflammation and behavioral impairments induced by chronic alcohol consumption."</strong></p> <p>This dataset contains small RNA sequencing data derived from extracellular vesicles (EVs) isolated from human adipose tissue-derived mesenchymal stem cells (MSC-EVs). The data was generated to study the potential therapeutic effects of repeated intravenous administration of MSC-EVs on neuroinflammation, cognitive dysfunction, and addictive-like behaviors induced by chronic alcohol consumption.</p> <p>Total RNA from MSC-EVs was extracted using the Total Exosome RNA Isolation Kit (Invitrogen). Small RNA libraries were constructed with the Small RNA-Seq Library Prep Kit (Lexogen) and sequenced on the Illumina NextSeq 550 platform with a 1 x 150 bp read length. Genetic architecture of alcohol consumption identified by a genotype-stratified GWAS, and impact on esophageal cancer risk in Japanese

<p><span>An East Asian-specific variant on <em>aldehyde</em> <em>dehydrogenase</em> <em>2</em> (<em>ALDH2</em> rs671, G&gt;A) is the major genetic determinant of alcohol consumption. We performed an rs671 genotype-stratified genome-wide association study (GWAS) meta-analysis in up to 175,672 Japanese individuals to uncover additional loci associated with alcohol consumption in an rs671-dependent manner. Three loci (<em>GCKR</em>, <em>KLB</em>, and <em>ADH1B</em>)</span> <span>satisfied the genome-wide significance threshold in wild-type homozygotes (GG), whereas six loci (<em>GCKR</em>, <em>ADH1B</em>, <em>ALDH1B1</em>, <em>ALDH1A1</em>, <em>ALDH2</em>, and <em>GOT2</em>) did so in heterozygotes (GA). Of these, five loci showed genome-wide significant interaction with rs671. Genetic correlation analyses revealed ancestry-specific genetic architecture in heterozygotes. Subsequent polygenic risk scoring depicted interactions highlighted by stratified GWAS. Further, most discovered loci showed significant effects on risk of esophageal cancer, a representative alcohol-related disease, and multiple other phenotypes. Our results identify the genotype-specific genetic architecture of alcohol consumption and reveal its potential impact on alcohol-related disease risk.</span></p>

Data from: Predictors of alcohol consumption among in-school adolescents in the Central Region of Ghana: a baseline information for developing cognitive-behavioural interventions

Background and purpose: Despite a recent shift in school going adolescents' engagement in health compromising behaviours has serious socio-economic implications on developing societies, it is surprising that baseline information through research that would influence planned interventions is sparse. The purpose of this study was to investigate the prevalence of alcohol drinking behaviours among in-school adolescents in the Junior High Schools (JHS) in the Central Region of Ghana. Methods and results: Using a descriptive cross-sectional design, multistage sampling procedures were used to sample 1400 school going adolescents in JHS in the Central Region. Preliminary results using simple frequencies and percentages revealed 42% alcohol drinking prevalence in the region. High prevalence of drunkenness (73%, n = 405) and early exposure to alcohol drinking when students were in primary school (52%, n = 286) were noted. Community festivals and use of alcohol as a form of medicine were enabling factors of alcohol consumption in the region. Binary logistic regression analysis also showed that geographical location was a significant predictor of alcohol drinking among school going adolescents, with students in the southern and central part of the region at greater risks of drinking alcohol than those from the northern part (OR = .696, 95% CI = 0.52-926, p = .013). However, no statistical significant variations were found in the odds of drinking alcohol within age (OR = 1.13, 95% CI = 0.86-1.48, p = .370), gender (OR = .81, 95% CI = 0.65-1.01, p = .06), religious affiliation (OR = 1.33, 95% CI = 0.94-1.89, p = .10), parental communication (OR = .86, 95% CI = 0.66-1.06, p = .13), academic performance (OR = 1.07, 95% CI = 0.79-1.45, p = .05) and socioeconomic status (OR = 1.20, 95% CI = 0.95-1.53, p = .12). Conclusions: With this baseline data, it was recommended that schools' curricula should include preventive cognitive-behavioural interventions that teach drug resistance skills and anti-drug norms. These interventions would foster the development of requisite knowledge and social skills (e.g., developing competence) for resisting social and peer influences that may trigger alcohol use and perhaps other drugs. Potentially, the motivation for alcohol use among school going adolescents in the region would be minimized, if not prevented.

Effects of Alcohol Consumption on Oxidative Stress in a Sample of Patients Recruited in a Dietary Center in a Southern University Hospital: A Retrospective Study

<p><em>Background and objectives</em>: The aim of this retrospective study was to evaluate the effects of alcohol consumption on oxidative stress. <em>Materials and Methods</em>: The study was conducted by analyzing the increase in lipid peroxidation, the reduction of antioxidant defenses and the alteration of the oxidation/antioxidant balance after the administration of ethanol in 25% aqueous solution (<em>v</em>/<em>v</em>) at a concentration of 0.76 g/kg of body weight daily in two doses for 3 days. The changes in oxidative stress indices were investigated by standard methods previously described. <em>Results</em>: Ethanol administration has determined a significant increase in plasma levels of lipid hydroperoxide (LOOH), malonilaldehyde (MDA) and oxidized glutathione (GSSH), and a decrease in total antioxidant capacity (TAC), reduced glutathione (GSH) and GSH/GSSH ratio. <em>Conclusions</em>: In the proposed experimental condition, the excessive and repeated consumption of ethanol causes oxidative damage, as shown by the increase in lipid peroxidation, the reduction of antioxidant defenses and the alteration of the oxidation/antioxidant balance, which, at least in part, are responsible for the harmful effects of excess ethanol.</p>
