Reference percentiles for carotid measures of subclinical atherosclerosis in children and adolescents – Data from the KiGGS study
<p>The dataset presents reference centile data for carotid intima-media thickness as well as four carotid stiffness parameters. The reference percentiles are based on data from the German Health Interview and Examination Survey for Children and Adolescents 2003-2006 (KiGGS). KiGGS started as a cross-sectional study conducted between 2003 and 2006 which was based on a nationally representative sample and aimed at obtaining comprehensive data on the health of children and adolescents aged 0 to 17 years living in Germany. Detailed information on study design and conduct has been published in peer reviewed journals as well as detailed papers on individual parameters. The documentation provides relevant references.<br> <br> 11 years later, KiGGS Wave 2 (2014-2017) was conducted as an interview and examination survey. Carotid sonography was attempted at follow-up (KiGGS2) in all 4,798 participants of the KiGGS cohort aged 14 to 28 years. Carotid intima-media thickness (CIMT) was successfully measured in 4,709 participants. Reference centiles for the distensibility coefficient, stiffness index ß, Young's elastic modulus and Peterson's elastic modulus were computed using data on 4,305 adolescents and young adults aged 14 to 28.</p>
GWAS to single cell: Intersecting single-cell transcriptomics and genome wide association studies identifies crucial cell-populations and candidate genes for atherosclerosis.
<p><strong>Background</strong></p> <p>Genome-wide association studies (GWAS) have discovered hundreds of common genetic variants for atherosclerotic disease and cardiovascular risk factors. The translation of susceptibility loci into biological mechanisms and targets for drug discovery remains challenging. Intersecting genetic and gene expression data has led to identification of candidate genes. However, the assayed tissues are often non-diseased and heterogeneous in cell composition confounding the candidate prioritization. We collected single-cell transcriptomics (scRNA-seq) from atherosclerotic plaques and aimed to identify cell-type-specific expression of disease-associated genes. </p> <p> </p> <p><strong>Methods and Results</strong></p> <p>To identify disease-associated candidate genes, we applied gene-based analyses using GWAS summary statistics from 46 atherosclerotic, cardiometabolic, and other traits. Next we intersected these candidates with single-cell transcriptomics (scRNA-seq) to identify those genes that are specifically expressed in individual cell (sub)populations of atherosclerotic plaques. We derive an enrichment score and show that loci that associated with coronary artery disease demonstrated a prominent substrate in plaque smooth muscle cells (<em>SKI</em>, <em>KANK2</em>, <em>SORT1</em>), endothelial cells (<em>SLC44A1</em>, <em>ATP2B1</em>), and macrophages (<em>APOE</em>, <em>HNRNPUL1</em>). Further sub clustering of SMC-subtypes revealed genes in risk loci for coronary calcification specifically enriched in a synthetic cluster of SMCs. To verify the robustness of our approach, we used liver-derived scRNAseq-data and showed enrichment of circulating lipids-associated loci in hepatocytes.</p> <p><br> <strong>Conclusion</strong></p> <p>We confirm known gene-cell pairs relevant for atherosclerotic disease, and discovered novel pairs pointing to new biological mechanisms amenable for therapy. We present an intuitive single-cell transcriptomics driven workflow rooted in human large-scale genetic studies to identify putative candidate genes and affected cells associated with cardiovascular traits.</p> <p> </p>

YB1 dephosphorylation attenuates atherosclerosis by promoting CCL2 mRNA decay
<p> Y-box binding protein 1 (YB1) is an RNA binding protein (RBP) that has been reported to play important roles in inflammation and atherosclerotic plaque formation. To explore the molecular mechanism of phosphorylated YB1 (pYB1) in atherosclerosis <em>in vitro</em>, we constructed YB1 phosphorylation site (Ser-100) mutant stable smooth muscle cell line (3ds-V5) and performed RNA profile screening through RNA-seq. We found inflammatory pathways and CCL2 was significantly decreased in the 3dS-V5 YB1 mutant compared with the control group (YB1-V5).</p>

Encompassing view of spatial and single-cell RNA-seq renews the role of the microvasculature in human atherosclerosis
<p>Here we generate an integrative high-resolution map of human atherosclerotic plaques combining single-cell RNA-seq from multiple studies and spatial transcriptomics data from 12 human specimens, with different stages of atherosclerosis. We show cell-type and atherosclerosis-specific expression changes and spatially constrained alterations in cell-cell communication.</p>
