Cross cultural tears: A systematic investigation of the interpersonal effects of emotional crying across different cultural backgrounds

<p>The present project wants to examine the importance of emotional crying as an attachment behaviour and its fundamental role across a number of diverse cultures.</p> <p>Emotional tears are uniquely human and have fascinated scholars across several decades (Vingerhoets, 2013). Some researchers argue that tearful crying played a significant role in the evolution of humankind with regard to social development and solidarity (Walter, 2006). Recent years have seen an increased interest in exploring the interpersonal effects of human tears (see Gračanin, Bylsma, &amp; Vingerhoets, 2018 for a review), with findings that emotional tears foster approach or support behavior (Gračanin, Krahmer, Rinck, &amp; Vingerhoets, 2018) and crying individuals being evaluated as more communal (e.g., Zickfeld, van de Ven, Schubert, &amp; Vingerhoets, 2018). These findings generally fit the hypothesis that emotional tears constitute a social act, promote social bonding and fulfill an attachment function (Nelson, 2005; Bowlby, 1982; Gračanin, Bylsma, et al., 2018; Murube, Murube, &amp; Murube, 1999; Radcliffe-Brown, 1922; Vingerhoets, 2013). Confocal microscopy images (CZI files) of human chondrocytes in different cell culture media with stained nuclei and primary cilia

<p>1. Methods</p> <p>1.1 Cell culture</p> <p>For investigating the influence of the cell culture medium composition on the lengths of primary cilia, human non-degenerative chondrocytes from a 30-year-old male donor (NHAC-kn, CC-2550; LONZA, Walkersville Inc., Walkersville, MD, USA) were used. These chondrocytes were seeded in passage four with a density of 28000 cells/cm<sup>2</sup> on collagen-coated glass coverslips (GG-15-Collagen; Neuvitro Corporation, Camas, WA, USA). The cells were cultivated in 12-well plates (Thermo Fisher Scientific Inc., Waltham, MA, USA) under hypoxic conditions at 37&deg;C, 5% CO<sub>2</sub> and 5% O<sub>2</sub> with different media compositions for three days.<br> The basal medium consisted of Dulbecco&rsquo;s Modified Eagle Medium (DMEM) (Gibco&trade;) including high glucose (GlutaMAX&trade;), sodium pyruvate supplements (Thermo Fisher Scientific Inc., Waltham, MA, USA), as well as 1% penicillin/streptomycin (Pen/Strep; Thermo Fisher Scientific Inc.), 1% Amphotericin B (Biochrom GmbH, Berlin, Germany), and 50 &micro;g mL<sup>&minus;1</sup> ascorbic acid (Sigma-Aldrich, Merck KGaA, Darmstadt, Germany). To this basal medium, different supplements were added, creating four groups:<br> 1) ITS: 1% Insulin-Transferrin-Selenium (ITS+&trade; Premix, BD Biosciences, Franklin Lakes, NJ, USA),<br> 2) ITS with Dexa: 1% Insulin-Transferrin-Selenium (ITS+&trade;) and 100 nM dexamethasone (Sigma-Aldrich, Merck KGaA, Darmstadt, Germany),<br> 3) ITS with Dexa + IGF-1 + TGF-&beta;1: 1% Insulin-Transferrin-Selenium (ITS+&trade;), 100 nM dexamethasone, 50 ng mL<sup>&minus;1</sup> insulin-like growth factor (IGF)-1 (R&amp;D Systems, Minneapolis, MN, USA) and 50 ng mL<sup>&minus;1</sup> transforming growth factor (TGF)-&beta;1 (Peprotec, Hamburg, Germany),<br> 4) FBS: 10% fetal bovine serum (FBS, Pan Biotech, Aidenbach, Germany).</p> <p>1.2 Immunocytochemistry</p> <p>After three days of cultivation in the different media compositions, the chondrocytes were washed once with phosphate-buffered saline (PBS; Biochrom GmbH, Berlin, Germany) and fixed for 10 min at room temperature (RT) with 4% paraformaldehyde (ROTI &reg; Histofix, Carl Roth GmbH + Co. KG, Karlsruhe, Germany). After fixation, cells were washed again and permeabilized with 0.2% Triton-X100 (Merck, Darmstadt, Germany) for 10 min. For blocking the unspecific binding sites, cell-seeded coverslips were incubated with bovine serum albumin (BSA; Sigma-Aldrich) with a concentration of 5% in PBS for one hour at RT after another washing step with PBS. To stain the primary cilium, cells were incubated with anti-acetylated &alpha;-tubulin (6-11B-1) (RRID: AB 628409) labeled with Alexa Fluor 647 (sc-23950 AF647, Santa Cruz Biotechnology, Dallas, TX, USA) diluted 1:200 in PBS at 4&deg;C overnight. Additionally, the Actin cytoskeleton was stained with Acti-stain 488 Fluorescent Phalloidin (Cytoskeleton, Inc., Denver, CO, USA) diluted 10 in PBS for 30 min at RT. Afterward, cells were washed three times with PBS, and the coverslips were fixed with Fluoroshield&trade; (Sigma-Aldrich) containing 4&rsquo;,6-Diamidino-2-phenylindole (DAPI).</p> <p>1.3&nbsp;Image acquisition</p> <p>Three-dimensional fluorescence images of stained cells were acquired with a ZEISS ELYRA LSM 780 confocal laser scanning microscope (CLSM) (Carl Zeiss AG, Oberkochen, Germany). Images were recorded using a Plan-Apochromat 63&times;/1.40 Oil DIC M27 objective (Carl Zeiss AG, Oberkochen, Germany). The distance of two layers was 0.2814 &micro;m and the resolution 1024 &times; 1024 pixels (scan magnification: 0.6, pixel length: 0.2196 &micro;m).</p>

"The sound comes from a meadow in the Sierra Nevada Mountains in California. The meadow is at an elevation of 2400 meters near a mountain named Olancha Peak, which is 3700 meters in altitude. Ihave a group of friends with which Ibackpack (trek) into the mountains. Our goal was to spend some time in the mountains and hike to the top of Olancha Peak (…) By the time we reached the meadow, we were in a forest and there was still snow on the ground in some places. We took the trip in June of 2006. The Sierra Nevada Mountains are a large mountain range. Much of the range is protected by national parks or preserved areas we call 'wilderness areas' (…) Ihave been backpacking for nearly 40 years and Iwill hopefully continue with this challenging activity for 40 years more! Many of my friends are much younger than Iam and it gives me much satisfaction to be able to have as much or more stamina for this activity than they have! When we are on these trips, we hike up peaks, catch fish, drink some whiskey around campfires and enjoy our time in the beautiful solitude. My memories of this trip were of the steep, hot hike from the desert to the cool meadow; the overall beauty of the nature, the absolute solitude of our campsite near the meadow; the strenuous hike to the top of Olancha Peak; the camaraderie of my friends; and, of course the sound of the frogs in the meadow. The frog sounds were astounding to me and Iwould listen in awe of the creature's instinctual desire to reproduce and continue the existence of their kind. Surely there were different species in the meadow for some of the frog sounds were different than others. The sounds only occurred after the Sun went down for the evening. Carbon availability affects already large species-specific differences in chemical composition of ectomycorrhizal fungal mycelia in pure culture

<p>Although ectomycorrhizal (ECM) contribution to soil organic matter processes receives increased attention, little is known about fundamental differences in chemical composition among species, and how that may be affected by carbon (C) availability. Here we study how 16 species (incl. 19 isolates) grown in pure culture at three different C:N ratios (10:1, 20:1 and 40:1) vary in chemical structure, using Fourier transform infrared (FTIR) spectroscopy. We hypothesised that C availability impacts directly on chemical composition, expecting increased C availability to lead to more carbohydrates and less proteins in the mycelia. There were strong and significant effects of ECM species (R<sup>2</sup> = 0.873 and P = 0.001) and large species-specific differences in chemical composition. Chemical composition also changed significantly with C availability, and increased C led to more polysaccharides and less proteins for many species, but not all. Understanding how chemical composition change with altered C availability is a first step towards understanding their role in organic matter accumulation and decomposition.</p>

Data from: Spatial and seasonal influences on culturable endophytic mycobiota associated with different tissues of Eugenia jambolana Lam. and their antibacterial activity against MDR strains

Background: Present study focuses on diversity and distribution analysis of endophytic fungi associated with different tissues of Eugenia jambolana. The influence of season and geographical location on diversity and distribution of endophytic fungi has been analyzed. Antibacterial activity of isolated fungal species has also been investigated against MDR bacterial strains. Result: A total of 1896 endophytic fungal isolates were obtained from healthy, surface sterilized tissues of leaf, stem and petiole tissues during summer, monsoon and winter season. Out of 24 fungal species isolated, 20 species belong to class Ascomycetes, 2 to Basidiomycetes and 2 to Zygomycetes. Maximum species diversity was in rainy season whereas colonization frequency was in winter. All the diversity indices showed maximum species diversity at site 5 (Yamunanager), rainy among the seasons and leaf among the tissues studied. Aspergillus genus was most frequently isolated. Aspergillus niger and Alternaria alternata were most dominant species. Three way ANOVA results showed that effect of season was highly significant on species diversity in relation to sites and tissues. 60% endophytic fungal extracts showed significant antibacterial activity against one or more than one MDR bacterial strain. Conclusion: Different fungal species were recovered from different sites but the inter-site comparisons were not significant according to Jaccard similarity coefficient. Diversity of such fungal endophytes indicates that Eugenia jambolana plant acts as an ecosystem facilitating survival of many microbes with impressive antibacterial potential.

Integrated multitrophic culture of shrimp Litopenaeus vannamei and mullet Mugil liza in biofloc system with different feeding rates for fish

<p>This study aimed to evaluate the effect of different feeding rates for mullets on the zootechnical performance of <em>Litopenaeus vannamei</em> and <em>Mugil liza</em> in an integrated multitrophic culture of shrimp and mullet in a biofloc system and on the efficiency of the system. For this, an experimental study of 46 days was carried out at the Marine Shrimp Laboratory of the Federal University of Santa Catarina. The experiment had four treatments, each one with four replicates: A0 (feed offer for shrimp and no feed offer for mullets), A1 (feed offer for shrimp and offer of 1% of fish biomass of feed for mullets), A2 (feed offer for shrimp and offer of 2% of fish biomass as feed for mullets) and A3 (feed offer for shrimp and offer of 3% of fish biomass for feed for the mullets). Each experimental unit consisted of an 800 L tank, in which shrimp were stocked, and a 90 L tank, used for mullets, connected to a recirculation system. Shrimp were stocked with an initial weight of 2.36 g &plusmn; 0.02 g. Mullets&rsquo; inicial weight was 17.06 g &plusmn; 0.65 g. The stocking density used was 375 shrimp m<sup>-3</sup> (300 shrimp per tank), while the fish density was 167 fish m<sup>-3</sup> (15 fish per tank). Animal growth performance, water quality, as well as total heterotrophic bacteria and <em>Vibrio </em>sp<em>.</em> concentration in the system were evaluated. It was observed that shrimp performance was not affected by different feed offer for fish, having in the end a survival above 82%, weekly growth around 1.50 g and feed conversion factor close to 1.60 in all treatments. Despite the higher input of nutrients into the system with higher food supply, there was no change in the water quality parameters with the treatments, or in the count of total heterotrophic bacteria and <em>Vibrio sp.</em> <a href="#_msocom_1">[1]</a>&nbsp;In conclusion, the best feed supply rate in the integrated cultivation of shrimp and mullet was 2%, due to the proximity of the results of zootechnical performance with the 3% rate, without affecting water quality or the count of total heterotrophic bacteria and genus <em>Vibrio sp.</em></p>
