Lattice Boltzmann simulation of liquid water transport in gas diffusion layers of proton exchange membrane fuel cells: Impact of gas diffusion layer and microporous layer degradation on effective transport properties

Underlying data to publication Sarkezi-Selsky et al., J. Pow. Sour. 556 (2023) 232415, https://doi.org/10.1016/j.jpowsour.2022.232415

Polymer Electrolyte Membrane Fuel Cells (PEMFCs) represent a promising technology for clean drivetrain solutions, in particular for heavy-duty applications. However, lifetime requirements demand high durability of each cell component.
In this work, transport of liquid water through pristine and degraded gas diffusion layers (GDL) was simulated with a 3D Color-Gradient Lattice Boltzmann model. The GDL microstructure was reconstructed from high-resolution X-ray micro-computed tomography (μ-CT) of an impregnated Freudenberg H14. The effect of a microporous layer (MPL) was considered by reconstruction of an impregnated and MPL-coated H14. Aged microstructures were generated artificially, assuming loss of polytetrafluoroethylene (PTFE) within the GDL and increase of MPL macroporosity as main degradation mechanisms. Liquid water transport within aged microstructures was simulated by imposing a liquid phase flow rate until breakthrough was reached. Subsequently, the GDL microstructures were analyzed for their breakthrough characteristics by means of saturation and effective gas transport properties. When the MPL was pristine, no distinct GDL degradation effect was observable, this was attributed to the MPL dominating capillary transport. MPL aging, however, led to increased saturations and thus to a deterioration of the effective gas transport. With a partially degraded MPL, aging of the GDL then appeared to affect the breakthrough characteristics.

spindle cell variant diffuse large B-cell lymphoma (NGS annotation file; high confidence calls) hematolrep-2136295

Diffuse large B-cell lymphoma with spindle cell morphology is a rare variant. We present the case of a 74-year-old male who initially presented with a right supraclavicular (lymph) node enlargement. Histological analysis showed a proliferation of spindle-shaped cells with narrow cytoplasms. An immunohistochemical panel was used to exclude other tumors, such as melanoma, carcinoma, and sarcoma. The lymphoma was characterized by a cell-of-origin subtype of germinal center B-cell-like (GCB) based on Hans’ classifier (CD10-negative, BCL6-positive, and MUM1-negative); EBER negativity, and the absence of BCL2, BCL6, and MYC rearrangements. Mutational profiling using a custom panel of 168 genes associated with aggressive B-cell lymphomas confirmed mutations in ACTB, ARID1B, DUSP2, DTX1, HLA-B, PTEN, and TNFRSF14. Based on the LymphGen 1.0 classification tool, this case had an ST2 subtype prediction. The immune microenvironment was characterized by moderate infiltration of M2-like tumor-associated macrophages (TMAs) with positivity of CD163, CSF1R, CD85A (LILRB3), and PD-L1; moderate PD-1 positive T cells, and low FOXP3 regulatory T lymphocytes (Tregs). Immunohistochemical expression of PTX3 and TNFRSF14 was absent. Interestingly, the lymphoma cells were positive for HLA-DP-DR, IL-10, and RGS1, which are markers associated with poor prognosis in DLBCL. The patient was treated with R-CHOP therapy, and achieved a metabolically complete response.

Carreras J, Kikuti YY, Miyaoka M, Hiraiwa S, Tomita S, Ikoma H, Kondo Y, Ito A, Nagase S, Miura H, Roncador G, Colomo L, Hamoudi R, Campo E, Nakamura N. Mapping of nanoparticles diffusion in cells

We mapped the diffusion of particles in the cytoplasm. To do so, we electroporated QDs-SB particles in cells as shown in Debayle et al 2019. Here we were using RPE1 cells. We present the recording of three cells and the scripts that we used to generate some maps of diffusion in the cell.

The scripts were made to use the output tracks from Slimfast 1.103e but can be adapted by changeing the beginning of the script.

Some functions were found in Matlab Central and their corresponding license is in their respective folders.

Diffusion of nanoparticles in the cell cytoplasm

In Etoc 2018, we presented the diffusive behavior inside the cytosol of living cells for different kind of nanometric sized nanoparticles. We showed that the passivation of the particles is the main parameter that regulate the diffusion properties of particles smaller than 70nm not only in Hela cells but also in RPE1 and MSC cells.

Here we open the data for the MSC cells.

Experiments consists in the recording of small subregions of the cytoplasm located at the periphery of the cell.

A README file is included in the archive with the recording parameters.

DATA were recorded by Elie BALLOUL

Scripts were written by Mathieu COPPEY, Fred ETOC and modified by Elie BALLOUL.(to use polyfit instead of ezyfit)

We also make available the scripts that we used to generate the maps.

Some functions were found in Matlab Central and their corresponding license is in their respective folders.

Single Cell Phenotypic Profiling to Identify a Set of Immune Cell Protein Biomarkers for Relapsed and Refractory Diffuse Large B Cell Lymphoma: A Single-Center Study

Diffuse large B-cell lymphoma (DLBCL) is the most common invasive type of non-Hodgkin lymphoma. Cell-of-origin (COO) classification is related to patients’ prognoses. Primary drug resistance in treatment for DLBCL has been observed. The specific serum biomarkers in these patients who suffer from relapsed and refractory (R/R)-DLBCL remains unclear. In the current study, using single-cell RNA sequencing (scRNA-seq) and mass cytometry (CyTOF), we determined and verified immune cell biomarkers at the mRNA and protein levels in single-cell resolution from 18 diagnostic peripheral blood mononuclear cell (PBMC) specimens collected from patients with R/R DLBCL. As controls, five PBMC specimens from healthy volunteers were obtained. We identified a panel of 35 surface marker genes for the features of R/R DLBCL unique cell cluster by scRNA-seq of eight R/R DLBCL patient samples and validated its efficiency in an external cohort consisting of 10 R/R DLBCL patients by CyTOF. The cell clustering and dimension reduction were compared among R/R DLBCL samples in CyTOF Space with COO as well as the C-MYC expression designation. Immune cells from each patient occupied unique regions in the 32-dimensional phenotypic space with no apparent clustering of samples into discrete subtypes. Significant heterogeneity observed in subgroups was mainly attributed to individual differences among samples and not to expression differences in a single, homogeneous immune cell subpopulation. The marker panel showed reliability in labeling R/R DLBCL without any influence from COO stratification and C-MYC expression designation. Furthermore, we compared all the markers between R/R DLBCL and normal samples. A total of 12 biomarkers were significantly overexpressed in R/R DLBCL relative to the normal samples. Therefore, we further optimized the diagnostic biomarker panel of R/R DLBCL comprising CD82, CD55, CD36, CD63, CD59, IKZF1, CD69, CD163, CD14, CD226, CD84, and CD31. In summary, we developed a novel set of biomarkers for the diagnoses of patients with R/R DLBCL. Detections procedures at single-cell resolution provide precise biomarkers which may substantially overcome intertumoral and intratumoral heterogeneity among primary samples. The findings confirmed that each case was unique and may comprise multiple, genetically distinct subclones.

Here we uploaded the dataset of CyTOF for external validation.
