PsPM-RRM1-2: SCR, ECG, respiration and eye tracker measurements in response to electric stimulation or visual targets

This dataset includes skin conductance response (SCR), electrocardiogram (ECG), respiration and eye tracker (including pupillometry) measurements for each of 29 healthy unmedicated participants (7 males and 22 females aged 23.5 +/- 3.6 years) in response to 10 discomforting electric stimulations to the forearm (RRM1) or 10 visual targets in a visual detection task (RRM2). The sample partly overlaps with data set PsPM-FR. Some participants did not take part in RRM1 or RRM2 such that there are 25 recordings for RRM1 and 26 recordings for RRM2. Electric shock stimuli are 0.2 ms wide square current pulse repeated at 500 Hz for 500 ms and individually adjusted amplitude just below the pain threshold. Visual stimuli are red crosses (+) embedded in a white digit stream; each stimulus is presented during 200 ms and separated by a 800 ms blank interval. ITI is selected randomly on each trial from 40 s, 45 s or 50 s. A baseline period with distractors but no targets concludes experiment RRM2. (This is in contrast to the methods description in Bach et al. (2016), according to which the baseline period was randomly either in the beginning or at the end of the experiment. This discrepancy was caused by an error in the code that controlled the experiment presentation.)

PsPM-SCRV6: Skin conductance responses to pain by electric stimulation

This dataset includes skin conductance response (SCR) measurements for each of 20 healthy unmedicated participants (10 males and 10 females aged 21.8+/-3.3 years) in response to 10 discomforting electric shocks. Stimuli are 0.5ms wide square current pulse repeated at 500Hz for 100ms. Amplitude is varied (mean +/- SD: 0.78mA +/- 0.43mA). ITI is selected randomly on each trial from 29s, 34s or 39s.

Bulk RNA Seq of Electrically Stimulated Retinal Organoids

Considering the significant role played by both intrinsic and extrinsic electric fields in the growth and maturation of the central nervous system, the impact of short exposure to external electric fields on the development and differentiation of retinal organoids was investigated.Organoids derived from human embryonic stem cells at day 80, a critical time point in their differentiation and maturation, were used. A single 60-minute exposure to a distinct biphasic electrical field influenced neuronal development and increased the population of photoreceptors. Initial immunohistochemistry and qPCR studies confirmed the influence of electrical stimulation on the development and differentiation of various RO cell types. RNA sequencing data revealed elevated expression of rod photoreceptors, Müller cells, horizontal cells, and amacrine cells, alongside the downregulation of retinal pigment epithelium and retinal ganglion cell genes.Furthermore, our study demonstrated varying degrees of organoid development and maturation depending on the specific electrical field applied. These findings highlight the significant impact of extrinsic electrical fields on early retinal development and suggest that optimizing electrical field parameters could effectively address certain limitations in retinal organoid technology, potentially reducing the reliance on chemicals and small molecules.

Effect of percutaneous electrical stimulation with high-frequency alternating currents at 30 kHz on the sensory-motor system of healthy volunteers.

Unmodulated high-frequency alternating currents (HFAC) are employed for producing peripheral nerves block. HFAC have been applied in humans with frequencies up to 20 kHz, whether transcutaneously, percutaneously, or via surgically-implanted electrodes. The aim of this study was to assess the effect of percutaneous HFAC, applied with ultrasound-guided needles at 30 kHz, on the sensory-motor nerve conduction of healthy volunteers.

Research data of Calcium Imaging after electrical stimulation

For intracellular calcium ions (Ca2+) mobilization analysis by Calcium Imaging, a human osteoblast-like cell line MG-63 (ATCC ®, CRL1427™) were used [1-3]. The MG-63 cell line has similar characteristics in terms of morphological behavior, adhesion and signaling properties as primary human osteoblasts [3]. Cells were cultured at 37 °C in a humidified atmosphere (5% CO2) in Dulbecco's modified eagle medium (DMEM; Gibco) containing 10% fetal calf serum (FCS; PAA Laboratories) and 1% antibiotic (gentamicin, Ratiopharm GmbH) [1-3]. Cells in the near-confluent state (70-80%) were used for the corresponding in vitro experiments. Therefore, cells were washed with PBS, trypsinized with 0.05% trypsin/0.02% EDTA (PAA) for 5 min and then treated with medium to stop the reaction. After centrifugation, 2x106 cells / 2ml were incubated shaking in complete medium at 37°C for a complete independent experiment. For the method, 2.5x105 cells were stained per electrical stimulation setup (non-stimulated, 1 V 7.9 Hz, 1 V 20 Hz, 5 V 7.9 Hz or 5 V 20 Hz). First, a wash step was performed with PBS, and after centrifugation, sedimented cells were stained with a membrane-permeable calcium indicator fluo-3/AM (Life Technologies Corporation; 5 μM) in slightly hypotonic 4-(2-hydroxyethyl)- 1-piperazineethanesulfonic acid (HEPES) buffer [1-3] shaking at 37°C. After incubation for 40 min, cells were centrifuged and then resuspended in 250 µl of isotonic HEPES. In a 12-well plate, 750 µl of isotonic HEPES was placed. Then the stained suspended cells were added, and immediately electrical stimulation by using IonOptix chamber was started under the LSM780 (Carl Zeiss). After 5 min of adhesion phase, the global calcium signal of the attaching cells was visualized using the inverted LSM 780 with a C Apochromat 40× water immersion objective. Fluo-3/AM dye was excited with the argon ion laser at 488 nm (emission at 515 nm). A full frame (512 x 512 pixels) at maximum pinhole aperture was evaluated using Zen2011 (black edition) software (Carl Zeiss) and "time series" mode. The first time series with electrical stimulation included 150 cycles each 2 s. After this time series (total electrical stimulation of 10 min), the electrical stimulation was stopped and a second time series without electrical stimulation was started. This second time series comprised 240 cycles every 2 s. After the 90th cycle of these series, fluo-3/AM stained cells were treated with 10 mM adenosine-triphosphate (ATP; SERVA). Mean fluorescence intensity of cells was evaluated using Zen2012 software (blue edition) and Mean ROI mode for defined areas of individual single cells [1-3].

[1] Staehlke S, Koertge A, Nebe B (2015) Intracellular calcium dynamics dependent on defined microtopographical features of titanium. Data from: Internal injuries in marine fishes caught in beam trawls using electrical versus mechanical stimulations

To improve the ecological and economic sustainability in the Dutch beam trawl fishery, tickler chains were replaced by electrical pulse stimulation to drive sole (Solea solea) out of the seabed. Because electrical stimulation may cause internal injuries, we quantified this risk by sampling fish species from commercial beam trawlers and recording spinal injuries and haemorrhages from X-radiographs and autopsy. To distinguish mechanically and electrical-pulse-induced injuries, we compared injuries in ten species sampled from pulse (PUL) and tickler-chain (TCK) trawlers and four species sampled from PUL trawlers with the stimulus switched on or off. Co-occurrence of a major spinal injury and major haemorrhage at the same location was only observed in PUL samples, and were frequently (40%) observed in cod (Gadus morhua) and in low numbers (0–2%) in whiting (Merlangius merlangus), grey gurnard (Eutrigla gurnardus), and greater sandeel (Hyperoplus lanceolatus), but not in flatfishes and other species. In cod, injury occurrence correlated with fish length, with lower probabilities for small fish. Major spinal injury or major haemorrhage occurrence in PUL (range: <1–16%) was lower than in TCK (range: <1–42%) in eight of the ten species studied. Population-level consequences of pulse-induced injuries are considered negligible.

Data_Discrimination between the effects of pulsed electrical stimulation and electrochemically conditioned medium on human osteoblasts

This publication contains the original data supporting our results concerning the application of biphasic pulsed electrical stimulation on MG-63 cells. We aim to discriminate between effects of the electric field and electrochemically conditioned medium. The dataset consists of the original output files, excel sheets to consolidate the information for the experiments as well as Readme.txt files with most important information concerning the data. For additional information on the material and methods as well as results see the corresponding research article.
