Data for: Sensitive and specific detection of tumor-derived exosomes using nanopore-crystal microchips

<p>Tumor-derived exosomes (tExos) have emerged as promising circulating biomarkers for early cancer diagnosis. However, the sensitivity and specificity of existing assays often limit the clinical translation of tExos. In this study, we present a highly versatile microfluidic platform, termed the nanopore-crystal microchip (NC-Chip), for specific isolation and ultrasensitive detection of tExos in as little as 0.5 μL of plasma samples from pancreatic cancer (PC) patients. The NC-Chip incorporates a herringbone-patterned hierarchical porous hydrogel scaffold, enabling fluid manipulation, size exclusion, immunoaffinity capture, and signal amplification. These integrated features significantly enhance the sensitivity and specificity of tExos assays in complex clinical scenarios. Utilizing an eight-protein signature, the NC-Chip can distinguish patients with pancreatitis and non-metastatic PC with 100% accuracy in the training cohort and 94.9% accuracy in the validation cohort. This platform is sensitive, specific, inexpensive, and only needs small-volume samples, offering a powerful exosome-based liquid biopsy tool for early PC diagnosis.</p>

Chronic hyperadiponectinemia induced by transgenic overexpression increases plasma exosomes without significantly improving glucose and lipid metabolism

<p><span>The fat</span><span>-derived factor, adiponectin, is considered a salutary circulating factor.</span> <span>We recently demonstrated that native adiponectin binds T-cadherin and promotes intracellular biogenesis and secretion of the exosome. Exosomes play important roles in various aspects of homeostasis, including glucose and energy metabolism. However, it remains unclear whether and how the promotion of exosome production by adiponectin <em>in vivo</em> is beneficial for glucose and lipid metabolism. In the present study, overexpression of human adiponectin in mice resulted in an increased number of circulating exosomes, but it did not significantly improve glucose metabolism, change body weights, or change triglyceride clearance under a high-fat diet. Multiple small doses of streptozotocin increased blood glucose and decreased triglyceride clearance similarly in both wild-type and transgenic mice. Thus, these results indicated that human adiponectin overexpression in mice increases plasma exosomes but does not significantly influence glucose and lipid metabolism.</span></p>

<p>The pearl oyster <em>Pinctada fucata martensii</em> is an economically important species of marine pearl culture, and tissue of mantle plays an essential role in pearl formation. Here, the extracted exosomes from mantle of <em>P. f. martensii</em> were analyzed by quantitative protein TMT sequencing. We wanted to verify if exosomes are the important vehicle in the process of biomineralization (e.g., pearl and shell formation), especially the color formation in shellfish. Finally, we got some results which indicated the importance of exosomes in sides of proteins, and it can also contribute to other researches related to the exosomes or pearl oyster.</p>

Data for: Simultaneous subset tracing and miRNA profiling of tumor-derived exosomes via dual-surface-protein orthogonal barcoding

<p><span>The clinical potential of miRNA-based liquid biopsy has been largely limited by the heterogeneous sources in plasma and tedious assay processes. </span><span>Here we develop a precise and robust one-pot assay called dual-surface-protein</span><span>-guided</span><span> orthogonal recognition of tumor-derived exosomes and in-situ profiling of microRNAs (SORTER) to detect tumor-derived exosomal miRNAs and enhance the diagnostic accuracy of prostate cancer (PCa). The SORTER utilizes two allosteric aptamers against exosomal marker CD63 and tumor marker EpCAM to create an orthogonal labeling barcode and achieve selective sorting of tumor-specific exosome subtypes. Furthermore, the labeled barcode on tumor-derived exosomes initiated targeted membrane fusion with liposome probes to import miRNA detection reagents, enabling in-situ sensitive profiling of tumor-derived exosomal miRNAs. With a signature of six miRNAs, SORTER differentiated PCa and benign prostatic hyperplasia with an accuracy of 100%. Notably, the diagnostic accuracy reached 90.6% in the classification of metastatic and non-metastatic PCa. We envision that the SORTER will promote the clinical adaptability of miRNA-based liquid biopsy.</span></p>
