Species diversity and plant dominance influence grassland stability in response to extreme climatic events and anthropogenic drivers across three LTER sites: Cedar Creek, Konza Prairie, and Kellogg Biological Station, 1982-2023.

The data in this package is associated with the analysis for a manuscript titled "Multiple community properties drive ecosystem resistance and resilience to extreme climate events across mesic grasslands". Neutrophil and emergency granulopoietic drivers of sepsis immune suppression and an extreme response to infection

<p>The dysregulated host response to infection leading to organ dysfunction is highly heterogeneous. It is currently poorly delineated by sepsis as a clinical syndromic classification, thus confounding immunotherapy trials. Here we establish the pathophysiology and potential therapeutic targets of a specific extreme response to infection state (sepsis response signature SRS1), characterised by immune compromise and poor outcome. We first derive a whole blood single-cell multi-omic atlas of the sepsis response (2727,993 cells, n=39), finding an increase in IL1R2+ immature neutrophils in SRS1, which we confirmed by CyTOF and RNA-sequencing (n=53). We next uncovered high activity of neutrophil STAT3 gene expression programs in SRS1, which were shared across multiple infectioius disease settings (n=1044) irrespective of the clinical definition of the patient cohorts. We observed elevated plasma G-CSF and IL-6 in SRS1, suggesting heightened emergency granulopoiesis (EG). We therefore characterised patient and healthy control hematopoietic stem cells (HSCs) using single-cell RNA/chromatin accessibility multi-omics (29,366 cells, n=27), identifying SRS1-specific EG transcriptional skewing, together with STAT3 and EG master regulator CEBPB epigenetic signatures. Our findings establish a common cellular axis present across extreme responses to infection, reveal its hematopoietic origin, and nominate G-CSF and IL-6 as potential therapeutic targets for the SRS1 state.</p> <p>&nbsp;</p> <p>The present data deposit includes processed and quality-controlled data tables for:</p> <p>1. Comparing climatic suitability and niche distances to explain populations responses to extreme climatic events

<p><span>Habitat suitability calculated from Species Distribution Models (SDMs) has been used to assess population performance, but empirical studies have provided weak or inconclusive support to this approach. Novel approaches measuring population distances to niche centroid and margin in environmental space have been recently proposed to explain population performance, particularly when populations experience exceptional environmental conditions that may place them outside of the species niche. Here, we use data of co-occurring species' decay, gathered after an extreme drought event occurring in the SE of the Iberian Peninsula which highly affected rich semiarid shrubland communities, to compare the relationship between population decay (mortality and remaining green canopy) and (1) distances between populations' location and species niche margin and centroid in the environmental space, and (2) climatic suitability estimated from frequently used SDMs (here MaxEnt) considering both the extreme climatic episode and the average reference climatic period before this. We found that both SDMs-derived suitability and distances to species niche properly predict populations performance when considering the reference climatic period; but climatic suitability failed to predict performance considering the extreme climate period. In addition, while distance to niche margins accurately predict both mortality and remaining green canopy responses, centroid distances failed to explain mortality, suggesting that indexes containing information about the position to niche margin (inside or outside) are better to predict binary responses. We conclude that the location of populations in the environmental space is consistent with performance responses to extreme drought. Niche distances appear to be a more efficient approach than the use of climate suitability indices derived from more frequently used SDMs to explain population performance when dealing with environmental conditions that are located outside the species environmental niche. The use of this alternative metrics may be particularly useful when designing</span><span> conservation measures to mitigate impacts of shifting environmental conditions.</span></p>

Site fidelity and behavioral plasticity regulate an ungulate's response to extreme disturbance

<p>1. With rapid global change, the frequency and severity of extreme disturbance events are increasing worldwide. The ability of animal populations to survive these stochastic events depends on how individual animals respond to their altered environments, yet our understanding of the immediate and short-term responses of animals to acute disturbances remains poor.</p> <p>2. We focused on animal responses to the environmental disturbance created by megafire. Specifically, we explored the effects of the 2018 Mendocino Complex Fire in northern California, USA on the behaviour and body condition of black-tailed deer (<i>Odocoileus hemionus columbianus</i>). We predicted that deer would be displaced by the disturbance or experience high mortality post-fire if they stayed in the burn area.</p> <p>3. We used data from GPS collars on 18 individual deer to quantify patterns of home range use, movement, and habitat selection before and after the fire. We assessed changes in body condition using images from a camera trap grid. The fire burned through half of the study area, facilitating a comparison between deer in burned and unburned areas.</p> <p>4. Despite a dramatic reduction in vegetation in burned areas, deer showed high site fidelity to pre-fire home ranges, returning within hours of the fire. However, mean home range size doubled after the fire and corresponded with increased daily activity in a severely resource-depleted environment. Within their home ranges, deer also selected strongly for patches of surviving vegetation and woodland habitat, as these areas provided forage and cover in an otherwise desolate landscape. Deer body condition significantly decreased after the fire, likely as a result of a reduction in forage within their home ranges, but all collared deer survived for the duration of the study.</p> <p>5. Understanding the ways in which large mammals respond to disturbance like wildfire is increasingly important as the extent and severity of such events increases across the world. While many animals are adapted to disturbance regimes, species that exhibit high site fidelity or otherwise fixed behavioural strategies may struggle to cope with increased climate instability and associated extreme disturbance events.</p>

Data from: Tree growth responses to extreme drought after mechanical thinning and prescribed fire in a Sierra Nevada mixed-conifer forest, USA

<p class="MsoNormal">An estimated 128 M trees died during the 2012-2016 California drought, largely in the southern Sierra Nevada Range. Prescribed burning and mechanical thinning are widely used to reduce fuels and restore ecosystem properties, but it is unclear if these treatments improve tree growth and vigor during extreme drought. This study examined tree growth responses after thinning, prescribed burning, and extreme drought at the Teakettle Experimental Forest, a historically frequent fire mixed-conifer forest in the southern Sierra Nevada of California, USA. Mechanical thinning (no thin, understory thin, and overstory thin) and prescribed burning (unburned, fall burning) were implemented in 2000-2001. Using annual growth data from increment cores, over 10,000 mapped and measured trees, and lidar-derived metrics of solar radiation and topographic wetness, we had two primary questions. First, what were the growth responses to thinning and prescribed burning treatments, and did these responses persist during the 2012-2016 drought? Second, what tree-level attributes and environmental conditions influenced growth responses to treatments and drought?</p> <p class="MsoNormal">Thinning increased residual tree growth and that response persisted through extreme drought 10 -15 years after treatments. Growth responses were higher in overstory versus understory thinning, with differences between thinning types more pronounced during drought. Species-specific growth responses were strongest with overstory thinning, with sugar pine (Pinus lambertiana) and incense-cedar (Calocedrus decurrens) having higher growth responses compared to white fir (Abies concolor) and Jeffery pine (Pinus jeffreyi). For individual trees, factors associated with higher growth responses were declining pretreatment growth trend, smaller tree size, and post-treatment low neighborhood basal area. Growth responses were initially not influenced by topography, but topographic wetness became important during extreme drought. Mechanical thinning resulted in durable increases in residual tree growth rates during extreme drought over a decade after thinning occurred, indicating treatment longevity in mitigating drought stress. In contrast, tree growth did not improve after prescribed burning, likely due to fire effects that reduced surface fuels, but had little effect on reducing tree density. Thinning treatments promoted durable growth responses, but focusing on stand-level metrics may ignore important tree-level attributes such as localized competition and topography associated with higher water availability. Mechanical thinning was effective at improving growth in trees that had been experiencing declining growth trends, but was less effective in improving growth responses in large old higher ecological importance.</p>

Nanoscale Simulation of the Thylakoid Membrane Response to Extreme Temperatures

<p>The thylakoid membrane is in a temperature-sensitive equilibrium that shifts repeatedly during the life cycle in response to ambient temperature or solar irradiance.<br> Plants respond to seasonal temperature by changing their thylakoid lipid composition, while a more rapid mechanism for short-term heat exposure is required.<br> The emission of the small organic molecule isoprene has been postulated as one such possible rapid mechanism.<br> The protective mechanism of isoprene is not known, but some plants emit isoprene during periods of high-temperature stress.<br> In this work, we investigate the dynamics and structure for lipids within a thylakoid membrane at different temperatures and varied isoprene content using classical molecular dynamics simulations.<br> The results are compared with experimental findings from across the literature for temperature-dependent changes in the lipid composition and shape of thylakoids.<br> We find that the surface area, volume, and flexibility of the membrane, as well as the lipid diffusion, increase with temperature, while the membrane thickness decreases.<br> Saturated thylakoid 34:3 glycolipids derived from eukaryotic synthesis pathways exhibit significantly different dynamics than lipids from prokaryotic synthesis paths, which could explain the upregulation of specific lipid synthesis pathways at different temperatures.<br> Increasing isoprene concentration was not observed to have a significant thermoprotective effect on the thylakoid membranes.</p>

Nonlinear response of productivity to precipitation extremes in the Inner Mongolia grassland

<p>As precipitation (PPT) regimes are becoming more extreme worldwide, our understanding of how terrestrial ecosystems will respond to this aspect of climate change remains unclear. We addressed this research gap by a PPT gradient experiment that included PPT amounts simulating extremely dry and wet years in combination with a long-term (40 years) observational study in a grassland ecosystem.&nbsp;This study illustrates how a grassland responded to greater PPT variability and highlights the role of limiting resource shift in ecosystem responses to climate extremes. Thus, to accurately forecast future ecosystem feedbacks to climate change, we should take into account the negative asymmetric feature of ecosystem response to stronger climate variability.</p>
