All-atom Molecular Dynamics Simulations of Meiosis 1-associated protein (M1AP) to Investagate the Impact of Known Missense Mutations Associated with Male Infertility through Non-obstructive Azoospermia

<p>Protein structure of meiosis 1-associated protein (M1AP) was modelled by using GalaxyWeb (from Seok Lab).&nbsp;We&nbsp;used this model to investigate the impact of variants (i.e., S50P, R266Q, P389L, G317R, and L430P)&nbsp;in M1AP&nbsp;which were recently&nbsp;associated with&nbsp;non-obstructive azoospermia (NOA). NOA is a male infertility-related condition causing&nbsp;absence of sperm in the seminal fluid due to meiosis failure. We aimed to elucidate the pathogenicity mechanisms of these five missense NOA-related mutations on M1AP by performing molecular modeling and molecular dynamics (MD)&nbsp;simulations. This dataset includes the results of 1000 ns MD simulations (two repeats, each 500 ns) for each of the mutant and wild-type systems.</p> <p>Systems were prepared in Visual Molecular Dynamics (VMD 1.9.3) by placing them in a TIP3P water box with approximately 20 &Aring; thickness from the protein surface and neutralizing the system charge with 0.15 M&nbsp;KCl. Of note, only protein parts&nbsp;were kept for the submission&nbsp;to reduce the size of files.&nbsp;Nanoscale Molecular Dynamics (NAMD 2.13-CUDA) was used to perform MD simulations with CHARMM36m force field. For pressure and temperature controls, Nos&eacute;-Hoover Langevin barostat&nbsp;and Langevin thermostat&nbsp;were used. ShakeH algorithm of NAMD was applied for water molecule constraints. 12 &Aring; cut-off distance was used for van der Waals interactions. Switching function starts at 10 &Aring; and reaches zero at 14 &Aring;. Integration time-step was 2 fs. To compute the long-range Coulomb interactions, the particle-mash Ewald&nbsp;method was used. NPT ensemble was applied for whole simulations.&nbsp;Two step minimization &amp; equilibration procedure was performed: (1) 5,000-step minimization and 1 ns equilibrium with constraints on the protein; (2)&nbsp;5,000-step minimization and 1 ns equilibrium without the constraints on the protein. All related configuration files for wild-type system were also included to the dataset. Production simulations were run twice along 500 ns by using different random seeds to assign the velocities from Boltzmann distribution (total simulation time for each system was 1000 ns, which are given as 500 ns repeat 1, and 500 ns repeat 2). Ectopic expression of BBS1 rescues male infertility, but not retinal degeneration, in a BBS1 mouse model

<p>Bardet-Biedl syndrome (BBS) is a rare ciliopathy for which there are no current effective treatments. BBS is a genetically heterogeneous disease, though the M390R mutation in <i>BBS1 </i>is involved in approximately 25% of all genetic diagnoses of BBS. The principle features of BBS include retinal degeneration, obesity, male infertility, polydactyly, intellectual disability, and renal abnormalities. Patients with mutations in BBS genes often present with night blindness within the first decade of life, which progresses to complete blindness. This is due to progressive loss of photoreceptor cells. Male infertility is caused by a lack of spermatozoa flagella, rendering them immobile. In this study, we have crossed the wild-type human <i>BBS1</i> gene, driven by the CAG promoter, onto the <i>Bbs1<sup>M390R/M390R </sup></i>mouse model to determine if ectopic expression of <i>BBS1</i> rescues male infertility and retinal degeneration. qRT-PCR indicates that the <i>BBS1 </i>transgene is expressed in multiple tissues throughout the mouse, with the highest expression seen in the testes, and much lower expression in the eye and hypothalamus. Immunohistochemistry of the transgene in the eye showed little if any expression in the photoreceptor outer nuclear layer. When male <i>Bbs1<sup>M30R/M390R</sup>;BBS1<sup>TG+</sup></i>mice are housed with WT females, they are able to sire offspring, indicating that the male infertility phenotype of BBS is rescued by the transgene. Using electroretinography (ERGs) to measure retinal function and optical coherence tomography to measure retinal thickness, we show that the transgene does not confer protection against retinal degeneration in <i>Bbs1<sup>M300R/M390R</sup>;BBS1<sup>TG+ </sup></i>mice.</p>

Extra-pair copulations can insure female blue tits against male infertility

Mating outside the pair-bond occurs frequently in socially monogamous birds, but the benefits that females gain from this behaviour remain debated. One hypothesis is that females engage in extra-pair copulations (EPCs) to ensure that their clutch is fertilised in case their own mate is infertile, but evidence for this idea is scarce. We report on a case of an infertile male blue tit that bred in three successive years with three different females. In the first year, all eggs were sired by an extra-pair male whereas in the second year all eggs were unfertilised and contained no sperm. In the third year, the female produced two clutches that were fertilised by an extra-pair male, but - unlike 'normal' clutches - sperm numbers on the perivitelline membrane decreased rapidly over the laying sequence. Our findings show that blue tit females mated to an infertile male can escape reproductive failure by engaging in EPCs and support previous suggestions that EPCs in blue tits cease after the onset of egg laying.

Research funding for male reproductive health and infertility in the UK and USA [2016 – 2019]

<p>There is a paucity of data on research funding levels for male reproductive health (MRH). We investigated the research funding for MRH and infertility by examining publicly accessible webdatabases from the UK and USA government funding agencies. Information on the funding was collected from the UKRI-GTR, the NIHR's Open Data Summary, and the USA's NIH RePORT webdatabases. Funded projects between January 2016 and December 2019 were recorded and funding support was divided into three research categories: (i) male-based; (ii) female-based; and (iii) not-specified. Between January 2016 and December 2019, UK agencies awarded a total of £11,767,190 to 18 projects for male-based research and £29,850,945 to 40 projects for female-based research. There was no statistically significant difference in the median funding grant awarded within the male-based and female-based categories (p=0.56, W=392). The USA NIH funded 76 projects totalling $59,257,746 for male-based research and 99 projects totalling $83,272,898 for female-based research Again, there was no statistically significant difference in the median funding grant awarded between the two research categories (p=0.83, W=3834). This is the first study examining funding granted by main government research agencies from the UK and USA for MRH. These results should stimulate further discussion of the challenges of tackling male infertility and reproductive health disorders and formulating appropriate investment strategies.</p>

Recovery from heat-induced infertility: A study of reproductive tissue responses and fitness consequences in male Drosophila melanogaster

<p><span>The predicted temperature increase caused by climate change is a threat to biodiversity. Across animal taxa, male reproduction is often sensitive to elevated temperatures leading to fertility loss and in more adverse scenarios, this can result in sterility when males reach their upper thermal fertility limit. Here we investigate temperature-induced changes in reproductive tissues, fertility reduction, sterility, and the associated fitness loss during the subsequent recovery phase in male <em>Drosophila melanogaster</em>. We heat-stressed males during development and either allowed them to recover or not in early adulthood, while measuring several determinants of male reproductive success. We found significant differences in recovery rate, organ sizes, sperm production, and other key reproductive traits among males from our different temperature treatments.</span> <span>Sperm maturation was impaired before reaching the upper thermal sterility threshold. While some effects were reversible, this did not compensate for the fitness loss due to damage imposed during development. Surprisingly, developmental heat stress was damaging to accessory gland growth, and female post-mating responses mediated by seminal fluid proteins were impaired regardless of the possibility of recovery. </span><span>We suggest </span><span>that sub-lethal thermal sterility and the subsequent fertility reduction is caused by a combination of inefficient functionality of both the accessory gland and testes. </span></p>
