Data for: Techno-economic analysis of a novel laccase production process utilizing perennial biomass and the aqueous phase of bio-oil, Iowa, USA 2023-2025

This dataset contains the experimental design, measurements, and derived variables used to parameterize a techno‑economic model of laccase production via two‑stage solid‑state fermentation of prairie biomass with bio‑oil aqueous phase induction. It includes nutrient screening data for Pleurotus ostreatus growth on prairie biomass with alternative nitrogen sources and a corn‑steep solids dose series; factorial/response‑surface experiments varying substrate bed depth, substrate‑to‑inoculum (S:I) ratio, and pre‑induction growth time; and time‑resolved induction measurements. For each run and replicate, the data record the full set of spectrophotometric absorbances at 0–210 s, fitted slopes and r-square values, dilution and volume factors, and laccase activities normalized per mL and per gram of biomass, alongside the exact culture timings and environmental conditions used in the ABTS assay at 420 nm. Results tables provide the fitted central‑composite design model terms (coefficients, F‑statistics, and p‑values) used directly as inputs to the minimum laccase selling price (MLSP) calculations, together with the underlying per‑condition raw results.

Spectrophotometric Assay for the Detection of 2,5-Diformylfuran and Its Validation through Laccase-Mediated Oxidation of 5-Hydroxymethylfurfural

<p>Modern biocatalysis requires fast, sensitive, and efficient high-throughput screening methods to screen enzyme libraries in order to seek out novel biocatalysts or enhanced variants for the production of chemicals. For instance, the synthesis of bio-based furan compounds like 2,5-diformylfuran (DFF) from 5-hydroxymethylfurfural (HMF) via aerobic oxidation is a crucial process in industrial chemistry. Laccases, known for their mild operating conditions, independence from cofactors, and versatility with various substrates, thanks to the use of chemical mediators, are appealing candidates for catalyzing HMF oxidation. Herein, Schiff-based polymers based on the coupling of DFF and 1,4-phenylenediamine (PPD) have been used in the set-up of a novel colorimetric assay for detecting the presence of DFF in different reaction mixtures. This method may be employed for the fast screening of enzymes (Z' values ranging from 0.68 to 0.72). The sensitivity of the method has been proved, and detection (8.4 μM) and quantification (25.5 μM) limits have been calculated. Notably, the assay displayed selectivity for DFF and enabled the measurement of kinetics in DFF production from HMF using three distinct laccase–mediator systems.</p>

Laccases from Pleurotus ostreatus Applied to the Oxidation of Furfuryl Alcohol for the Synthesis of Key Compounds for Polymer Industry

<p>Laccases are oxidative enzymes with high synthetic potential. In this work, their value in biocatalysis is shown through the green and selective oxidation of furfuryl alcohol into furfural with the aid of mediators. The influence of different parameters, such as pH, enzyme/mediator composition, buffer type, cosolvent tolerance, and reaction times, is investigated. Under the optimal conditions, 20 mol&thinsp;% of TEMPO as mediator and 5.8 U&thinsp;mL<sup>&minus;1</sup> of laccases POXC and POXA1b from <em>Pleurotus ostreatus</em>, quantitative production of furfural is attained after 16 h. POXC laccase stands out for its ability to catalyze the reaction at pH 6.5, whereas POXA1b is notable for its high stability. Furfural conversions reach excellent values (95&thinsp;%) after 72 h using only 5 mol&thinsp;% of TEMPO at 100 mM. Furthermore, furfuryl alcohol bioamination is achieved by employing the amine transaminase from <em>Chromobacterium violaceum</em>, providing furfuryl amine, a key compound for the polymer industry, through a one-pot sequential approach.</p>

Bio-oil aqueous phase as a laccase inducer in Pleurotus ostreatus: laccase properties and applications, Iowa, USA 2021-2023

This dataset documents the experimental conditions, protocols, and measurements generated during the investigation of bio-oil aqueous phase (AP) as an inducer for laccase enzyme production by Pleurotus ostreatus using prairie biomass as a substrate. The data include detailed descriptions of AP preparation from corn stover pyrolysis, chemical characterization of AP and prairie biomass, and the design of two-stage solid-state fermentation experiments with variables such as AP and copper concentrations, fermentation times, and environmental parameters (pH, temperature). The dataset also contains raw and processed measurements of laccase activity, protein concentration, reducing capacity, and other relevant biochemical properties across multiple experimental runs. In addition to fermentation and enzyme production data, the dataset provides information on downstream application assays. These include quantitative data on dye decolorization (crystal violet, coomassie blue, methyl orange), saccharification yields from prairie biomass pretreated with laccase, and antibiotic detoxification assays using tetracycline.
