Biochemical and biophysical drivers of the hydrogen isotopic composition of carbohydrates and acetogenic lipids

<p>The hydrogen isotopic composition (δ2H) of plant compounds is increasingly used as a hydro-climatic proxy, however, the interpretation of δ2H values is hampered by potential co-effecting biochemical and biophysical processes. Here, we studied δ2H values of water and carbohydrates in leaves and roots, and of leaf n-alkanes, in two distinct tobacco (Nicotiana sylvestris) experiments. Large differences in plant performance and biochemistry resulted from (a) soil fertilization with varying nitrogen (N) species ratios and (b) knockout-induced starch deficiency. We observed a strong 2H-enrichment in sugars and starch with a decreasing performance induced by increasing NO3-/NH4+ ratios and starch deficiency, and from leaves to roots. However, δ2H values of cellulose and n-alkanes were less affected. We show that relative concentrations of sugars and starch, interlinked with leaf gas-exchange, shape δ2H values of carbohydrates. We thus provide novel insights into drivers of hydrogen isotopic composition of plant compounds and the mechanistic modelling of plant cellulose δ2H values.</p>

Lipid extraction alters amino acid composition and bulk, but not amino acid, carbon and nitrogen isotope values

<p>Rationale: Concerns exist over observed shifts in value and variance of nitrogen isotopes following physicochemical extraction of lipids from organic matter. The mechanisms behind these apparent changes in bulk tissue δ15N values are not fully understood yet have major implications for analytical costs and integrity of data interpretations.</p> <p>Methods: Changes in proximate analysis, amino acid composition, C:N ratios, bulk tissue and amino acid δ13C and δ15N values, and resulting isotope‐based food web metrics were compared between lipid‐intact and lipid‐extracted muscle tissue of fishes spanning &lt;1% to &gt;20% muscle fat content to identify mechanisms of nitrogen isotope fractionation associated with physicochemical lipid extraction.</p> <p>Results: Bulk δ13C and δ15N values increased and %N, C:N ratios and crude protein content decreased following lipid extraction. Resulting bulk isotope niche spacing and overlap varied significantly between lipid‐intact and lipid‐extracted tissues. While amino acid composition significantly changed during lipid extraction, particularly for lipid‐associated amino acids (e.g., Glu, Lys, Ser), individual amino acid δ13C and δ15N values, and their associated compound‐specific isotope analysis of amino acids (CSIA‐AA)‐based food web metrics, did not.</p> <p>Conclusions: Physicochemical lipid extraction caused significant tissue composition changes (e.g., leaching of amino acids and 15N‐deplete nitrogenous waste) that affected δ13C and δ15N values and tissue %C and %N beyond simply removing lipids. However, lipid extraction did not alter individual amino acid δ13C or δ15N values or their associated CSIA‐AA‐based food web metrics.</p>

Data from: Biological stoichiometry of oleaginous microalgal lipid synthesis: The role of N:P supply ratios and growth rate on microalgal elemental and biochemical composition

<p>Biological Stoichiometry is an ecological framework connecting the balance of elements to the functioning of organisms. Here, we applied this framework to study the relationships between carbon:nitrogen:phosphorus (C:N:P) ratios and synthesis of industrial high value biochemicals in the highly oleaginous alga <em>Tetradesmus bernardii.</em> We expected an increase in protein content with increasing cellular N content and decreasing C:N stoichiometry, and an increase in lipid content with increasing C:N and C:P stoichiometry. We tested these hypotheses by exposing <em>T. bernardii</em> to N and P limitation at a range of N:P supply ratios in chemostats set at low and high dilution rates. Following expectations, the cellular protein content increased with the N content, and decreased with cellular C:N ratios across all treatments. Carbohydrates and lipids largely followed the relative availability of C and increased under both N and P limitation, with higher C:N and C:P ratios. Specifically, lipid content increased by 100–125% upon N and P limitation, with a shift towards more neutral lipids at the cost of glycolipids and phospholipids. Generally, we observed a re-allocation of cellular C from protein to carbohydrates upon modest N limitation, and towards lipids under P and severe N limitation. Our results demonstrate stoichiometrically predictable patterns of industrially valuable compounds in an oleaginous microalga.</p>

Alkyl lipid distribution, elemental composition and compound-specific 14C isotope composition in marine and terrestrial sedimentary archives from sub-Antarctic South Georgia

<p>The dataset comprises elemental data, alkyl lipid data and radiocarbon (14C) data that was obtained on sediment samples from sub-Antarctic South Georgia. The sediments were collected from a range of different sites, including lakes, peat deposits, a marine inlet and a fjord during RV Polarstern expedition PS81 in 2013. Data from: The cutaneous lipid composition of bat wing and tail membranes: a case of convergent evolution with birds

The water vapour permeability barrier of mammals and birds resides in the stratum corneum (SC), the outermost layer of the epidermis. The molar ratio and molecular arrangement of lipid classes in the SC determine the integrity of this barrier. Increased chain length and polarity of ceramides, the most abundant lipid class in mammalian SC, contribute to tighter packing and thus to reduced cutaneous evaporative water loss (CEWL). However, tighter lipid packing also causes low SC hydration, making it brittle, whereas high hydration softens the skin at the cost of increasing CEWL. Cerebrosides are not present in the mammalian SC; their pathological accumulation occurs in Gaucher's disease, which leads to a dramatic increase in CEWL. However, cerebrosides occur normally in the SC of birds. We tested the hypothesis that cerebrosides are also present in the SC of bats, because they are probably necessary to confer pliability to the skin, a quality needed for flight. We examined the SC lipid composition of four sympatric bat species and found that, as in birds, their SC has substantial cerebroside contents, not associated with a pathological state, indicating convergent evolution between bats and birds.
