MicroRNA Panel Predicts Lung Adenocarcinoma in Patients Presenting with Ground-Glass Nodules
<p>MicroRNA (miRNA) expression is correlated with tumor histology, differentiation, invasiveness and treatment outcome. We aimed to identify miRNAs whose differential expression might enable early diagnosis of lung adenocarcinoma in patients presenting with ground-glass nodules (GGNs). To identify potential miRNAs of interest, we analyzed the miRNA expression profile of tumor and adjacent non para-tumor tissue in 3 participants by next-generation sequencing (NGS). We then assessed the expression levels of the miRNAs of interest in 73 lung adenocarcinoma presenting with GGNs with matched adjacent non-tumor tissue by quantitative real-time polymerase chain reaction (qRT-PCR). Target genes of our selected miRNA panel were predicted using Miranda with default parameters. Twenty-three miRNAs showed differential expression between tumor and adjacent non-tumor tissue by NGS. Five miRNAs exhibited higher expression in tumor tissue compared to adjacent non-tumor tissue (P<0.05), eighteen miRNAs demonstrated lower expression in tumor tissue versus adjacent non-tumor tissue (P<0.05). When qRT-PCR was performed for the 23 miRNAs identified by NGS in the pilot stage, seven were found to have statistically significant expression in tumor versus adjacent non-tumor tissue (P<0.05). The predicted targets of our miRNAs of interest are frequently associated with cancer signaling pathways. We developed a miRNA panel that could potential predict the presence of lung adenocarcinoma in patients presenting with GGNs. </p>
Single-cell RNA-seq profiles of lung adenocarcinoma patients and tumor-bearing mice
<p>single-cell RNA sequencing (scRNA-seq) profiles from eight patients with lung adenocarcinoma (LUAD) and four samples of tumor tissues from tumor bearing mice were performed. By integrating other scRNA-seq data and clinical information, we identified activated adaptive immune responses in older patients, reflected by enriched dysfunctional T cell signature scores and immune checkpoint molecules. Our study shows increased efficacy of immune checkpoint blockade therapy in older patients, addressing the prominent role of age when considering immunotherapy.</p>

Single-cell and spatial transcriptomics delineate molecular traits and immunosuppressive landscape during histological progression of lung adenocarcinoma
<p>Two specimens of lung adenocarcinoma, each corresponding to the lepidic and solid histologic patterns as confirmed through histologic scrutiny, were procured in accordance with standard surgical protocols. These specimens underwent a process of formalin fixation and were subsequently encapsulated within paraffin-embedded tissue blocks. The specimens were then sectioned and subjected to hematoxylin and eosin (H&E) staining to facilitate subsequent imaging at a resolution of 40x (equivalent to 0.25 micron/pixel) via the use of Aperio GT450 scanners. The tissue slides were then conveyed to the Genomics core, where following the decoverslipping of the tissue, the Visium CytAssist device was employed to transfer transcriptomic probes from the original glass slides to capture areas on Visium slides measuring 11mm x 11mm. Comprehensive transcriptomic profiling was achieved post mRNA permeabilization, through poly(A) capture and probe hybridization. The resultant libraries were sequenced utilizing the Illumina Novaseq 6000, using paired-end sequencing with a read length of 150 base pairs.</p>

Integrated multi-omics analysis of early lung adenocarcinoma links tumor biological features with predicted indolence or aggressiveness
<p>This is a collection of datasets presented in the manuscript "Multi-omics data analysis identifies correlations between tumor biology features and predicted behaviors in early lung adenocarcinoma". This study provides a comprehensive profiling of LUAD indolence and aggressiveness at the biological bulk and single cell levels, as well as at the clinical and radiomics levels. This hypothesis generating study uncovers several potential future research avenues. It also highlights the importance and power of data integration to improve our systemic understanding of LUAD and to help reduce the gap between basic science research and clinical practice.</p>
