Multiple genetic trajectories to extreme abiotic stress adaptation in Arctic Brassicaceae

<p>Extreme environments offer powerful opportunities to study how different organisms have adapted to similar selection pressures at the molecular level. The Arctic is one of the most hostile environments on Earth, and the few plant species inhabiting this region typically possess suites of similar morphological and physiological adaptations to extremes in light and temperature. Here we compare patterns of molecular evolution in three Brassicaceae species that have independently colonized the Arctic, and present some of the first genetic evidence for plant adaptations to the Arctic environment. By testing for positive selection and identifying convergent substitutions in orthologous gene alignments for a total of 15 Brassicaceae species, we find that positive selection has been acting on different genes, but similar functional pathways in the three Arctic lineages. The positively selected gene sets identified in the three Arctic species showed convergent functional profiles associated with extreme abiotic stress characteristic of the Arctic. However, there was little evidence for independently fixed mutations at the same sites and for positive selection acting on the same genes. The three species appear to have evolved similar suites of adaptations by modifying different components in similar stress response pathways, implying that there could be many genetic trajectories for adaptation to the Arctic environment. By identifying candidate genes and functional pathways potentially involved in Arctic adaptation, our results provide a framework for future studies aimed at testing for the existence of a functional syndrome of Arctic adaptation in the Brassicaceae and perhaps flowering plants in general. </p>

Ecological consequences of plant genotypic diversity within a foundation plant, Spartina alterniflora, are pervasive but not universal across multiple stress gradients

<ol> <li>Plant genotypic diversity can influence population- and community-level processes, yet we have a limited understanding of how these effects vary across environmental gradients that are ubiquitous in nature. </li> <li>We conducted a 2-year field experiment manipulating plant (<em>Spartina</em> <em>alterniflora</em>) genotypic diversity across a natural stress gradient in tidal elevation, both with and without the addition of nutrients. </li> <li> <em>Spartina</em> diversity increased stem production, but the magnitude of this effect was reduced at both the most stressful and most benign endpoints of the combined elevation and nutrient gradient, consistent with recent species diversity studies. Complementarity among individuals likely underpins the observed benefit of <em>Spartina</em> diversity. </li> <li> <em>Spartina</em> diversity also affected the associated marsh community, with higher consumer (<em>Littoraria</em> <em>irrorata</em>) abundance in more diverse plots, owing to both greater <em>Spartina</em> density and increased variation in <em>Spartina</em> traits.</li> <li> <em>Synthesis</em>: The positive effects of <em>Spartina</em> diversity on population- and community-level responses under most environmental conditions highlights the ecological importance of plant genotypic diversity for the maintenance of function across the marsh landscape. </li> </ol>

Data from: Loss of heterosis and family-dependent inbreeding depression in plant performance and resistance against multiple herbivores under drought stress

1. Inbreeding depression (ID), outbreeding depression (OD) and heterosis can occur concurrently in plant populations. ID often increases under environmental stress, but the combined effects of inbreeding, outbreeding between populations and environmental stress, such as drought, on plant performance and herbivore resistance remain unclear. 2. In order to determine environment-dependent and family-dependent ID, OD and heterosis we conducted a common garden experiment with plants from five populations of Brassica nigra. Inbred, within-population outbred and between-population outbred plant families were exposed to drought or ambient water levels. We recorded the abundance and damage caused by specialist herbivores from contrasting feeding guilds, i.e. the phloem-feeding Brevicoryne brassicae, the leaf-chewing Psylliodes chrysocephalus and the stem-boring Ceutorhynchus quadridens larvae. 3. Drought stress had negative effects on growth, herbivore resistance and resistance against B. brassicae and positive effects on investments in reproductive output and plant secondary metabolites (sinigrin). We found drought stress-induced loss of heterosis for plant height and investment in reproductive output. Between-population outbred plants were more sensitive to drought stress in terms of above-ground biomass compared to within-population outbred plants. 4. Drought and inbreeding synergistically negatively influenced traits related to growth and reproductive output (environment-dependent inbreeding depression, EDID). There was high variation among families within populations in the degree of ID and EDID. Genetic variation in EDID could buffer the negative effects of genetic stress associated with habitat fragmentation and concurrent environmental stress. In order to capture fully the effects of both inbreeding and between-population outbreeding under stress the different spatial scales of the effects of inbreeding and between-population outbreeding should be taken into account. 5. Synthesis. Our results indicate that drought stress influences not only inbreeding depression (ID), but also heterosis. These findings shed new light on the combined effects of anthropogenic environmental change and the genetic consequences of habitat fragmentation on plants and their interactions with other organisms. Conservation programmes aiming to restore genetically degraded populations with the translocation of individuals between populations should consider environmental stress as a risk factor.

Additive and mostly adaptive plastic responses of gene expression to multiple stress in Tribolium castaneum

Gene expression is known to be highly responsive to the environment and important for adjustment of metabolism but there is also growing evidence that differences in gene regulation contribute to species divergence and differences among locally adapted populations. However, most studies so far investigated populations when divergence had already occurred. Selection acting on expression levels at the onset of adaptation to an environmental change has not been characterized. Understanding the mechanisms is further complicated by the fact that environmental change is often multivariate, meaning that organisms are exposed to multiple stressors simultaneously with potentially interactive effects. Here we use a novel approach by combining fitness and whole-transcriptome data in a large-scale experiment to investigate responses to drought, heat and their combination in Tribolium castaneum . We found that fitness was reduced by both stressors and their combined effect was almost additive. Expression data showed that stressor responses were acting independently and did not interfere physiologically.  Since we measured expression and fitness within the same individuals, we were able to estimate selection on gene expression levels. We found that variation in fitness can be attributed to gene expression variation and that selection pressures were environment dependent and opposite between control and stress conditions. We could further show that plastic responses of expression were largely adaptive, i.e.  in the direction that should increase fitness.

<p><strong>Abstract&nbsp;</strong></p> <p>Studying multiple sclerosis (MS) and its treatments requires the use of biomarkers for&nbsp;underlying pathological mechanisms. We aim to estimate the required sample size for detecting&nbsp;variations of biomarkers of inflammation and oxidative stress. This is a post-hoc analysis on&nbsp;60 relapsing-remitting MS patients treated with Interferon-1a and Coenzyme Q10 for 3 months&nbsp;in an open-label crossover design over 6 months. At baseline and at the 3 and 6-month visits, we&nbsp;measured markers of scavenging activity, oxidative damage, and inflammation in the peripheral&nbsp;blood (180 measurements). Variations of laboratory measures (treatment eect) were estimated using&nbsp;mixed-eect linear regression models (including age, gender, disease duration, baseline expanded&nbsp;disability status scale (EDSS), and the duration of Interferon-1a treatment as covariates; creatinine&nbsp;was also included for uric acid analyses), and were used for sample size calculations. Hypothesizing&nbsp;a clinical trial aiming to detect a 70% eect in 3 months (power&nbsp;<strong>=&nbsp;</strong>80% alpha-error&nbsp;<strong>=&nbsp;</strong>5%), the sample&nbsp;size per treatment arm would be 1 for interleukin (IL)-3 and IL-5, 4 for IL-7 and IL-2R, 6 for IL-13,&nbsp;14 for IL-6, 22 for IL-8, 23 for IL-4, 25 for activation-normal T cell expressed and secreted (RANTES),&nbsp;26 for tumor necrosis factor (TNF)-, 27 for IL-1, and 29 for uric acid. <p><strong>Abstract</strong></p> <p>Background: Oxidative stress is a driver of multiple sclerosis (MS) pathology. We evaluated&nbsp;the effect of coenzyme Q10 (CoQ10) on laboratory markers of oxidative stress and&nbsp;inflammation, and on MS clinical severity.</p> <p>Methods: We included 60 relapsing&ndash;remitting patients with MS treated with interferon beta1a&nbsp;44<strong>&mu;</strong>g (IFN-<strong>&beta;</strong>1a) with CoQ10 for 3 months, and with IFN-<strong>&beta;</strong>1a 44<strong>&mu;</strong>g alone for 3 more months (in&nbsp;an open-label crossover design). At baseline and at the 3 and 6-month visits, we measured&nbsp;markers of scavenging activity, oxidative damage and inflammation in the peripheral blood,&nbsp;and collected data on disease severity.</p> <p>Results: After 3 months, CoQ10 supplementation was associated with improved scavenging activity&nbsp;(as mediated by uric acid), reduced intracellular reactive oxygen species production, reduced&nbsp;oxidative DNA damage, and a shift towards a more anti-inflammatory milieu in the peripheral&nbsp;blood [with higher interleukin (IL)-4 and IL-13, and lower eotaxin, granulocyte-macrophage colonystimulating&nbsp;factor (GM-CSF), hepatocyte growth factor (HGF), interferon (IFN)-<strong>&gamma;</strong>, IL-1<strong>&alpha;</strong>, IL-2R,&nbsp;IL-9, IL-17F, macrophage inflammatory proteins (MIP)-1<strong>&alpha;</strong>, regulated on activation-normal T cell&nbsp;expressed and secreted (RANTES), tumor necrosis factor (TNF)-<strong>&alpha;</strong>, and vascular endothelial growth&nbsp;factor (VEGF). Also, CoQ10 supplementation was associated with lower Expanded Disability Status&nbsp;Scale, fatigue severity scale, Beck&rsquo;s depression inventory, and the visual analogue scale for pain.</p> <p>Conclusions: CoQ10 supplementation improved scavenging activity, reduced oxidative&nbsp;damage, and induced a shift towards a more anti-inflammatory milieu, in the peripheral blood&nbsp;of relapsing&ndash;remitting MS patients treated with 44<strong>&mu;</strong>g IFN-<strong>&beta;</strong>1a 44<strong>&mu;</strong>g.
