W. Dekkers, Jeroen A. A. Demmers, Kay Grünewald, Charlotte Uetrecht and Marina Mikhaylova. The EMBO Journal (2021) e106523. doi:10.15252/embj.2020106523

Description:

Native mass spectrometry (MS) analysis of the stoichiometry and ion occupancy of recombinant human calmodulin (CaM) and recombinant rat caldendrin (CaD) complex with synthetic mouse myosinV IQ1 (myoIQ) motif in the presence / absence of excess Ca2+ and Mg2+ ions.

Sample processing:

Full-length CaD and CaM as well as the synthetic myoVa peptide were buffer exchanged into 150 mM aqueous ammonium acetate solution (pH 7.4). CaM was twice passed through a Bio-Spin P-6 gel filtration spin column (6 kDa cut-off, Bio-Rad), CaD and the myoVa peptide were buffer exchanged through five cycles of tenfold dilution and re-concentration using centrifugal concentrators Vivaspin 500 (10 kDa cut-off, Sartorius) or Amicon Ultra 0.5mL (3 kDa cut-off, Merck/Millipore), respectively. Desalted proteins were introduced into an Orbitrap Q Exactive UHMR mass spectrometer (Thermo Scientific) via static nanoelectrospray ionization from in-house prepared gold-coated borosilicate glass capillaries Kwik-Fil 1B120F-4 (World Precision Instruments). Proteins were sprayed and analysed at 8.5 µM concentration in ammonium acetate alone or supplemented with 200 µM calcium acetate and 100 µM magnesium acetate (both for trace metal analysis, Sigma-Aldrich). For interaction analysis, CaM and/or caldendrin were mixed with myoVa peptide which had final concentration of 8.5 µM (low concentration) or 34 µM (high concentration). The mass spectrometer was tuned for best signal quality and intensity, keeping ion activation and unfolding minimal. Namely, electrospray voltage was kept at 1.3 kV, source desolvation temperature 250°C, in-source desolvation -50 V, ion transfer profile "high m/z", analyzer profile "low m/z", analyzer target resolution 12500 acquiring in mass range 500 – 9000 m/z. Nitrogen was used as collision gas in HCD cell at relative gas pressure setting 7.0 with gentle collisional activation by 10 V HCD voltage gradient.

Data processing:

Raw spectra were averaged over at least 50 scans for mass deconvolution and peak assignment in UniDec 4.4.1 package (Marty et al., 2015). The averaged spectra were exported for ZENODO deposition using Thermo Scientific FreeStyle 1.5.93.34 as single-scan Thermo .raw files (including instrumental parameters metadata) as well as in plain m/z vs intensity .txt files.

A role for myosin II cluster and membrane energy in cortex rupture for Dictyostelium discoideum cells

Blebs, pressure driven protrusions of the cell membrane, facilitate the movement of eukaryotic cells such as the soil amoeba Dictyostelium discoideum, white blood cells and cancer cells. Blebs initiate when the cell membrane separates from the underlying cortex. A local rupture of the cortex, has been suggested as a mechanism by which blebs are initiated. However, much clarity is still needed about how cells inherently regulate rupture of the cortex in locations where blebs are expected to form. In this work, we examine the role of membrane energy and the motor protein myosin II (myosin) in facilitating the cell driven rupture of the cortex. We perform under-agarose chemotaxis experiments, using Dictyostelium discoideum cells, to visualize the dynamics of myosin and calculate changes in membrane energy in the blebbing region. To facilitate a rapid detection of blebs and analysis of the energy and myosin distribution at the cell front, we introduce an autonomous bleb detection algorithm that takes in discrete cell boundaries and returns the coordinate location of blebs with its shape characteristics. We are able to identify by microscopy naturally occurring gaps in the cortex prior to membrane detachment at sites of bleb nucleation. These gaps form at positions calculated to have high membrane energy, and are associated with areas of myosin enrichment. Myosin is also shown to accumulate in the cortex prior to bleb initiation and just before the complete disassembly of the cortex. Together our findings provide direct spatial and temporal evidence to support cortex rupture as an intrinsic bleb initiation mechanism and suggests that myosin clusters are associated with regions of high membrane energy where its contractile activity leads to a rupture of the cortex at points of maximal energy.

Gliding motility of the diatom Craspedostauros australis correlates with the intracellular movement of raphid-specific myosins

Abstract:

Raphid diatoms are one of the few eukaryotes capable of gliding motility, which is remarkably fast and allows for quasi-instantaneous directional reversals. Besides other mechanistic models, it has been suggested that an actomyosin system provides the force for diatom gliding. However, in vivo data on the dynamics of actin and myosin in diatoms are lacking. In this study we demonstrate that the raphe-associated actin bundles required for diatom movement do not exhibit a directional turnover of subunits and thus their dynamics do not contribute directly to force generation. By phylogenomic analysis we identified four raphid diatom-specific myosins in Craspedostauros australis (CaMyoA-D) and investigated their in vivo localization and dynamics through GFP-tagging. Only CaMyoB-D but not CaMyoA exhibited coordinated movement during gliding, consistent with a role in force generation. The characterization of raphid diatom-specific myosins lays the foundation for unraveling the molecular mechanisms that underlie the gliding motility of diatoms.

This dataset contains all microscopy data used for confocal and TIRFM imaging, with all additional elements required to reproduce figures in the manuscript "Gliding motility of the diatom Craspedostauros australis correlates with the intracellular movement of raphid-specific myosins". The dataset also contains code used to generate plots and analyze microscopy data, as well as protein sequences and code used to generate the phylogenomic tree in the paper.

Data from: The selective myosin II inhibitor blebbistatin reversibly eliminates gastrovascular flow and stolon tip pulsations in the colonial hydroid Podocoryna carnea
Blebbistatin reversibly disrupted both stolon tip pulsations and gastrovascular flow in the colonial hydroid Podocoryna carnea. Epithelial longitudinal muscles of polyps were unaffected by blebbistatin, as polyps contracted when challenged with a pulse of KCl. Latrunculin B, which sequesters G actin preventing F actin assembly, caused stolons to retract, exposing focal adhesions where the tip epithelial cells adhere to the substratum. These results are consistent with earlier suggestions that non-muscle myosin II provides the motive force for stolon tip pulsations and further suggest that tip oscillations are functionally coupled to hydrorhizal axial muscle contraction.
