Maximum performance expression is affected by octopamine and antennae removal in Acheta domesticus

<p>Animals in nature seldom use their maximum performance abilities, likely in part due to context-dependent differences in performance motivation. Despite interest in the factors affecting performance expression, the physiological mechanisms underlying variation in performance motivation are poorly understood. We manipulated levels of the biogenic amine octopamine (OA) to test the hypothesis that OA drives motivation to express maximum bite force in male house crickets. We also tested the effect of antenna removal on bite force given prior evidence of potential links among antennaectomy, aggression, and OA. We found that administration of an OA antagonist, epinastine, significantly decreases realized maximum bite force, as does antenna removal. In addition, the performance decrement induced by antennaectomy is abolished by administration of excess OA, and that rescue effect is itself nullified by the simultaneous administration of epinastine. These data show that OA is an important mediator of performance, and thus of performance motivation, in insects, and potentially a promising candidate for the short term manipulation of performance as well.</p>

Bite force, body size, and octopamine mediate mating interactions in the house cricket (Acheta domesticus)

<p>Mating interactions are rife with conflict because the evolutionary interests of males and females seldom coincide. Intersexual conflict affects sexual selection, yet the proximate factors underlying male coercive ability and female resistance are poorly understood. Male combat outcomes are often influenced by bite force, with superior biters being more likely to achieve victory over poorer biters in a range of species, including crickets. If good performers also achieve mating success through sexual coercion, then bite force might play a role in intersexual conflict as well. We tested the capacity of bite force to influence mating interactions in house crickets both directly by measuring bite forces of males and females and by altering male bite capacity through neuropharmacological manipulation. In addition, the invertebrate neurotransmitter octopamine both mediates aggression and underlies motivation to bite in male house crickets. By blocking octopamine receptors through the application of an antagonist, epinastine, we tested the effects of reduced bite force on male mating success. Our results show that male bite capacity, in combination with body size, influences both the likelihood and the outcomes of mating interactions, whereas treatment of males with epinastine eliminates motivation to mate. Our results suggest a functional role for bite force in affecting both sexual conflict and sexual selection and expand our knowledge of the influence of biogenic amines on reproductive behavior.   </p>

Neuronal octopamine signaling regulates mating-induced germline stem cell increase in female Drosophila melanogaster

<p>Stem cells fuel the development and maintenance of tissues. Many studies have addressed how local signals from neighboring niche cells regulate stem cell identity and their proliferative potential. However, the regulation of stem cells by tissue-extrinsic signals in response to environmental cues remains poorly understood. Here we report that efferent octopaminergic neurons projecting to the ovary are essential for germline stem cell (GSC) increase in response to mating in female <i>Drosophila</i>. The neuronal activity of the octopaminergic neurons is required for mating-induced GSC increase as they relay the mating signal from Sex peptide receptor-positive cholinergic neurons. Octopamine and its receptor Oamb are also required for mating-induced GSC increase via intracellular Ca<sup>2+</sup> signaling. Moreover, we identified Matrix metalloproteinase-2 as a downstream component of the octopamine-Ca<sup>2+</sup> signaling to induce GSC increase. Our study provides a mechanism describing how neuronal system couples stem cell behavior to environmental cues through stem cell niche signaling.</p>

Data from: Octopamine increases individual and collective foraging in a neotropical stingless bee

<p>The biogenic amine octopamine is a key modulator of individual and social behaviours in honeybees, but its role in the other group of highly eusocial bees, the stingless bees, remains largely unknown. In honeybees, octopamine mediates reward perception and affects a wide range of reward-seeking behaviours. Thus, we tested the hypothesis that octopamine increases individual foraging effort and collective food source exploitation in the neotropical stingless bee Plebeia droryana. Octopamine treatment caused a significant increase in the number of bees at artificial sucrose feeders and a 1.73-times higher individual foraging frequency. This effect can be explained by octopamine lowering the sucrose response threshold and, thus, increasing the perceived value of the food source. Our results demonstrate that, similar to its effects on honeybees, octopamine increases both individual and collective food source exploitation in P. droryana. This suggests that, despite having evolved many complex behaviours independently, OA might have similar regulatory effects on foraging behaviours in the two groups of highly eusocial bees.</p>
