Response of humic acids and soil organic matter to vegetation replacement in subtropical high mountain forests
In this study, we used solid-state 13 C NMR spectroscopy, photometric analyses, and chemical fractionation to examine carbon (C) components and lability of SOM in a Japanese cedar (Cryptomeria japonica) forest and moso bamboo (Phyllostachys edulis) plantation reforested on a cutover primary broadleaf forest in Taiwan. The data was collected in Feb 2014.
Polymer-assisted modification of metal-organic framework MIL-96 (Al): influence on particle size, crystal morphology and perfluorooctanoic acid (PFOA) removal

A low cost and water-soluble polymer, hydrolyzed polyacrylamide (HPAM), was added in varying quantities into the synthesis reaction to achieve >200% particle size enlargement with controlled crystal morphology. The modified adsorbent, MIL-96-RHPAM2, was systematically characterized by SEM, XRD, FTIR, BET and TGA-MS. Using activated carbon (AC) as a reference adsorbent, the effectiveness of MIL-96-RHPAM2 for perfluorooctanoic acid (PFOA) removal from water was examined. The study confirms stable morphology of hydrated MIL-96-RHPAM2 particles as well as a superior PFOA adsorption capacity (340 mg/g) despite its lower surface area, relative to standard MIL-96. MIL-96-RHPAM2 suffers from slow adsorption kinetics as the modification significantly blocks pore access. The strong adsorption of PFOA by MIL-96-RHPAM2 was associated with the formation of electrostatic bonds between the anionic carboxylate of PFOA and the amine functionality present in the HPAM backbone. Thus, the strongly held PFOA molecules in the pores of MIL-96-RHPAM2 were not easily desorbed even after eluted with a high ionic strength solvent (500 mM NaCl). Nevertheless, this simple HPAM addition strategy can still chart promising pathways to impart judicious control over adsorbent particle size and crystal shapes while the introduction of amine functionality onto the surface chemistry is simultaneously useful for enhanced PFOA removal from contaminated aqueous systems.</p>
Dataset: Survey on the actual situation of antibiotic resistant bacteria detection by nucleic acid amplification test in clinical microbiology laboratories at hospitals in Japan: Online survey of participants in workshops organized by the Nara Association of Medical Technologists
<p>The coronavirus disease 2019 pandemic has led to the widespread use of the nucleic acid amplification test (NAAT), along with an increase in demand for SARS-CoV-2 tests. NAAT has been used to detect antimicrobial resistance (AMR) genes since before the pandemic, but the test has been performed in a limited number of facilities. We investigated the current status and background of Japanese clinical laboratories by surveying the implementation of genotypic AST in NAAT, which has become widespread owing to the pandemic. This means that 59% of the respondents possessed NAAT and were using it for genotypic AST. GeneXpert and FilmArray were introduced in the majority of cases (62.5% and 82.6%, respectively), with the pandemic as the trigger. More than half of the respondents cited “rapid detection” (56.0%) and “ICT requests” (52.4%) as the reasons for introducing the system. Regarding usefulness, “contribution to infectious disease treatment” (74.1%) showed the highest percentage. Among the respondents who cited “not implemented”, the most frequent responses were “I have no plans, but I want to do it.” (38.1%) and “would do so if requested by a physician” (33.3%). The most common reason for not implementing the system was concern about increased workload (52.9%). We believe that this is due to changes in the working environment caused by the pandemic and the characteristics of Japanese society. In the future, to promote the adoption of genotypic AST, it will be necessary to approach it through reports on its usefulness from domestic facilities, and simultaneously, improving and enhancing efficiency in work processes will also be essential.</p>
Quantification of phosphorylated metabolites, organic acids, and intermediates of the TCA cycle using capillary ion chromatography tandem mass spectrometry (capIC-MS/MS) following treatment of Escherichia coli with ciprofloxacin
<p>Capillary ion chromatography tandem mass spectrometry (capIC-MS/MS) was used to quantify phosphorylated metabolites, organic acids, and intermediates of the TCA cycle of Escherichia coli treated with ciprofloxacin, BTP-001 (a novel antimicrobial peptide), and a combination of the two . Metabolite extracts were analyzed with a Xevo TQ-XS triple quadrupole mass spectrometer (Waters, USA).</p><p>Samples were gathered from E. coli cultures grown in batch cultivations using 1 liter bioreactors. Briefly, intracellular metabolites were extracted by cycling samples between −20 °C EtOH and N2 (<i>l</i>) in three consecutive freeze–thaw cycles, with vortexing every 10 min during the thawing phase. Filters were removed and the cell debris was pelleted (4500 rcf, 10 min, -9 °C). The supernatants were transferred to a new tube, snap frozen in N2 (<i>l</i>), and lyophilized. Lyophilized extracts were reconstituted in 500 µL cold Milli-Q H2O and cleared by spin-filtration with a 10 kDa molecular cutoff (20817 rcf, 10 min, 0 °C). A mix of 80 µL centrifuged sample and 20 µL 13C-labeled ISTD extract from yeast was sent to analysis. </p><p>Data processing and absolute quantification was performed as earlier described using the TargetLynx application manager of MassLynx v 4.1 (Waters) to interpolate calibration curves made with appropriate dilutions of analytical grade standards (Sigma-Aldrich). The response factor of the corresponding U13C-isotopologues were used to correct the standard and sample extract response factors. Extract concentrations were normalized to the CDW, which was calculated from interpolation of the OD600 vs. CDW (g/L) curve. </p><p>Further statistical analysis in MetaboAnalyst v 5.0 replaced missing values with 1/5 of the minimum value of the respective metabolite. An unpaired T-test with unequal variance determined differential enriched metabolites with a false discovery rate (FDR) < 0.05 which are presented as log2 fold-change compared to control. </p>
High performance cation exchange membranes synthesized via in-situ emulsion polymerization without organic solvents and corrosive acids
<p>Dataset supporting journal publication:</p> <p><strong>Abstract:</strong> The synthesis of cation exchange membranes (CEMs) usually involves using organic solvents and/or sulfonation process. In this study, green and scalable synthesis of high performance CEMs is achieved without organic solvents and sulfonation. The synthesis is carried out via in-situ polymerization of lithium styrene sulfonate in porous support. Different preparation procedures are developed and optimized. Functional sulfonate groups were successfully loaded onto and into the membrane support, as verified by FTIR. Besides, water plays an important role during membrane synthesis. By reducing the amount of water used, the ratio of functional polymers to membrane support in the synthesized CEMs is increased. Therefore, the synthesized CEMs show increased ion exchange capacity (IEC). This is significant because it means that high IEC can be achieved without introducing cation exchange resins to the membranes. Finally, the synthesized membranes demonstrate high desalination performance. This new methodology may shed new light on preparing CEMs in an efficient and eco-friendly way.</p>
Seasonal dynamics of sinking organic matter in the Pacific Arctic Ocean revealed by nitrogen isotope ratios of amino acids
<p><span>The Pacific Arctic Ocean has experienced a rapidly changing climate, sea-ice retreat, and enhanced primary production over the past few decades. The export production generated by photoautotrophs and heterotrophs has been characterized in the Arctic Ocean, but their seasonal variations in relative proportion are largely unknown due to the limited access in the ice-covered season. We measured the concentration and nitrogen isotope ratio of individual amino acids from sinking particles in the northern east Siberian Sea (KAMS1), northern Chukchi Sea (KAMS2), and Northwind Ridge (KAMS4) from August 2017 to July 2019. </span><span>The average trophic position, based on differences in the nitrogen isotope ratios of glutamic acid and phenylalanine, can indicate the relative proportions of biogenic organic matters derived from photoautotrophs and heterotrophs in sinking particles. Decreasing values (close to 1.0) in summer at KAMS2 in 2018 suggest that primary producers are responsible for most of the downward flux of sinking particles. However, the average trophic position at KAMS1 in 2017 increased to > 1.5 in autumn and was maintained at approximately 1.7 during ice-covered winter periods, likely due to greater contributions from heterotrophic organisms. Exceptionally high average trophic positions (close to 2.0) of sinking particles in summer at KAMS1 in 2017 and KAMS4 in 2018 were likely due to small export of photoautotrophs due to the surface seawater stratification and limited pelagic production. </span><span>T</span><span>he average trophic position in sinking particles should reflect the spatiotemporal variation in export particle composition in the </span><span>Pacific Arctic Ocean</span><span>.</span></p>
