Photomorphogenesis for Robot Self-assembly: Adaptivity, Collective Decision-making, and Self-repair

Self-assembly in biological systems is an inspiration for engineered large-scale multi-modular systems with desirable characteristics, such as robustness, scalability, and adaptivity. Previous works have shown that simple mobile robots can be used to emulate and study self-assembly behaviors. However, many of these studies were restricted to rather static and inflexible aggregations in predefined shapes, and were limited in adaptivity compared to that observed in nature. We propose a photomorphogenesis approach for robots using our vascular morphogenesis model---a light-stimuli directed method for multi-robot self-assembly inspired by the tissue growth of trees. Robots in the role of `leaves' collect a virtual resource that is proportional to a real, sensed environmental feature. This resource is then shared throughout the whole robot aggregate and determines where it grows or shrinks as a reaction to the dynamic environment. In our approach the robots use supplemental bioinspired models to collectively select a seed robot to decide who starts to self-assemble (and where), or to assemble static aggregations. The robots then use our vascular morphogenesis model to aggregate in a directed way preferring bright areas, hence resembling natural phototropism (growth towards light). In this assembly, they are adaptive and able to react to a dynamic environment by collectively and autonomously rearranging the aggregate, discarding outdated parts and growing new ones. In representative experiments, the self-assembling robots collectively make rational decisions on where to grow. Cutting off parts of the aggregate triggers a self-organizing repair process in the robots, and the parts regrow. All these capabilities of adaptivity, collective decision-making, and self-repair in our robot self-assembly originate directly from self-organized behavior of the vascular morphogenesis model. Our approach opens up opportunities for self-assembly with reconfiguration on short time-scales with high adaptivity of dynamic forms and structures.

Data from: Alternative splicing substantially diversifies the transcriptome during early photomorphogenesis and correlates with the energy availability in Arabidopsis
Plants use light as source of energy and information to detect diurnal rhythms and seasonal changes. Sensing changing light conditions is critical to adjust plant metabolism and to initiate developmental transitions. Here we analyzed transcriptome-wide alterations in gene expression and alternative splicing (AS) of etiolated seedlings undergoing photomorphogenesis upon exposure to blue, red, or white light. Our analysis revealed massive transcriptome reprograming as reflected by differential expression of ~20% of all genes and changes in several hundred AS events. For more than 60% of all regulated AS events, light promoted the production of a presumably protein-coding variant at the expense of an mRNA with nonsense-mediated decay-triggering features. Accordingly, AS of the putative splicing factor REDUCED RED-LIGHT RESPONSES IN CRY1CRY2 BACKGROUND 1 (RRC1), previously identified as a red light signaling component, was shifted to the functional variant under light. Downstream analyses of candidate AS events pointed at a role of photoreceptor signaling only in monochromatic but not in white light. Furthermore, we demonstrated similar AS changes upon light exposure and exogenous sugar supply, with a critical involvement of kinase signaling. We propose that AS is an integration point of signaling pathways that sense and transmit information regarding the energy availability in plants.
High resolution imaging as a tool for identifying quantitative trait loci that regulate photomorphogenesis in Arabidopsis thaliana

A primary component of seedling establishment is the photomorphogenic response as seedlings emerge from the soil. This process is characterized by a reduced growth rate in the hypocotyl, increased root growth, opening of the apical hook, and expansion of the cotyledons as photosynthetic organs. While fundamental to plant success, the photomorphogenic response can be highly variable. Additionally, studies of Arabidopsis thaliana are made difficult by subtle differences in growth rate between individuals. High-resolution imaging and computational processing have emerged as useful tools for quantification of such phenotypes. This study sought to: 1) develop an imaging methodology which could capture changes in growth rate as seedlings transition from darkness to blue light in real time, and 2) apply this methodology to single-QTL analysis using the Cvi x Ler RIL mapping population. Significant differences in the photomorphogenic response were observed between the parent lines and analysis of 158 RILs revealed a wide range of growth rate phenotypes. QTL analysis detected significant loci associated with dark growth rate on chromosome 5 and significant loci associated with light growth rate on chromosome 2. Candidate genes associated with these loci, such as the previously characterized ER locus, highlight the application of this approach for QTL analysis. Genetic analysis of Landsberg lines without the erecta mutation also supports a role for ER in modulating the photomorphogenic response, consistent with previous QTL analyses of this population. Strengths and limitations of this methodology are presented, as well as means of improvement.
