Abundance of eukaryote picophytoplankton and Synechococcus from a moored submersible flow cytometer at Martha's Vineyard Coastal Observatory, ongoing since 2003 (NES-LTER since 2017)
This is a decadal-scale time series of the abundance of eukaryote picophytoplankton and Synechococcus at 4 meters depth at the Martha's Vineyard Coastal Observatory, about 3 km south of Katama Beach, Edgartown, Massachusetts, USA. Picophytoplankton were sensed in situ by a submersible flow cytometer (FlowCytobot, or FCB). Sampling frequency was continuous at approximately 20-minute intervals binned to hourly resolution with some exceptions (e.g., winter in some years). This time series is ongoing for Northeast U.S. Shelf Long-Term Ecological Research (NES-LTER).
Data set for the paper: Intercomparison of ocean colour algorithms for picophytoplankton carbon in the ocean

This dataset contains the phytoplankton carbon,Cphy, obtained from in situ counts of phytoplankton cells using ow cytometry presented in the paper [13]. The location and time of the samples have been matched with the satelllite data in the Ocean Colour Climate Change Initiative (OCCCI) dataset. This dataset is the match between the in situ Cphy and the products from using the OCCCI inputs (i.e. chlorophyll concentration, backscattering coecient, phytoplankton absorption) with 6 different algorithms. This document describes the dataset details: data sources, computation of Cphy, selected data.

Picophytoplankton and bacteria abundances analyzed with flow cytometry (FCM) from CCE-CalCOFI Augmented cruises in the California Current System, 2004 - 2023 (ongoing).
Picophytoplankton populations and non-pigmented prokaryotes are sampled within the California Current Ecosystem (CCE) for abundances from 3 to 8 depths at CalCOFI stations. Seawater is collected from Niskin bottles and cells are fixed in the field aboard the survey cruises (since 2004, ongoing) with paraformaldehyde, and stained with a DNA-specific dye back in the laboratory. The cells are enumerated by an Altra flow cytometer (with a syringe pump for volumetric sample delivery) simultaniously with argon ion lasers, to distinguish three major populations of photoautotrophs (Prochlorococcus, Synechococcus, and pico-eukaryotes) and the assemblage of heterotrophic prokaryotes collectively referred to as H-Bact.

FCM abundance estimates for each are converted to carbon biomass equivalents using mixed-layer estimates.

Time series measurements of group-specific picophytoplankton primary production

14C-based primary production by flow cytometrically determined pico plankton (Prochlorococcus, Synechococcus, and photosynthetic picoeukaryotes) were measured at six discrete depths (5, 25, 45, 75, 100, and 125 m) at Station ALOHA. Seawater samples from each depth were subsampled into triplicate 30-mL polycarbonate centrifuge tubes from a pre-dawn cast, inoculated under subdued light with 70 µL of NaH14CO3-, then incubated over the full photoperiod (~12-14 hours). At the end of the incubation period (after sundown), 5 mL of each radiolabeled sample was preserved in cryotubes containing 30 µL of 16% (final concentration 0.24% w/v) microscopy-grade paraformaldehyde, flash-frozen in liquid nitrogen, and stored at -80°C for subsequent flow cytometric sorting. Group-specific rates of 14C-assimilation by Prochlorococcus, Synechococcus, and PPE were determined by measuring the amount of 14C assimilated into populations sorted using a BD Influx sorting flow cytometer.
