Pervasive relaxed selection on spermatogenesis genes in gorillas coincident with the evolution of polygyny

<p>Gorillas have a polygynous social system in which the highest-ranking male has almost exclusive access to females and sires most of the offspring in the troop. Such behavior results in a  dramatic reduction of sperm competition, which is ultimately associated with  numerous traits leading to a low efficacy of gorilla spermatogenesis. However, the molecular basis behind the remarkable erosion of the gorilla male reproductive system remains unknown. Here, we explored the genetic consequences of the polygynous social system in gorillas by testing for altered selection intensity across 13,310 orthologous protein-coding genes from 261 Eutherian mammals. We identified 578 genes with relaxed purifying selection in the gorilla lineage, compared with only 96 that were positively selected. Genes under relaxed purifying selection in gorillas have accumulated numerous deleterious amino acid substitutions; their expression is biased towards male germ cells, and are enriched in functions related to meiosis and sperm biology. We tested the function of gorilla relaxed genes previously not implicated in sperm biology using the Drosophila model system and identified 41 novel spermatogenesis genes required for normal fertility. Furthermore, by exploring exome/genome sequencing data of infertile men with severe spermatogenic impairment, we found that the human orthologs of the gorilla relaxed genes are enriched for loss-of-function variants in infertile men. These data provide compelling evidence that reduced sperm competition in gorillas is associated with  relaxed purifying selection on genes related to male reproductive function. The accumulation of deleterious mutations in these genes likely provides the mechanistic basis behind the low efficacy of gorilla spermatogenesis and uncovers new candidate genes for human male infertility.</p>

Data from: Where are the beachmasters? Unexpectedly weak polygyny among southern elephant seals on a South Shetland Island

<p><strong>Abstract</strong></p> <p>Intraspecific variation in animal mating systems can have important implications for ecological, evolutionary and demographic processes in wild populations. For example, patterns of mating can impact social structure, dispersal, effective population size and inbreeding. However, few species have been studied in sufficient detail to elucidate mating system plasticity and its dependence on ecological and demographic factors. Southern elephant seals (Mirounga leonina) have long been regarded as a textbook example of a polygynous mating system, with dominant &lsquo;beachmaster&rsquo; males controlling harems of up to several hundred females. However, behavioural and genetic studies have uncovered appreciable geographic variation in the strength of polygyny among elephant seal populations. We therefore used molecular parentage analysis to investigate patterns of parentage in a small satellite colony of elephant seals at the South Shetland Islands. We hypothesised that dominant males would be able to successfully monopolise the relatively small numbers of females present in the colony, leading to relatively high levels of polygyny. A total of 424 individuals (comprising 33 adult males, 101 adult females and 290 pups) sampled over eight years were genotyped at 20 microsatellites and reproductive success was analysed by genetically assigning parents. Paternity could only be assigned to 31 pups (10.7%), despite our panel of genetic markers being highly informative and the genotyping error rate being very low. The strength of inferred polygyny was weak in comparison to previous genetic studies of the same species, with the most successful male fathering only seven pups over the entire course of the study. Our results show that, even in a species long regarded as a model for extreme polygyny, male reproductive skew can vary substantially among populations.</p>

1. The Acacia drepanolobium (also known as Vachellia drepanolobium) ant-plant symbiosis is considered a classic case of species coexistence, in which four species of tree-defending ants compete for nesting space in a single host tree species. Coexistence in this system has been explained by trade-offs in the ability of the ant associates to compete with each other for occupied trees versus the ability to colonize unoccupied trees. 2. We seek to understand the proximal reasons for how and why the ant species vary in competitive or colonizing abilities, which are largely unknown. 3. In this study, we use RADseq derived SNPs to identify relatedness of workers in colonies to test the hypothesis that competitively dominant ants reach large colony sizes due to polygyny, i.e., the presence of multiple egg-laying queens in a single colony. 4. We find that variation in polygyny is not associated with competitive ability; in fact, the most dominant species, unexpectedly, showed little evidence of polygyny. We also use these markers to investigate variation in mating behavior among the ant species, and find that different species vary in the number of males fathering the offspring of each queen. Finally, we show that the nature of polygyny varies between the two commonly polygynous species, Crematogaster mimosae and Tetraponera penzigi: in C. mimosae, queens in the same colony are often related, while this is not the case for T. penzigi. 5. These results shed light on factors influencing the evolution of species coexistence in an ant-plant mutualism, as well as demonstrating the effectiveness of RADseq-derived SNPs for parentage analysis.

Data from: Polyandry and polygyny in a social rodent: an integrative perspective based on social organization, copulations, and genetics

Animal mating systems have fascinated biologists for thousands of years. Ways to describe amating systeminclude determining social organization, observing copulations, or using genetics to assign parentage. Social organization can be difficult to quantify, however, documentation of copulations is often challenging, many copulations do not produce offspring, and genetic variation is sometimes minimal. Here we use data from a 7-year study of wild white-tailed prairie dogs (WTPDs, Cynomys leucurus) living in Colorado USA to estimate the frequencies of polyandry (i.e., copulation with ≥2 males) and polygyny (i.e., copulation with ≥2 females) from three independent approaches: (1) determination of the number of males and females living in the same territory (social organization); (2) observations of copulations; and (3) genetic assignments of paternity from seven polymorphic microsatellites. We predicted that our three approaches would yield similar estimates of polyandry and polygyny. Because a WTPD female's period of sexual receptivity each spring is limited to several hours on a single day, we also predicted that frequencies of polyandry and polygyny would be lower for WTPDs than for animals with longer periods of receptivity. Our results did not support these predictions. For polyandry, observations of copulations and genetics indicated similar overall percentages (27%), but social organization indicated a much lower percentage (2%). For polygyny, observations of copulations indicated the highest overall percentage (84%), then social organization (59%), then genetics (46%). All three approaches showed striking annual variation in the frequencies of WTPD polyandry and polygyny. Long-term studies that integrate behavioral and genetic insights can provide a detailed view of a mating system, but feasibility will depend on ease of capture, visibility of copulations, length of mating season, research objectives, and genetic variation.

Data from: Climatic conditions cause spatially dynamic polygyny thresholds in a large mammal

The polygyny threshold (PT) is a critical transition point in the sexual selection process for many organisms in natural populations, characterizing when females choose to mate with an already mated male over an unmated one to improve fitness. Understanding its causes and consequences is therefore of high interest. While both theoretical and empirical work suggest that the degree of polygyny within a species is plastic and a function of male inequality, the functional relationship between underlying availability of resources occupied by breeding males under variable climatic conditions and the dynamics of PTs across space and time has received less attention. Here, we use a standardized measure of male mating inequality as the culmination of female mate choices to analyse how spatially dynamic PTs in a naturally regulated feral horse (Equus ferus caballus) population emerge along a geographic gradient in a known, limiting resource (freshwater) each year from variable climatic conditions. Polygyny threshold distance from permanent freshwater increased with increasing precipitation during the breeding season of each year, suggesting a relationship between annual resource availability and female mate choice. The mechanism by which climatic conditions underpin the spatial dynamics of PTs was likely through precipitation providing ephemeral freshwater sources across the study area that effectively weakened the gradient in availability of permanent freshwater, thereby providing mating males that occupied home ranges far from permanent water with access to this limiting resource and enabling them to attract and retain females. Increased precipitation also coincided with a decreased proportion of males in the population that experienced sexual selection pressure attributed to female mate choice in relation to the acquisition and/or defence of freshwater sources. Climatic conditions caused spatial shifts in PTs annually along the geographic gradient in resource availability. Our findings reveal that such environmental gradients may either buffer or amplify impacts of climatic variation on selection pressure operating in natural populations, and emphasize the importance of integrating spatially explicit PTs with atmospheric fluctuations when predicting the effect of climatic change on selection processes within populations that occupy environmental gradients.

1. Mate searching is assumed to be performed mostly by males, but when females benefit from multiple mating or are under risk of failing to mate, they may also perform mate searching. This is especially important in scramble competition polygynies, in which mate searching is the main mechanism of mate competition. Typically, more mobile individuals are expected to achieve higher mating success because mobility increases their probability of finding mates. 2. If we assume individual movements are mainly explained by mate searching in scramble competition polygynies, we can investigate searching strategies by asking when individuals should leave their location and where they should go. We hypothesize that individuals will leave their locations when mating opportunities are scarce and will seek spatially close sites with better mating opportunities. 3. We tested these hypotheses for males and females of Leptinotarsa undecimlineata, a leaf beetle with scramble competition polygyny in which both sexes are promiscuous. Individuals mate and feed exclusively on Solanum plants, thus individual movements can be described as switches between plants. 4. Females were less likely than males to leave isolated plants, and both males and females moved preferentially to neighbouring plants. Males were more likely to leave when the local number of females was low, and the number of males was high. They moved to plants with more females, a behaviour consistent with a mate searching strategy. Females were more likely to move to plants with fewer males and many females, a behaviour consistent with male harassment avoidance. 5. Strategic movement is widely considered in foraging context, but seldom in a mate searching context. Considering that selection to minimize searching costs, maximize mating success, and minimize harassment may be ubiquitous in nature, we argue that strategic movements by mate searching individuals are likely to occur in many species.

1. Patterns of male reproductive allocation provide insight into life-history characteristics. The trade-offs associated with resource and female group defense are well-defined. However, less is understood about trade-offs in species that practice scramble-competition polygyny, where successful strategies may favor competitive mate-searching rather than contest competition and fighting. 2. White-tailed deer (Odocoileus virginianus) practice scramble-competition polygyny where solitary males search for and assess receptivity of females scattered across the landscape. Physically mature males are expected to do most of the breeding because of the high energetic costs of reproduction and high social status. However, young males may collectively sire one-third of offspring. To gain a better understanding of trade-offs associated with scramble-competition polygyny, we quantified metrics associated with reproductive effort and success. 3. We quantified changes in body mass of harvested males, energetic costs of locomotion based on movements of GPS radio-collared males, and timing of reproduction via temporal genetic parentage assignments. 4. Young males (1.5 and 2.5 years old) sired offspring, but their mating success was mainly limited to peak rut, when most females were in estrus. Furthermore, multiple paternity was common, indicating opportunistic reproduction. Reproductive effort, indexed by body mass loss, was highest in prime-age males (5.5-6.5 years old). Surprisingly, young and post-prime males also exhibited significant body mass loss, indicative of investment in reproductive effort. Movement rates increased 2 to 4-fold during rut as a function of mate-search activities, but cost of locomotion would cause only about one-third of observed body mass loss. Because males are capital breeders, we infer most of body mass loss is due to reduced foraging. 5. In scramble-competition polygyny, the repeated location of potential mates and assessment of their estrous status appear to be important constituents of male mating strategies. Therefore mating success may be influenced by time management and spatial memory, and not based solely on social dominance. Thus, reproductive effort should be greater for individuals capable of reducing time foraging. For those that cannot, opportunistic mating opportunities may arise when operative adult sex ratios are low. Our analyses reveal valuable insight into the trade-offs associated with scramble-competition polygyny.

Data from: Polygyny without wealth: popularity in gift games predicts polygyny in BaYaka Pygmies

The occurrence of polygynous marriage in hunter–gatherer societies, which do not accumulate wealth, remains largely unexplored since resource availability is dependent on male hunting capacity and limited by the lack of storage. Hunter–gatherer societies offer the greatest insight in to human evolution since they represent the majority of our species' evolutionary history. In order to elucidate the evolution of hunter–gatherer polygyny, we study marriage patterns of BaYaka Pygmies. We investigate (i) rates of polygyny among BaYaka hunter–gatherers; (ii) whether polygyny confers a fitness benefit to BaYaka men; (iii) in the absence of wealth inequalities, what are the alternative explanations for polygyny among the BaYaka. To understand the latter, we explore differences in phenotypic quality (height and strength), and social capital (popularity in gift games). We find polygynous men have increased reproductive fitness; and that social capital and popularity but not phenotypic quality might have been important mechanisms by which some male hunter–gatherers sustained polygynous marriages before the onset of agriculture and wealth accumulation.

Sexual selection is one of the earliest areas of interest in evolutionary biology. And yet, the evolutionary history of sexually dimorphic traits remains poorly characterized for most vertebrate lineages. Here, we report on evidence for the early evolution of dimorphism within a model mammal group, the pinnipeds. Pinnipeds show a range of sexual dimorphism and mating systems that span the extremes of modern mammals, from monomorphic taxa with isolated and dispersed mating to extreme size dimorphism with highly ordered polygynous harem systems. In addition, the degree of dimorphism in pinnipeds is closely tied to mating system, with strongly dimorphic taxa always exhibiting a polygynous system, and more monomorphic taxa possessing weakly polygynous systems. We perform a comparative morphological description, and provide evidence of extreme sexual dimorphism (similar to sea lions), in the Miocene-aged basal pinniped taxon Enaliarctos emlongi. Using a geometric morphometric approach and combining both modern and fossil taxa we show a close correlation between mating system and sex-related cranial dimorphism, and also reconstruct the ancestral mating system of extant pinnipeds as highly polygynous. The results suggest that sexual dimorphism and extreme polygyny in pinnipeds arose by 27 Ma, in association with changing climatic conditions.
