AGO2 localises to cytokinetic protrusions in a p38 dependent manner and is needed for accurate cell division.

Argonaute 2 (AGO2) is an indispensable component of the RNA-induced silencing complex, operating at the transcriptional or posttranscriptional level. It is compartmentalized into structures such as GW- and P-bodies, stress granules and adherens junctions as well as the midbody. Here we show using immunofluorescence, image- and bioinformatic analysis and cytogenetics that AGO2 also resides in membrane protrusions such as open- and close-ended tubes. The latter are cytokinetic bridges where AGO2 colocalizes at the midbody-arms with cytoskeletal components such as α-Τubulin and Aurora B and various kinases. AGO2, phosphorylated on serine 387 is located together with Dicer at the midbody ring in a manner dependent on p38 MAPK activity. We further show that AGO2 is stress sensitive and important to ensure the proper chromosome segregation and cytokinetic fidelity. We suggest that AGO2 is part of a regulatory mechanism triggered by cytokinetic stress to generate the appropriate micro-environment for local transcript homeostasis.

Statement: AGO2 resides in open-ended tunneling nanotubes and close-ended cytokinetic bridges. At the latter location AGO2 colocalises with cell division components and the authors show that AGO2 deregulation impairs cell division fidelity.

Computational exploration of treadmilling and protrusion growth observed in fire ant rafts

Condensed active matter systems regularly achieve cooperative emergent functions that individual constituents could not accomplish alone. The rafts of fire ants (Solenopsis invicta) are often studied in this context for their ability to create structures comprised entirely of their own bodies, including tether-like protrusions that facilitate exploration of flooded environments. While similar protrusions are observed in cytoskeletons and cellular aggregates, they are generally dependent on morphogens or external gradients leaving the isolated role of local interactions poorly understood. Here we demonstrate through an ant-inspired, agent-based numerical model how protrusions in ant rafts may emerge spontaneously due to local interactions and how phases of exploratory protrusion growth may be induced by increased ant activity. These results provide an example in which functional morphogenesis of condensed active matter may emerge purely from locally-driven collective motion and may provide a source of inspiration for the development of autonomous active matter and swarm robotics.

Suction feeding turned on its head: a functional novelty facilitates lower jaw protrusion

Functional novelties play important roles in creating new ways for organisms to access resources. In fishes, jaw protrusion has been attributed to the massive diversity of suction-based feeding systems, facilitating the dominant mode of prey capture in this group. Nearly all fishes that feed by suction use upper jaw protrusion, achieved by rotation of the mandible at its base, which then transmits forward motion to independently mobile upper jaw bones. In this study, by contrast, we explore an unusual form of lower jaw protrusion in the freshwater invertivore, Nannocharax fasciatus, enabled by a novel intramandibular joint (IMJ). We combine morphological, kinematic, and biomechanical data to show that the added mobility created by the IMJ influences the pattern of suction-based prey capture movements and contributes to lower jaw protrusion (increasing it by 25%, based on biomechanical modeling). Interestingly, the upper jaw bones are fused in N. fasciatus and rotate about a single fixed joint, like the lower jaws of most other suction feeding fishes. We suggest that this vertical inversion of the jaw protrusion mechanism for ventrally directed suction-feeding on benthic prey is a likely exaptation, as the IMJ is used for biting in related taxa. This work highlights the ability of novelties to facilitate ecological specialization by enabling new functional capabilities.

Experimental data of an article on the impact of coarse-grain protrusion on near-bed hydrodynamics

This deposit contains experimental data related to the article entitled " impact of coarse-grain protrusion on near-bed hydrodynamics" that will be publised in Journal of Geophysical Research - Earth surface. It consists in post-processed data (turbulence statistics) for flows around hemispheres protruding from a bed of fine sediments. Calculated statistics (time-averaged and double-averaged) are provided along other inferred quantities (friction velocity, etc...) and experimental parameters (protrusion level, discharge, etc..) in matlab metadata format. Matlab functions are given to plot all the figures of the article.

X-ray computed tomography reveals that grain protrusion controls entrainment shear stress for entrainment of fluvial gravels: Dataset

This is the dataset that accompanies a paper in Geology:

Hodge RA, Voepel H, Leyland J, Sear DA, Ahmed S (2020). X-ray computed tomography reveals that grain protrusion controls entrainment shear stress for entrainment of fluvial gravels. Geology, 48(2), 149-153.

The aim of this work was to understand how the properties of a sediment grain in a river bed affect the forces required to entrain that grain. This dataset presents the properties of 1055 sediment grains, which were meaured using CT scanning.
