Case report: Large-cell calcifying Sertoli cell tumor

Case history

21-year-old male with a painless mass in the right testicle and a prior history of cutaneous lesions.

Histologic findings

Figures show a proliferation composed of large, polygonal cells with ample, eosinophilic cytoplasm, round nuclei, evident nucleoli, with a trabecular, cord-like pattern of growth, associated with foci of dystrophic calcification and psammomatous bodies, within a myxoid loose stroma.

Discussion

Large-cell calcifying Sertoli cell tumor is an unusual morphological variant of Sertoli cell tumors composed of large polygonal Sertoli cells associated with calcification. Around one third of all cases appear in patients with the Carney or, less frequently, with the Peutz-Jeghers syndrome.

Clinically, it presents as a small (<2 cm), painless testicular mass, which can be bilateral and multifocal in about 40% of the cases, especially in syndromic settings. In non-sporadic cases extratesticular manifestations are related to the associated syndrome, such as myxomas of the skin, soft-tissue, and heart and myxoid lesions of the breast, facial lentigines, cutaneous blue nevi, secondary Cushing syndrome due to pigmented adrenocortical nodular hyperplasia, pituitary somatotroph adenomas, and psammomatous melanotic schwannomas.

Microscopically, solid, tubular, and cord-like growth patterns can be observed. Tumor cells are large polygonal with abundant eosinophilic, finely granular cytoplasm, vesicular nuclei with prominent nucleoli, within a myxohyaline stroma. Distinct calcifications characterized by large, laminated calcified nodule, psammoma bodies and focal ossification, and neutrophilic infiltration, can be also found.

The presence of large tumor size (>4 cm), overt nuclear atypias, increased mitotic activity (>3 mitosis/10 HPF), necrosis, extratesticular extension, and vascular invasion are associated with clinical aggressive behavior. Malignant tumors appear in patients older than 25 years old and are usually not associated with syndromes.

Tumor cells are positive for vimentin, S-100, EMA, melan-A, desmin, and inhibin, and focally positive for cytokeratins. The main differential diagnoses of large-cell calcifying Sertoli cell tumor include Sertoli cell and Leydig cell tumors.

Most of the cases behave indolently, and surgical excision is the treatment of choice.

Effects of titanium dioxide nanoparticles on porcine pre-pubertal sertoli cells

This dataset contains data from an in vitro experiment described in the paper: "Mancuso F, Arato I, Di Michele A, Antognelli C, Angelini L, Bellucci C, Lilli C, Boncompagni S, Fusella A, Bartolini D, Russo C, Moretti M, Nocchetti M, Gambelunghe A, Muzi G, Baroni T, Giovagnoli S, Luca G. Effects of Titanium Dioxide Nanoparticles on Porcine Prepubertal Sertoli Cells: An "In Vitro" Study. Front Endocrinol (Lausanne). 2022 Jan 3;12:751915. doi: 10.3389/fendo.2021.751915. PMID: 35046890; PMCID: PMC87623342".

The data describes the X-ray diffraction (XRD), scanning electron microscopy (SEM) analysis to confirm the correct synthesis of titanium dioxide nanoparticles (TiO2 NPs), and the transmission electron microscopy (TEM) analysis to investigate the morphological/ultrastructural effects on porcine pre-pubertal Sertoli cells (SCs), after an acute (24 hours) and chronic (from 1 up to 3 weeks) exposure, at both sub-toxic (5 µg/ml) and toxic (100 µg/ml) doses of TiO2 NPs.

The diffractogram confirmed the correct synthesis of TiO2 NPs in accordance with the International Centre for Diffraction Data Sample (JCPDS 00-001-0562) and the asterisk (*) represents the peaks of TiO<sub> </sub>NPs in anatase form; SEM analysis showed that the mean size distribution of TiO<sub>2</sub> NPs in dry form was 20 ± 5 nm and confirmed that TiO<sub>2</sub> NPs tend to form aggregates of submicrometric dimensions;

Representative TEM images of unexposed SCs showed elongated oval or spindle-shaped SCs containing quite large nuclei (N) and abundant mitochondria. SCs did not show substantial and/or severe ultrastructural alterations in SCs treated with 5 µg/ml TiO2 NPs for 1 and 2 weeks, in all the other exposure conditions (5 µg/ml for 3 weeks and 100 µg/ml for 1, 2, and 3 weeks), SCs showed several ultrastructural changes with a high percentage of severely damaged SCs, presenting different features of cell stress such as deeply invaginated and shrunk nuclei, disorder/ marginalization of chromatin components, swollen very fragmented, or almost completely missing endoplasmic reticulum membranes. In addition, mitochondrial morphology was severely affected and mitochondria number dramatically decreased with the presence of several large vacuoles, probably as a result of apoptotic mitochondria and/or enlarged endoplasmic reticulum and/or increased frequency of lipid droplets.

The main results of the experiments are that long time constant exposure of SCs at both TiO2 NPs concentrations inevitably led to an altered ultrastructure.

miR-1285-3p targets TPI1 to regulate the glycolysis metabolism signaling pathway of Tibetan sheep Sertoli cells

Glycolysis in sertoli cells (SCs) can provide energy substrates for the development of spermatogenic cells. Triose phosphate isomerase 1 (TPI1) is one of the key catalytic enzymes involved in glycolysis. However, the biological function of TPI1 in SCs and its role in glycolytic metabolic pathways are poorly understood. On the basis of previous research, we isolated primary SCs from Tibetan sheep and overexpressed TPI1 gene to determine its effect on the proliferation, glycolysis, and apoptosis of SCs. Secondly, we investigated the relationship between TPI1 and miR-1285-3p, and whether miR-1285-3p regulates the proliferation and apoptosis of SCs, and participates in glycolysis by targeting TPI1. Results showed that overexpression of TPI1 increased the proliferation rate and decreased apoptosis of SCs. In addition, overexpression of TPI1 altered glycolysis and metabolism signaling pathways and significantly increased the amount of the final product lactic acid. Further analysis showed that miR-1285-3p inhibited TPI1 by directly targeting its 3'untranslated region. Overexpression of miR-1285-3p suppressed the proliferation of SCs, and this effect was partially reversed by restoration of TPI1 expression. In summary, this study shows that the miR-1285-3p/TPI1 axis regulates glycolysis in SCs. These findings add to our understanding of the regulation of spermatogenesis in sheep and other mammals.
