An in vivo stable isotope labeling method to investigate individual matrix protein synthesis, ribosomal biogenesis, and chondrocyte proliferation in murine articular cartilage

<p>These experiments have used a stable-isotope method using <em>in vivo</em> deuterium oxide labeling and mass spectrometry to measure protein concentration, protein half-life, cell proliferation, and ribosomal biogenesis in a single sample of murine articular cartilage. We hypothesized that a 60-day labeling period would capture age-related declines in cartilage matrix protein content, protein synthesis rates, and chondrocyte proliferation. Knee cartilage was isolated from 25- and 90-week-old female C57BL/6J mice treated with deuterium oxide for 15, 30, 45 and 60 days. We measured protein abundance and half-lives using high resolution accurate mass spectrometry (HRAM) and d2ome data processing software.&nbsp;</p>

Facile synthesis of Cu2O nanorods in the presence of NaCl by SILAR method and its characterizations

<p>Cu2O nanorods have been deposited on soda-lime glass (SLG) substrates by the modified SILAR technique by varying the concentration of NaCl electrolyte into the pre-cursor complex solution. The structural, electrical, and optical properties of synthesized Cu2O nanorod films have been studied by a variety of characterization tools. Structural analyses by XRD confirmed the polycrystalline Cu2O phase with (111) preferential growth. Raman scattering spectroscopic measurements conducted at room temperature also showed characteristic peaks of the pure Cu2O phase. The surface resistivity of the Cu2O nanorod films decreased from 15,142 to 685 Ω.cm with the addition of NaCl from 0 to 4 mmol, and then exhibited an opposite trend with further addition of NaCl. The optical bandgap of the synthesized Cu2O nanorod films was observed as 1.88~2.36 eV, while the temperature-dependent activation energies of the Cu2O films were measured as about 0.14~0.21 eV. SEM morphologies demonstrated Cu2O nanorod as well as closely packed spherical grains with the alteration of NaCl concentration. The Cu2O phase of nanorods was found stable up to 230<i>℃</i><span><span> </span></span>corroborating the optical bandgap results of the same. The film fabricated in presence of 4 mmol of NaCl showed the lowest resistivity and activation energy as well comparatively uniform nanorod morphology. Our studies demonstrate that the nominal presence of NaCl electrolytes in the pre-cursor solutions has a significant impact on the physical properties of Cu2O nanorod films which could be beneficial in optoelectronic research.</p>

Synthesis of novel mesoporous silicate molecular sieves by Aerosol-assisted method for loading and release of drug

<p>The novel mesoporous silicate molecular sieves were synthesized with polyether F127 as the template by aerosol-assisted method for loading and release of ibuprofen. The synthesized samples were characterized by scanning electron microscopy (SEM), transmission electron microscopy (TEM), X-ray diffraction (XRD) and N2 adsorption-desorption isotherms. The drug ibuprofen was applied as a model drug to investigate the drug release behavior by UV spectrophotometry measurements. The investigation results demonstrate that mesoporous silicate molecular sieves by aerosol-assisted method are spherical with the core-shell structure. As the drug carrier, it has good structural stability and can achieve drug controlled release which is expected. Therefore, the aerosol-assisted synthesis method provides a new idea for the synthesis of sustained-release drug carriers.</p>

Data from: Assessment of intrathecal free light chain synthesis: comparison of different quantitative methods with the detection of oligoclonal free light chains by isoelectric focusing and affinity-mediated immunoblotting

Objectives: We aimed to compare various methods for free light chain (fLC) quantitation in cerebrospinal fluid (CSF) and serum and to determine whether quantitative CSF measurements could reliably predict intrathecal fLC synthesis. In addition, we wished to determine the relationship between free kappa and free lambda light chain concentrations in CSF and serum in various disease groups. Methods: We analysed 166 paired CSF and serum samples by at least one of the following methods: turbidimetry (Freelite™, SPAPLUS), nephelometry (N Latex FLC™, BN ProSpec), and two different (commercially available and in-house developed) sandwich ELISAs. The results were compared with oligoclonal fLC detected by affinity-mediated immunoblotting after isoelectric focusing. Results: Although the correlations between quantitative methods were good, both proportional and systematic differences were discerned. However, no major differences were observed in the prediction of positive oligoclonal fLC test. Surprisingly, CSF free kappa/free lambda light chain ratios were lower than those in serum in about 75% of samples with negative oligoclonal fLC test. In about a half of patients with multiple sclerosis and clinically isolated syndrome, profoundly increased free kappa/free lambda light chain ratios were found in the CSF. Conclusions: Our results show that using appropriate method-specific cut-offs, different methods of CSF fLC quantitation can be used for the prediction of intrathecal fLC synthesis. The reason for unexpectedly low free kappa/free lambda light chain ratios in normal CSFs remains to be elucidated. Whereas CSF free kappa light chain concentration is increased in most patients with multiple sclerosis and clinically isolated syndrome, CSF free lambda light chain values show large interindividual variability in these patients and should be investigated further for possible immunopathological and prognostic significance.

Understanding multidisciplinary product development through a synthesis mapping of approaches, processes, methods and tools from the scientific literature - Searchable map

<p>This document introduces a synthesis map of approaches, processes, methods and tools for multidisciplinary product development. The presented map is associated with an open-access article &ndash; see the citation box below&nbsp;&ndash; and differs from the one provided in the article in that it is searchable, which makes it easier to pinpoint the approaches, processes, methods and tools. This document comprises a legend and the map.&nbsp;</p> <p>To contextualize the map, multidisciplinary products arise from the integration of contributions from mechanical, electrical, electronics, software, and broadly information and communication technologies. <br>This &ldquo;multidisciplinarity&rdquo; implies a higher technical and organizational complexity that can invite companies to adapt their development. To support companies in the adaptation of their development and navigate the dense and fragmented scientific literature corpus on multidisciplinary product development, the authors proposed to graphically organize&nbsp;it.</p> <p>In a first phase, multidisciplinary product development was investigated by analyzing three specific types of multidisciplinary products that can be referred to as &ldquo;cyber-physical systems&rdquo;, &ldquo;mechatronics&rdquo;, and &ldquo;smart products and systems&rdquo; in the literature. This first phase led to three maps which graphically organized a total of&nbsp;236&nbsp;&ldquo;concepts and techniques&rdquo; identified from 167&nbsp;scientific papers through an extensive literature review and categorized based on a four-level model paired with a decision tree. A second phase, which result is represented below, introduced some simplifications and filters to the initial maps, which narrow down the number of concepts and techniques to 56. The following map represents a preliminary database of concepts and techniques for multidisciplinary product development and serves to support companies in their transformation from the product development perspective by providing them with a synthesized overview of the related literature. This work is particularly suited for companies and researchers looking for getting acquainted with the scientific literature related to multidisciplinary product development and how the different concepts and techniques can be associated.</p>
