Exposure to 3, 3', 4, 4', 5-pentachlorobiphenyl (PCB 126) causes widespread DNA hypomethylation in adult zebrafish testis
<p>Exposure to environmental toxicants during preconception have been shown to affect offspring health and epigenetic mechanisms such as DNA methylation are hypothesized to be involved in adverse outcomes. However, studies addressing the effects of exposure to environmental toxicants during preconception on epigenetic changes in gametes are limited. The objective of this study is to determine the effect of preconceptional exposure to a dioxin-like PCB (PCB126) on DNA methylation and gene expression in testis. Adult zebrafish were exposed to 3 and 10 nM PCB126 for 24 hours and testis tissue was sampled at 7 days post-exposure for histology, DNA methylation and gene expression profiling. Reduced Representation Bisulfite Sequencing revealed 37 and 92 differentially methylated regions (DMRs) in response to 3 and 10nM PCB126 exposures, respectively. Among them 19 DMRs were found to be common between both PCB126 treatment groups. Gene ontology (GO) analysis of DMRs revealed that enrichment of terms such as RNA processing, iron-sulfur cluster assembly and gluconeogenesis. Gene expression profiling showed differential expression of 40 and 1621 genes in response to 3 and 10nM PCB126 exposures, respectively. GO analysis revealed differential expression of genes related to xenobiotic metabolism, oxidative stress and immune function. There is no overlap in the GO terms or individual genes between DNA methylation and RNAseq results, but functionally many of the altered pathways have been shown to cause spermatogenic defects. Our results indicate that exposure to dioxin-like PCBs during preconception could affect testicular function by altering DNA methylation patterns, with significant implications for reproductive health. </p>

Data from: Sperm competition and the evolution of precopulatory weapons: testis size and amplexus position, but not arm strength, affect fertilization success in a chorusing frog
Trade-offs between pre- and postcopulatory traits influence their evolution, and male expenditure on such traits is predicted to depend on the number of competitors, the benefits from investing in weapons, and the risk and intensity of sperm competition. Males of the chorusing frog Crinia georgiana use their arms as weapons in contest competition. Previously, we showed that increased numbers of rivals elevated the risk and intensity of sperm competition due to multimale amplexus, and caused a reversal in the direction of precopulatory selection on arm girth. Here we focused on the factors affecting postcopulatory fertilization success during group spawning, using paternity data from natural choruses. Competitive fertilization success depended on the time spent amplexed and amplexus position. Relative testes size but not arm girth, contributed to fertilization success, but the effect of testes size depended on amplexus position. Our findings offer within species empirical support for recent sperm competition models that incorporate precopulatory male-male competition, and show why an understanding of the evolution of animal weapons requires a consideration of both pre- and postcopulatory episodes of sexual selection.

Porcine reproductive and respiratory syndrome virus infects the reproductive system of male piglets and impairs development of the blood–testis barrier
<p>Porcine reproductive and respiratory syndrome virus (PRRSV) causes a highly contagious disease that threatens the global swine industry. Recent studies have focused on the damage that PRRSV causes to the reproductive system of male pigs, although <span>pathological research is lacking</span>. Therefore, <span>we </span><span>examined the pathogenic mechanisms in male piglets infected with PRRSV</span>. Gross and histopathological changes indicated that PRRSV affected the entire reproductive system, as confirmed via immunohistochemical analysis. PRRSV infected Sertoli cells and spermatogonia. To test the <span>new </span>hypothesis that PRRSV infection in piglets impairs blood–testis barrier (BTB) development, we investigated the <span>pathology</span> of PRRSV damage in the BTB. PRRSV infection significantly decreased the quantity and proliferative capacity of Sertoli cells constituting the BTB. Zonula occludens-1 and β-catenin were downregulated in cell–cell junctions. Transcriptome analysis revealed that several crucial genes and signaling pathways involved in the growth and development of Leydig cells, Sertoli cells, and tight junctions in <span>the </span>testes were downregulated. <span>Apoptosis, necroptosis</span>, inflammatory, and oxidative stress-related pathways were activated, whereas hormone secretion-related pathways were inhibited. Many Sertoli cells and spermatogonia underwent apoptosis during early differentiation. Infected piglets exhibited disrupted androgen secretion, leading to significantly reduced testosterone and anti-Müllerian hormone levels. A cytokine storm occurred, notably upregulating cytokines such as tumor necrosis factor-α and interleukin-6. Markers of oxidative-stress damage (i.e., H2O2, malondialdehyde, and glutathione) <span>were upregulated</span>, whereas antioxidant-enzyme activities (i.e., superoxide dismutase, total antioxidant capacity, and catalase) <span>were downregulated</span>. Our results demonstrated that PRRSV infect<span>ed</span> multiple organs in the male reproductive system, <span>which imparied</span> growth in the BTB. </p>
