Data from: Ovarian cycling and reproductive state shape the vaginal microbiota in wild baboons

Background: The vaginal microbiome is an important site of bacterial-mammalian symbiosis. This symbiosis is currently best characterized for humans, where lactobacilli dominate the microbial community and may help defend women against infectious disease. However, lactobacilli do not dominate the vaginal microbiota of any other mammal studied to date, raising key questions about the forces that shape the vaginal microbiome in non-human mammals. Results: We used Illumina sequencing of the bacterial 16S rRNA gene to investigate variation in the taxonomic composition of the vaginal microbiota in 48 baboons (Papio cynocephalus), members of a well-studied wild population in Kenya. Similar to prior studies, we found that the baboon vaginal microbiota was not dominated by lactobacilli. Despite this difference, and similar to humans, reproductive state was the dominant predictor of baboon vaginal microbiota, with pregnancy, postpartum amenorrhea, and ovarian cycling explaining 18% of the variance in community composition. Furthermore, among cycling females, a striking 39% of variance in community composition was explained by ovarian cycle phase, with an especially distinctive microbial community around ovulation. Peri-ovulatory females exhibited the highest relative abundance of lactic acid-producing bacteria compared to any other phase, with a mean relative abundance of 44%. To a lesser extent, sexual behavior, especially a history of shared sexual partners, also predicted vaginal microbial similarity between baboons. Conclusions: Despite striking differences in their dominant microbes, both human and baboon vaginal microbiota exhibit profound changes in composition in response to reproductive state, ovarian cycle phase, and sexual behavior. We found major shifts in composition during ovulation, which may have implications for disease risk and conception success. These findings highlight the need for future studies to account for fine-scale differences in reproductive state, particularly differences between the various phases of the ovarian cycle. Overall, our work contributes to an emerging understanding of the forces that explain intra- and inter-individual variation in the mammalian vaginal microbiome, with particular emphasis on its role in host health and disease risk.

The canine vaginal microbiome in heat does not influence fertility in healthy breeding bitches

A healthy and balanced vaginal microbiome is often thought to be an important prerequisite for successful breeding and healthy litters. Previous studies investigating the influence of canine vaginal bacteria on fertility mostly relied on aerobic culturing. In recent years, culture-independent methods, such as next-generation sequencing (NGS), have become popular. With the ability to analyze the microbiome as a whole, research in this field has made notable advances. This is the first study to correlate NGS data of the canine vaginal microbiome in heat with fertility data. Healthy breeding bitches (n=80) presented for routine pre-breeding examination were sampled during early heat and mated after ovulation. A vaginal sample was taken for NGS analysis and microbiological culture. Additionally, a blood sample was collected. Fertility data (mating, pregnancy, delivery, litter size) were assessed. NGS revealed a diverse microbiome in all the samples. Bioinformatics and statistical analysis did not provide evidence of larger differences in the microbiome of those bitches that became pregnant and those that did not.

Comparison of different suture techniques for laparoscopic vaginal cuff closure

Laparoscopic hysterectomy is a commonly performed procedure. However, one high-risk complication is vaginal cuff dehiscence. Currently, there is no standardization regarding thread material or suturing technique for vaginal cuff closure. Therefore, this study aimed to compare extracorporeal and intracorporeal suturing techniques for vaginal cuff closure using a pelvic trainer model. Eighteen experts in laparoscopic surgery performed vaginal cuff closures with interrupted sutures using intracorporeal knotting, extracorporeal knotting and continuous, unidirectional barbed sutures. While using an artificial tissue suturing pad in a pelvic trainer, experts performed vaginal cuff closure using each technique according to block randomization. Task completion time, tension resistance, and the number of errors were recorded. After completing the exercises, participants answered a questionnaire concerning the suturing techniques and their performance. Experts completed suturing more quickly (p < 0.001, p < 0.001, respectively) and with improved tension resistance (p < 0.001, p < 0.001) when using barbed suturing compared to intracorporeal and extracorporeal knotting. Furthermore, the intracorporeal knotting technique was performed faster (p = 0.04) and achieved greater tension resistance (p = 0.023) compared to extracorporeal knotting. The number of laparoscopic surgeries performed per year was positively correlated with vaginal cuff closure duration (p = 0.007). Barbed suturing was a time-saving technique with improved tension resistance for vaginal cuff closure.

Data from: Vaginal lactobacilli produce anti-inflammatory beta-carboline (BC) compounds. RNAseq dataset of BC-treated human monocytes.

Primary human monocytes were isolated from peripheral blood mononuclear cells via CD14+ magnetic selection. Monocytes were then treated with 3 beta-carboline compounds isolated from the supernatant of vaginal Lactobacillus crispatus strain MV-1A-US (HM-637). The beta-carbolines are labled 322/BC1; 325/BC3 and 361/BC6. We find that BC6 suppresses inflammatory signaling genes in untreated and in LPS-treated monocytes. FIles include raw reads, count table and code for DESeq2-generated differentially expressed genes.

Assessing the prevalence of Female Genital Schistosomiasis and comparing the acceptability and performance of health worker-collected and self-collected cervical-vaginal swabs using PCR testing among women in North-Western Tanzania: the ShWAB study

Female genital schistosomiasis (FGS) is a severe neglected disease, caused by infection with Schistosoma haematobium. The WHO has prioritized the improvement of diagnostics for FGS and previous studies have explored the PCR-based detection of Schistosoma DNA on genital specimens, with encouraging results. We aimed to determine the prevalence of FGS among women living in an endemic district in North-western Tanzania, applying and preliminary comparing self-collected and operator-collected cervical-vaginal swabs followed by PCR, and to assess the acceptability of these sampling procedures.

A INTERVENTIONAL CROSS SECTIONAL STUDY BETWEEN INTRA VAGINAL PLATELET RICH PLASMA AND ESTROGEN FOR TREATMENT OF GENITO-URINARY SYNDROME IN POST MENOPAUSAL WOMEN.

Genitourinary syndrome of menopause (GSM) is a new term for a condition more renowned as atrophic vaginitis. It is used to describe a variety of menopausal symptoms and signs that are related to the physical changes of the vulva, vagina and lower urinary tract. The etiology of GSM is secondary to decreased levels of endogenous estrogens and represents a common but underreported condition. Currently Intra-Vaginal application of oestrogen has been considered an effective treatment of atrophic vaginitis.Recently, PRP has becoming popular as a non-operative treatment option for GSM symptoms. Our study focuses on the use platelet rich plasma for the treatment of GSM as it has natural mechanisms of cell regeneration and requires less frequent dosing and have better compliance when compared to estrogen creams. Purpose of the study is to compare the effectiveness of intra vaginal estrogen cream with intravaginal PRP in treatment of GSM.
