Outdoor mesocosm study evaluating how mass, NaCl tolerance, and pesticide tolerance affect oxidative stress biomarkers (CAT, SOD, GR, GPx, TBARS) in larval wood frogs (Rana sylvatica) exposed to baseline and NaCl-contaminated conditions, 2019

Biomarkers of oxidative stress can aid in wildlife monitoring by allowing conservationists to detect sublethal environmental shifts. However, interpretation of stress responses can be complicated by multiple interacting factors (e.g., individual development, evolved physiological tolerance to stressors) which alter biomarker expression. Here, we investigated how individual ontogenetic traits and population-level tolerance traits influence oxidative stress responses under baseline and contaminated environmental conditions. For our model contaminant, we used NaCl (common freshwater contaminant due to factors such as coastal flooding, irrigation, airborne salt circulation, drought, runoff from road deicing salts). For our model wildlife populations, we used larval wood frogs (Rana sylvatica) from six noninteracting populations known to vary in two population-level tolerance traits: NaCl tolerance (calculated as average time to death from lethal NaCl exposure) and pesticide tolerance (determined by proxy of distance to agriculture - a consistent and highly repeatable relationship). At an outdoor research facility, R. sylvatica tadpoles were exposed to either baseline conditions (0 g/L NaCl added) or NaCl-contaminated conditions (1 g/L NaCl added for 21 days, then reduced to 0.5 g/L NaCl). Exposures were conducted in individual units with 40 replicates per population for each treatment. The experiment was terminated per individual to capture the full term of larval development (Developmental stage: Gosner stage 36), lasting between 33-48 days. For each individual, we measured mass, Snout-Vent-Length, and developmental stage before processing for biomarker expression. Growth, development, activity, and survival of wood frog tadpoles from different populations in eastern NY in 2022.

These data were collected as two lab experiments that examined larval (i.e., tadpole) wood frog populations. The first was a time-to-death experiment in which we exposed individuals tadpoles to either a control or lethal concentrations of NaCl. The second was a growth and development experiment in which we raised tadpoles from the same populations in a sublethal concentration of NaCl for several weeks to examine their growth and development (i.e. Gosner developmental stage).

Leaf litter quality induces morphological changes in wood frog (Lithobates sylvaticus) metamorphs, Oakland University Biological Preserve (MI, USA) 2010.

For organisms that exhibit complex life cycles, resource conditions experienced by individuals before metamorphosis can strongly affect phenotypes later in life. Such resource-induced effects are known to arise from variation in resource quantity, yet little is known regarding effects stemming from variation in resource quality (e.g., chemistry). For larval anurans, we hypothesized that variation in resource quality will induce a gradient of effects on metamorph morphology. We conducted an outdoor mesocosm experiment in which we manipulated resource quality by rearing larval wood frogs (Lithobates sylvaticus) under 11 leaf litter treatments. The litter species represented plant species found in open- and closed-canopy wetlands and included many plant species of current conservation concern (e.g., green ash, common reed). Consistent with our hypothesis, we found a gradient of responses for nearly all mass-adjusted morphological dimensions. Hindlimb dimensions and gut mass were positively associated with litter nutrient content and decomposition rate. In contrast, forelimb length and head width were positively associated with concentrations of phenolic acids and dissolved organic carbon. Limb lengths and widths were positively related with the duration of larval period, and we discuss possible hormonal mechanisms underlying this relationship. There were very few, broad differences in morphological traits of metamorphs between open- and closed-canopy litter species or between litter and no-litter treatments. This suggests that the effects of litter on metamorph morphology are litter species-specific, indicating that the effects of changing plant community structure in and around wetlands will largely depend on plant species composition.

Relyea, R. A. 2002. Local population differences in phenotypic plasticity: Predator-induced changes in wood frog tadpoles. Ecological Monographs 72:77-93

Taxa that are divided into separate populations with low levels of interpopulation dispersal have the potential to evolve genetically based differences in their phenotypes and the plasticity of those phenotypes. These differences can be due to random processes, including genetic drift and founder effects, or they can be the result of different selection pressures among populations. I investigated population-level differences in predator- induced phenotypic plasticity in eight populations of larval wood frogs (Rana sylvatica) over a small geographic scale (interpopulation distances of 0.3–8 km). Using a common-garden experiment containing predator and no-predator environments, I found population differences in behavior, morphology, and life history. These responses exhibited a habitat-related pattern: the four populations from closed-canopy ponds did not differ from each other in any of their phenotypes whereas the four populations from opencanopy ponds did differ from each other in these traits. This phenotypic pattern matches the pattern of competitors and predators found in these two types of ponds. Based on two years of pond surveys, the four closed-canopy ponds contained very similar competitor and predator assemblages while the assemblages of the four open-canopy ponds were more diverse and highly variable among open-canopy ponds. When combined with past studies, which demonstrate that predators and competitors select for alternative behavioral and morphological traits, these patterns suggest that the population differences may have arisen via natural selection and not via random mutation or drift. In a second experiment, I cross-transplanted two of the populations into each other’s ponds to determine if the populations were locally adapted to the conditions of their native pond (using low and high competition crossed with the presence or absence of a lethal predator). Model data to investigate wood frog abundance in 17-year post harvest variable retention mixed wood forests

<p>Variable retention forest harvesting aims to reduce negative effect of harvesting on forest biodiversity, but its effectiveness is not well understood for many taxa. To better understand the effects of variable retention forest management and environmental features on amphibians, we used pitfall traps to capture wood frogs (<em>Lithobates sylvaticus</em>) across 4 levels of retention harvest (clearcut [0%], 20%, 50%, and unharvested control [100%]), and 2 forest types (deciduous and coniferous), in 17-year post-harvest forests in northwest Alberta. We mapped breeding sites and used a terrain moisture index (Depth-to-Water) derived from airborne LiDAR to examine relationships between relative abundance, breeding site proximity and soil moisture. Retention level alone had no effect on relative abundance, but in late summer (July and August) there was a significant interaction between retention level and forest type: capture rates decreased with amount of retention for deciduous forests, but increased with amount of retention in conifer forests. During late summer, capture rates were higher in conifer forests than in deciduous forests, with soil moisture (lower Depth-to-Water) positively related to capture rates. Though timber retention may be beneficial to wood frogs in the short-term, any impacts of forest harvesting on wood frog abundance was undetectable in stands 17 years post-harvest.  </p>

Data from: Phenological delay despite warming in wood frog (Rana sylvatica) reproductive timing: a 20-year study

<p>Across all taxa, amphibians exhibit some of the strongest phenological shifts in response to climate change. As climates warm, amphibians and other animals are expected to breed earlier in response to temperature cues. However, if species use fixed cues such as daylight, their breeding timing might remain fixed, potentially creating disconnects between their life history and environmental conditions. Wood frogs (<i>Rana </i>sylvatica) are a cold-adapted species that reproduce in early spring, immediately after breeding ponds are free of ice. We used long-term surveys of wood frog oviposition timing in 64 breeding ponds over 20 years to show that, despite experiencing a warming of 0.29 °C per decade in annual temperature, wood frog breeding phenology has shifted later by 2.8 days since 2000 (1.4 days per decade; 4.8 days per °C). This counterintuitive pattern is likely the result of changes in the timing of snowpack accumulation and melting. Finally, we used relationships between climate and oviposition between 2000 and 2018 to hindcast oviposition dates from climate records to model longer-term trends since 1980. Our study indicates that species can respond to fine-grained seasonal climate heterogeneity within years that is not apparent or counterintuitive when related to annual trends across years.</p>

Temperature-mediated tradeoff between development and performance in larval wood frogs (Rana sylvatica)

<p>Countergradient variation has been detected in diverse taxa. In a common manifestation, individuals from colder environments develop faster than conspecifics from warmer environments when placed in a common garden. Where such a pattern exists, it implies a tradeoff: individuals from warmer environments have intrinsic rates of development lower than those demonstrated by other individuals of the same species. We explored a tradeoff between development rate and locomotor performance in the wood frog (Rana sylvatica), an amphibian for which countergradient variation has been well documented. We reared wood frogs from 10 populations under two temperature regimes, bracketing the temperatures observed in local natural ponds. Individuals reared under warmer conditions developed more rapidly but exhibited burst speeds 20% lower than individuals reared under colder conditions. The shape of the relationship was consistent across the 10 populations and thus, we found no evidence of countergradient variation in performance. Burst speed assays of wild-caught tadpoles from the same populations showed a similar but nonsignificant trend, with greater variability among ponds. Overall, our findings support the existence of a tradeoff that may be of broad importance and which may help explain widespread occurrence of countergradient variation.</p>

Data from: Development, validation, and evaluation of an assay for the detection of wood frogs (Rana sylvatica) in environmental DNA

We developed and describe a qPCR assay for the detection of wood frogs (Rana sylvatica) using environmental DNA (eDNA) sampling. A single primer set was designed to amplify a 115-bp region of the wood frog cytochrome B gene and assessed for target specificity. There was no evidence of amplification in 11 non-target species. We evaluated the utility of the primer set in qPCR assay by conducting geo-referenced eDNA field surveys in Interior Alaska. Results indicate that the assay consistently detects wood frog DNA in the environment to 1.83 × 10−3 pg/μL. The assay provides a complement to traditional survey methods and can be readily applied in a wider conservation and management context.

Data from: The effect of habitat fragmentation on finescale population structure of wood frogs (Rana sylvatica)

We examined the impact of recent anthropogenic habitat fragmentation on the genetic structure of wood frog (Rana sylvatica) breeding sites in Wellington County of Ontario, Canada. In addition to geographic distance (average pairwise distance ~22 km, greatest distance ~50.22 km), four landscape features hypothesized to contribute to genetic differentiation between breeding sites were considered: road density, a major highway (highway 401), canopy cover, and watershed discontinuity. Analysis of data from 396 samples across nine breeding sites using eight microsatellite DNA loci, revealed a small degree of significant genetic structure between breeding sites. The presence of highway 401 and road density were correlated with small but statistically significant structure observed between several groups of sites. One outlier breeding site outside of Wellington County located within the city of Toronto, had significantly lower allelic richness and much larger population differentiation with the Wellington sites. Our data suggest that recent fragmentation has had an effect on wood frog population structure and also demonstrate the importance of dispersal for this species in maintaining levels of genetic diversity.

Female choice scores and Peak Frequency and Duration in calls from Wood frog chorus recordings

<p>A limitation in bioacoustic studies has been the inability to differentiate individual sonic contributions from group-level dynamics. We present a novel application of acoustic-camera technology to investigate how individual wood frogs calls influence chorus properties, and how variation influences mating opportunities. We recorded mating calls and used playback trials to gauge preference for different chorus types in the laboratory. Males and females preferred chorus playbacks with low variance in dominant frequency. Females preferred choruses with low mean peak frequency. Field studies revealed more egg masses laid in ponds where males chorused with low variance in dominant frequency. We also noted a trend towards more egg masses laid in ponds where males called with low mean frequency. Nearest neighbor distances influenced call timing (neighbors called in succession) and distances increased with variance in chorus frequency. Results highlight the potential fitness implications of individual-level contributions to a bioacoustic signal produced by groups.</p>

Ranavirus infection-induced avoidance behavior in wood frog juveniles: Do amphibians socially distance?

<p>Hosts may limit exposure to pathogens through changes in behavior, such as avoiding infected individuals or contaminated areas. Here, we tested for a behavioral response to ranavirus infection in juvenile wood frogs (<em>Rana sylvatica</em>) because the majority of dispersal between populations occurs during this life stage. We hypothesized that if infections are transmissible and detectable at this life stage, then susceptibles would display avoidance behaviors when introduced to an infected conspecific. Despite no apparent signs of infection, we observed a greater distance between susceptible-infected pairs, compared to pairs of either two infected or two susceptible animals. Further, distances between susceptible-infected pairs were positively related to the infection intensity of the focal exposed frog, suggesting the cue to avoid infected conspecifics may become more detectable with more intense infections. Although we did not quantify whether transmission was affected by their distancing, our findings suggest that juvenile frogs have the potential to reduce terrestrial transmission of ranaviruses through avoidance behaviors.</p>
