Bar=20 &mu;m. <strong><em>B.</em></strong> mRNA expression of myosin7a and prestin decreased sharply after 7days and nestin increased on day 9. Aminoglycosides, such as kanamycin and gentamicin reduce the viability of HEI-OC1 by about 10%. Therefore, kanamycin concentration was selected to 10mM, and 50uM was selected for furosemide which does not affect cells. <strong><em>B.</em></strong> When treated with kanamycin and furosemide, cell viability was reduced by 10% more than kanamycin alone. *P&lt;0.05, **P&lt;0.005 and ***P&lt;0.0001 in comparison to untreated group. <sup>##</sup>P&lt;0.005 is compared with kanamycin-furosemide treatment group.</p> <p>&nbsp;</p> <p>&nbsp;</p> <p><strong>Supplementary Figure S3. </strong>Effects of kanamycin and furosemide on HEI-OC1. <strong><em>A.</em></strong> The expression of ER stress-related markers was increased in all three groups and there was no difference in the combination treatment of kanamycin-furosemide. <strong><em>B.</em></strong> As expected, cleaved caspase -3, -9 and p-p53 from kanamycin-furosemide treated HEI-OC1 were not detected. Staurosporin was used as a positive control for caspase-3 and -9. **P&lt;0.005 and ***P&lt;0.0001 in comparison to untreated group.</p> <p>&nbsp;</p> <p><strong>Supplementary Figure S4. </strong>NOX inhibition itself did not cause any damage to the cells. NOX inhibitors, DPI and APO did not affect the IL-1ꞵ expression and even slightly reduced the expression of TNF-&alpha;. A slight increase in the IL-6 mRNA level was not significant.</p> <p>&nbsp;</p>
