Dataset related to the publication "New Technique for Probing the Protecting Character of the Solid Electrolyte Interphase as a Critical but Elusive Property for Pursuing Long Cycle Life Lithium-Ion Batteries"
<p>The formation of a protecting nano-layer, so-called Solid Electrolyte Interphase (SEI), on the negative electrode of Li-ion batteries (LIBs) from product precipitation of the cathodic decomposition of the electrolyte is a blessing since the electrically-insulating nature of this nano-layer protect the electrode surface preventing continuous electrolyte decomposition and enabling the large nominal cell voltage of LIBs, e.g. 3.3 – 3.8 V. Thus, the protecting performance of the nano-layer SEI is essential for LIBs to achieve long cycle life. Unfortunately, evaluation of this critical property of the SEI is not trivial. Herein, a new, cheap and easily-implementable methodology is presented to estimate the protecting quality of the SEI; the redox-mediated enhanced coulometry. The key element of the methodology is the addition of a redox-mediator in the electrolyte during degassing step (after the SEI formation cycle). The redox-mediator leads to an internal self-discharge process that is inversely proportional to the protecting character of the SEI. And the self-discharge process results in an easily-measurable decrease in coulombic efficiency. The influence of vinylene carbonate as electrolyte additive in the resulting SEI is used as case study to showcase the potential of the proposed methodology</p>

Data from: Towards high-resolution modeling of small molecule - ion channel interactions
<p>Ion channels are critical drug targets for a range of pathologies, such as epilepsy, pain, itch, autoimmunity, and cardiac arrhythmias. To develop effective and safe therapeutics, it is necessary to design small molecules with high potency and selectivity for specific ion channel subtypes. There has been increasing implementation of structure-guided drug design for the development of small molecules targeting ion channels. We evaluated the performance of two Rosetta ligand docking methods, RosettaLigand and GALigandDock, on structures of known ligand - cation channel complexes. Ligands were docked to voltage-gated sodium (NaV), voltage-gated calcium (CaV), and transient receptor potential vanilloid (TRPV) channel families. For each test case, RosettaLigand and GALigandDock methods were able to frequently sample a ligand binding pose within 1-2 Å root mean square deviation (RMSD) relative to the experimental ligand coordinates. However, RosettaLigand and GALigandDock scoring functions cannot consistently identify experimental ligand coordinates as top-scoring models. Our study reveals that the proper scoring criteria for RosettaLigand and GALigandDock modeling of ligand - ion channel complexes should be assessed on a case-by-case basis using sufficient ligand and receptor interface sampling, knowledge about state specific interactions of the ion channel and inherent receptor site flexibility that could influence ligand binding.</p>

Mobility-assisted psuedo-MS3 sequencing of protein ions
<p>The sequencing of intact proteins within a mass spectrometer has many benefits but is frequently limited by the fact that tandem mass spectrometry (MS/MS) techniques often generate poor sequence coverages when applied to protein ions. To overcome this limitation exotic MS/MS techniques that rely on lasers and radical chemistry have been developed. These techniques generate high sequence coverages, but they require specialized instrumentation, create products through multiple dissociation mechanisms, and often require long acquisition times. Recently, we demonstrated that protein ions can be dissociated in a trapped ion mobility spectrometry (TIMS) device prior to mobility separation in a commercial timsTOF. All generated product ions were distributed throughout the mobility dimension and this separation enabled deconvolution of complex tandem mass spectra and could enable facile pseudo-MS3 interrogation of generated product ions with the downstream quadrupole and collision cell. A second activation step improves sequence coverage because the most labile bonds have been depleted during the first dissociation and subsequent dissociation events are more evenly distributed throughout the product ion backbone. In this work, we explore the potential of this mobility-assisted pseudo-MS3 (MAP) method on a commercial timsTOF and timsTOF Pro 2. We demonstrate that while MAP only generates 92% of the sequence coverage of the most effective MS/MS technique, it accomplished this feat in 1.5 mins and could be facilely integrated with liquid chromatographic separations.</p>
Data from: Remote neurostimulation through an endogenous ion channel using a near infrared light-activatable nanoagonist
<p>The development of noninvasive approaches to precisely control neural activity in mammals is highly desirable. Here we utilized the ion channel TRPA1 as a proof of principle, demonstrating remote near-infrared (NIR) activation of endogenous channels in the neural structures of living mice through an engineered nanoagonist. This achievement enables specific neurostimulation in wild-type, non-genetically modified mice. Initially, target-based screening identified flavins as photopharmacological agonists, allowing for the photoactivation of TRPA1 in sensory neurons upon UVA/blue light illumination. Subsequently, upconversion nanoparticles (UCNPs) were customized with an emission spectrum aligned to flavin absorption and conjugated with flavin adenine dinucleotide, creating a nanoagonist capable of NIR activation of TRPA1. Following the intrathecal injection of the nanoagonist, noninvasive NIR stimulation allows precise bidirectional control of nociception in mice through the remote activation of spinal TRPA1. This study demonstrates a noninvasive NIR neurostimulation method with the potential for adaptation to various endogenous ion channels and neural processes by combining photochemical toolboxes with customized UCNPs.</p>
