Toxicological Effect and Risk Assessment (TERA) Knowledge Graph
<p>The Toxicological Effect and Risk Assessment (TERA) Knowledge Graph is based on chemical effect data from U.S. EPA ECOTOX. This data is aligned to non-proprietary identifiers using ontology alignment tools and external sources (eg. wikidata). This enables the use of external chemical knowledge graphs (eg. ChEBI, PubChem, Mesh, "Courtesy of the U.S. National Library of Medicine"). Data from: Temperature alters the toxicological impacts of plant terpenoids on the polyphagous model herbivore Vanessa cardui
<p>Terpenes are a major class of secondary metabolites present in all plants, and long hypothesized to have diversified in response to specific plant-herbivore interactions. Herbivory is a major biotic interaction that plays out across broad temporal and spatial scales that vary dramatically in temperature regimes, both due to climatic variation across geographic locations as well as the effect of seasonality. In addition, there is an emerging understanding that global climate change will continue to alter the temperature regimes of nearly every habitat on Earth over the coming centuries. Regardless of source, variation in temperature may influence herbivory, in particular via changes in the efficacy and impacts of plant defensive chemistry. This study aims to characterize temperature-driven variation in toxicological effects across several structural classes of terpenes in the model herbivore Vanessa cardui, the painted lady butterfly. We observed a general increase in monoterpene toxicity to larvae, pupa, and adults at higher temperatures, as well as an increase in development time as terpene concentration increased. Results obtained from this study yield insights into possible drivers of seasonal variation in plant terpene production as well as inform effects of rising global temperatures on plant-insect interactions. In the context of other known effects of climate change on plant-herbivore interactions like carbon fertilization and compensatory feeding, temperature-driven changes in plant chemical defense efficacy may further complicate the prediction of climate change impacts on the fundamental ecological process of herbivory.</p>

Data from 90-day quantitative inhalation toxicology study evaluating the dose-response and fate in the lung and pleura of chrysotile-containing brake dust
<p>The final results from this multi-dose, 90-day inhalation toxicology study in the rat with life-time post-exposure observation have shown a significant fundamental difference in pathological response and tumorgenicity between brake dust generated from brake pads manufactured with chrysotile or from chrysotile alone in comparison to the amphiboles, crocidolite and amosite asbestos.</p> <p>The groups exposed to brake dust showed no significant pathological or tumorigenic response in the respiratory track compared to the air control group at exposure concentrations and deposited doses well above those at which humans have been exposed. Slight alveolar/interstitial macrophage accumulation of particles was noted. Wagner grades were 1–2 (1 = control group), similar to the TiO2 particle control group. Chrysotile was not biopersistent, exhibiting in the lung a deterioration of its matrix which results in breakage into particles and short fibers which can be cleared by alveolar macrophages and which can continue to dissolve. Particle-laden macrophage accumulation was observed, leading to a very-slight interstitial inflammatory response (Wagner grade 1–3). There was no peribronchiolar inflammation, occasional very-slight interstitial fibrosis (Wagner grade 4), and no exposure-related tumorigenic response.</p> <p>The pathological response of crocidolite and amosite compared to the brake dust and chrysotile was clearly differentiated by the histopathology and the confocal analysis. Crocidolite and amosite induced persistent inflammation, microgranulomas, persistent fibrosis (Wagner grades 4), and a dose-related lung tumor response. Confocal microscopy quantified extensive inflammatory response and collagen development in the lung, visceral and parietal pleura as well as pleural adhesions.</p> <p>These results provide a clear foundation for differentiating the innocuous effects of brake dust exposure from the adverse effects following amphibole asbestos exposure.</p>

FunShield4Med e-learning module on Computational Toxicology
<p>This is a two-module presentation about the use of computational methods in toxicology providing the audience with an introduction of molecular modelling techniques applied to toxicology assessment (first module, theoretical part). Taking advantage of selected case studies targeting ochratoxin A, interested persons are learning how in silico methods can be broadly applied to investigate the toxicodynamics and toxicokinetic of mycotoxins (second module, e-learning part), supporting the early stage of risk assessment (i.e. the hazard identification and characterization). The two modules are explaining in depth and step-by-step the procedure used and the underpinning rationale and base of knowledge to make anyone virtually able to reproduce such kind of analysis.</p>

Primary hepatocyte culture from Oreochromis niloticus fish as a tool for environmental toxicology
<p>Accumulated chemicals have been impairing water quality over decades of human activities. Organisms such as fish are often used for risk assessment, as chemical analysis is insufficient to assess the impact on health. Consequently, researchers have faced challenges in ecotoxicology, concerning the use of animals. Primary and cell line cultures are good alternatives for reducing animal use. This study aims to evaluate the use of primary hepatocytes culture from Oreochromis niloticus as an in vitro tool for screening environmental contaminants and pollutants. Metal cadmium (Cd) is the pollutant model.</p>
