Microscopy image sequences and annotated kymographs of laser ablation experiments in Drosophila embryos

Content

This dataset contains 15 2D time-lapse fluorescence microscopy image sequences recorded with confocal laser-scanning microscopy. Each movie shows an epithelial tissue laser nanoablation experiment conducted in a Drosophila embryo.

For each sequence, the dataset contains kymographs (one-dimensional space-time plots) of a supracellular cable that is cut during the ablation, and manually created tracks of visible features, such as the resulting cut ends. These tracks allow to estimate, for instance, recoil velocities of the cut tissue and may be used to evaluate automated methods for estimating said velocities.

This dataset is used in the manuscript to evaluate various variational approaches for joint motion estimation and source identification:

L. Joint Motion Estimation and Source Identification using Convective Regularisation with an Application to the Analysis of Laser Nanoablations. 2019.

Description

The movies depict a square region of approximately \(42.2 \times 42.2 \, \mathrm{\mu m}^{2}\) at a spatial resolution of \(250 \times 250\) pixels. A typical sequence contains between 60 and 100 frames. They temporal interval between recorded frames was \(727.67 \, \mathrm{ms}\).

Each sequence features cell membranes labelled with E-cadherin:GFP and shows a single plasma-induced laser nanoablation. The destructed tissue region is roughly of \(2 \, \mathrm{\mu m}\) length. This ablation is expected to have a width of the order of the size of one pixel. During the ablation the acquisition is paused, resulting in a black image.

For the used microscopy techniques and for the preparation of flies, as well as for the details of the laser ablation method, see the paper:

E. Chemical and sediment characteristics of ice cores collected from the ablation zones of Canada, Commonwealth, Howard, Hughes, and Seuss Glaciers in the McMurdo Dry Valleys, Antarctica from 2015 to 2019

This data package contains chemical and sediment characteristics of ice cores collected from the ablation zones of five glaciers in Taylor Valley, located in the McMurdo Dry Valleys region of Antarctica, during the 2015-16, 2016-17, 2017-18, and 2018-19 austral summers. Specifically, shallow ice cores were collected from the ablation zones of Hughes, Howard, Seuss, Commonwealth, and Canada Glaciers in order to characterize the spatial and temporal evolution of ice chemistry and sediment concentration across Taylor Valley. Cores were collected in triplicate from each sampling location and measured 79 mm in diameter and up to 1 m in depth. Cores were sectioned in 5 cm (0-25 cm depth) and 25 cm increments (25 cm to the maximum depth of the core) prior to analyzing chemical and sediment characteristics.

Data from: Ablation of Hoxb3os in mouse polycystic kidney disease exacerbates cystogenesis

Autosomal dominant polycystic kidney disease (ADPKD) is a monogenic disorder characterized by the formation of cysts in the kidney and is primarily caused by mutations in two genes, PKD1 and PKD2. In this study, we investigated the role of the long noncoding RNA (lncRNA) Hoxb3os in ADPKD by ablating its expression in the mouse. Hoxb3os-null mice were viable and had grossly normal kidney morphology but displayed activation of mTOR/Akt signaling and subsequent increase in kidney cell proliferation. To determine the role of Hoxb3os in cystogenesis, we crossed the Hoxb3os-null mouse to two orthologous Pkd1 mouse models: Pkhd1/Cre;Pkd1F/F (rapid cyst progression) and Pkd1RC/RC (slow cyst progression). Ablation of Hoxb3os exacerbated cyst growth in both models. To gain insight into the mechanism whereby Hoxb3os inhibition promotes cystogenesis, we performed proteomic analysis of mTOR/Akt pathway between Pkd1 single-knockout (SKO) and Pkd1-Hoxb3os double-knockout (DKO) mice. Compared to SKO, DKO mice presented with enhanced levels of total and phosphorylated Rictor. This was accompanied by increased phosphorylation of AktSer473, a known mTORC2 effector site. Physiologically, kidneys from DKO mice displayed between 40-50% increase in cell proliferation. Results from this study indicated that ablation of Hoxb3os in mouse PKD exacerbated cystogenesis and dysregulated mTORC2.

Raw Data for the article: Atrial fibrillation ablation improves late survival after concomitant cardiac surgery

Objective: Preoperative atrial fibrillation (AF) increases risk of stroke, heart failure, and all-cause mortality after cardiac surgery. Despite encouraging results and guideline recommendations, surgical ablation (SA) for AF concomitant with other heart surgery remains low. In the current study we aimed to address the long-term mortality after SA concomitant with cardiac surgery.

Methods: This report pertains to the HEart surgery In atrial fibrillation and Supraventricular Tachycardia (HEIST) registry. We identified 20,765 adult patients (62% male) with preoperative AF who underwent conventional sternotomy heart surgery between 2010 and 2021 in 8 tertiary centers in Poland, Netherlands, and Italy. We used Cox proportional hazards models for computations and propensity score matching to minimize differences in baseline characteristics.

Results: Of included patients, 2755 (13.4%) underwent SA for AF. The highest rates of SA were observed for mitral interventions (mitral valve repair or replacement and tricuspid intervention, 25.2%), lowest for isolated coronary artery bypass grafting (6.2%). Patients in the SA group were younger (mean age 64.5 ± 9.0 years vs 68.7 ± 16.0 years; P < .001) and lower risk (mean European System for Cardiac Operative Risk Evaluation [EuroSCORE] II, 4.1 vs 5.7; P < .001). During the 11-year study period, there was a mortality reduction associated with SA (hazard ratio, 0.57; 95% CI, 0.52-0.62; P < .001). After propensity matching, 2750 pairs with similar baseline characteristics were identified. SA was associated with 16% mortality decline (hazard ratio, 0.84; 95% CI, 0.75-0.94; P = .003).

Conclusions: In this multicenter, retrospective, propensity matched study, SA concomitant with other cardiac surgery was associated with improved long-term survival regardless of baseline surgical risk.
