Figure 4. Four sequential positions of a male (1-4) during the successful courtship of a female Paraphidippus aurantius (Lucas 1833) on a plant in the laboratory. Males were placed on plants with three recently molted females, all from Greenville County, South Carolina. In each case the female stopped moving soon after she sighted the male, and the male advanced to mate successfully when she did not turn to face him as he stepped from side to side. In this example the female stopped moving at a distance of about 10 cm from the approaching male. The subsequent mating included insertion of each pedipalp on the respective side of the epigynum, and occupied more than 20 minutes. P. aurantius males will cohabit with and defend penultimate females (Thurlow 2016), and their large chelicerae are associated with the ritual male-male combat (agonistic behavior) that may ensue. This represents a pattern seen in many other salticids (e.g., Lyssomanes viridis, Tedore & Johnsen 2012, 2013, 2015), in which male-male contests account for most sexual selection. At the other end of the spectrum of sexual selection lie the highly ornamented salticids of the genus Maratus, for which male-male contests are virtually unknown (save one species), and a female may examine the details of male ornamentation up-close before acceptance (Otto & Hill 2021). Most salticids appear to fall somewhere between these two extremes, relying to some extent on both male-male combat and active selection by females.

The strength and form of natural selection on transcript abundance in the wild

<p>Gene transcription variation is known to contribute to disease susceptibility and adaptation, but we currently know very little about how contemporary natural selection shapes transcript abundance. Here, we propose a novel analytical framework to quantify the strength and form of ongoing natural selection at the transcriptome level in a wild vertebrate. We estimated selection on transcript abundance in a cohort of a wild salmonid fish (<i>Salmo trutta</i>) affected by an extracellular myxozoan parasite (<i>Tetracapsuloides bryosalmonae</i>) through mark-recapture field sampling and the integration of RNA-seq with classical regression-based selection analysis. We show, based on fin transcriptomes of the host, that infection by the parasite and subsequent host survival is linked to upregulation of mitotic cell cycle process. We also detect a <span class="Heading3Char">widespread signal of disruptive selection</span> on transcripts linked to host immune defence, host-pathogen interactions, cellular repair and maintenance. Our results provide insights about how selection can be measured at the transcriptome level to dissect the molecular mechanisms of contemporary natural selection driven by climate change and emerging anthropogenic threats. We anticipate that the approach described here will enable critical information on the molecular mechanisms and targets of natural selection to be obtained in real time.</p>

Dcp2 C-terminal cis-binding elements control selective targeting of the decapping enzyme by forming distinct decapping complexes

<p>A single Dcp1-Dcp2 decapping enzyme targets diverse classes of yeast mRNAs for decapping-dependent 5' to 3' decay, but the molecular mechanisms controlling selective mRNA targeting by the enzyme remain elusive. Through extensive genetic analyses we uncover <i>cis</i>-regulatory elements in the Dcp2 C-terminal domain that control selective targeting of the decapping enzyme by forming distinct decapping complexes. Two Upf1-binding motifs target the decapping enzyme to NMD substrates, and a single Edc3-binding motif targets both Edc3 and Dhh1 substrates. Pat1-binding leucine-rich motifs target Edc3 and Dhh1 substrates under selective conditions. Although it functions as a unique targeting component of specific complexes, Edc3 is a common component of multiple complexes. Xrn1 also has a specific Dcp2 binding site, allowing it to be directly recruited to decapping complexes. Collectively, our results demonstrate that Upf1, Edc3, and Pat1 function as regulatory subunits of the holo-decapping enzyme, controlling both its targeting specificity and enzymatic activation</p>

Microbial symbionts are instrumental to the ecological and long-term evolutionary success of their hosts, and the central role of symbiotic interactions is increasingly recognized across the vast majority of life. Lichens provide an iconic group for investigating patterns in species interactions; however, relationships among lichen symbionts are often masked by uncertain species boundaries or an inability to reliably identify symbionts. The species-rich lichen-forming fungal family Parmeliaceae provides a diverse group for assessing patterns of interactions of algal symbionts, and our study addresses patterns of lichen symbiont interactions at the largest geographic and taxonomic scales attempted to date. We analysed a total of 2356 algal internal transcribed spacer (ITS) region sequences collected from lichens representing ten mycobiont genera in Parmeliaceae, two genera in Lecanoraceae and 26 cultured Trebouxia strains. Algal ITS sequences were grouped into operational taxonomic units (OTUs); we attempted to validate the evolutionary independence of a subset of the inferred OTUs using chloroplast and mitochondrial loci. We explored the patterns of symbiont interactions in these lichens based on ecogeographic distributions and mycobiont taxonomy. We found high levels of undescribed diversity in Trebouxia, broad distributions across distinct ecoregions for many photobiont OTUs and varying levels of mycobiont selectivity and specificity towards the photobiont. Based on these results, we conclude that fungal specificity and selectivity for algal partners play a major role in determining lichen partnerships, potentially superseding ecology, at least at the ecogeographic scale investigated here. To facilitate effective communication and consistency across future studies, we propose a provisional naming system for Trebouxia photobionts and provide representative sequences for each OTU circumscribed in this study.
