The different environments that humans experience are likely to impact physiology and disease susceptibility. In order to estimate the magnitude of the impact of environment on transcript abundance, we examined gene expression in peripheral blood leukocyte samples from 46 desert nomadic, mountain agrarian and coastal urban Moroccan Amazigh individuals. Despite great expression heterogeneity in humans, as much as one third of the leukocyte transcriptome was found to be associated with differences among regions. Genome-wide polymorphism analysis indicates that genetic differentiation in the total sample is limited and is unlikely to explain the expression divergence. Methylation profiling of 1,505 CpG sites suggests limited contribution of methylation to the observed differences in gene expression. Genetic network analysis further implies that specific aspects of immune function are strongly affected by regional factors and may influence susceptibility to respiratory and inflammatory disease. Our results show a strong genome-wide gene expression signature of regional population differences that presumably include lifestyle, geography, and biotic factors, implying that these can play at least as great a role as genetic divergence in modulating gene expression variation in humans.
Data from: Time-keeping programme can explain seasonal dynamics of leukocyte profile in a migrant bird
It has been proposed that immune functioning in wild animals is shaped by the trade-off between a probability of encountering pathogens and an availability of resources for maintaining immune system in active state. Due to resources' seasonality one can expect that immune functioning, e.g., absolute and relative counts of white blood cells, WBC (leukocyte profile) also follows an annual cycle. However, dynamics of the seasonal changes of leukocyte profile are controversial and specific inquiries so far addressed only a portion of annual cycle. To study the seasonal dynamics of the leukocyte profile and to test its possible endogenous basis we experimentally simulated annual cycle in a migratory passerine bird, chiffchaff (Phylloscopus collybita abietinus). We report here a clear seasonal pattern of leukocyte profile in that species. The highest WBC counts were observed during pre-alternate moult and the lowest during the subsequent spring migration; the seasonal dynamics of HL ratio showed the opposite tendency. Surprisingly, HL ratios during autumn migration were relatively low, indicating little to no stress. Observed seasonal changes in controlled experimental conditions suggest an existence of time-keeping programme underlying these variations. Additionally we found negative relationship between the body condition index and WBC counts and positive relationship with HL (heterophil to lymphocyte) ratio; these findings are controversial with the data obtained with other bird species in wild populations. Understanding the origins of variation of leukocyte profile per se and in relation with other parameters of physiological condition can be a useful tool in studying physiological response to environmental changes.
Leukocyte proportions in Pteropus alecto blood smears
<p>The black flying fox (<em>Pteropus alecto</em>) is a natural reservoir for Hendra virus, a paramyxovirus that causes fatal infections in humans and horses in Australia. Increased excretion of Hendra virus by flying foxes has been hypothesized to be associated with physiological or energetic stress in the reservoir hosts. The objective of this study was to explore the leukocyte profiles of wild-caught <em>P. alecto</em>, with a focus on describing the morphology of each cell type to facilitate identification for clinical purposes and future virus spillover research. To this end, we have created an atlas of images displaying the commonly observed morphological variations across each cell type. We provide quantitative and morphological information regarding the leukocyte profiles in bats captured at two roost sites located in Redcliffe and Toowoomba, Queensland, Australia, over the course of two years. We examined the morphology of leukocytes, platelets, and erythrocytes of <em>P. alecto</em> using cytochemical staining and characterization of blood films through light microscopy. Leukocyte profiles were broadly consistent with previous studies of <em>P. alecto</em> and other Pteropus species. A small proportion of individual samples presented evidence of hemoparasitic infection or leukocyte morphological traits that are relevant for future research on bat health, including unique large granular lymphocytes. Considering hematology is done by visual inspection of blood smears, examples of the varied cell morphologies are included as a visual guide. To the best of our knowledge, this study provides the first qualitative assessment of <em>P. alecto</em> leukocytes, as well as the first set of published hematology reference images for this species. </p>

Data from: Leukocyte profiles are associated with longevity and survival, but not migratory effort: a comparative analysis of shorebirds
1. Assessment of leukocyte profiles has become an increasingly popular tool in the fields of ecology and ecophysiology. The ratio of heterophils to lymphocytes (H/L ratio) is of special utility, as it reflects physiological adaptation of an organism to cope either with an infection through injury (via heterophils) or a communicable disease (via lymphocytes). Thus, elevated H/L ratios may constitute an adaptation to risky environments (i.e. associated with high risk of injury). While intra-population variation in avian H/L ratio has been extensively studied, we are aware of no studies that linked this trait to life-history components at the inter-specific level. 2. We measured H/L ratio in over 400 shorebirds from 19 species during their autumn migration through Central Europe. Phylogenetically informed comparative methods were used to test whether H/L ratio was related to: i) annual survival and lifespan; ii) migratory effort, as measured with total migration distance and migratory energy reserves; and iii) confounding variables such as body size or breeding latitude. 3. A relatively strong phylogenetic signal and spatial phylogenetic autocorrelation indicated that most diversification in shorebird H/L ratios occurred relatively early in radiation of this group. 4. Comparative analysis gave strong support for the negative associations of H/L ratio with residual lifespan and annual survival. In contrast, we found no support for the effect of migratory behaviour on H/L ratios, suggesting that leukocyte profiles in shorebirds may not constitute an important physiological adaptation for long-distance migration. 5. Our study provided the first comparative evidence for a link between H/L ratios and important life-history traits in birds. Strong negative associations with annual survival and residual lifespan indicate that leukocyte profiles may form an adaptive basis for life-history strategies in birds.
Endothelial and leukocyte-derived microvesicles and cardiovascular risk after stroke: PROSCIS-B
<p><strong>Objective:</strong> To determine the role of circulating endothelial microvesicles (EMV) and microvesicles (MV) of other origins on long-term cardiovascular outcomes after stroke, we measured them in a cohort of first-ever stroke patients and observed them for three years.</p> <p><strong>Methods:</strong> In the PROSpective Cohort with Incident Stroke Berlin (PROSCIS-B), patients with first-ever ischemic stroke were followed for three years. The primary combined endpoint consisted of recurrent stroke, myocardial infarction, and all-cause mortality. Levels of EMV, leukocyte-derived MV (LMV), monocytic MV (MMV), and platelet-derived MV (PMV) were measured in citrate blood using flow cytometry. Kaplan-Meier curves and Cox proportional hazards models were used to estimate the effect of MV levels on the combined endpoint after adjustment confounding.</p> <p><strong>Results: </strong>571 patients were recruited (median age 69y; 39% female; median NIHSS 2, interquartile range 1-4). During the follow-up, 95 endpoints occurred. Patients with levels of EMV [adjusted hazard ratio (HR)=2.5, 95% confidence interval (CI) 1.2-4.9] or LMV (HR=3.1, 95%CI 1.4-6.8) in the highest quartile were more likely to experience an event than participants with lower levels using the lowest quartile as reference category. The association was less pronounced for PMV (HR=1.7, 95%CI 0.9-3.2) and absent for MMV (HR=1.1, 95%CI 0.6-1.8).</p> <p><strong>Conclusion:</strong> High levels of EMV and LMV after ischemic stroke were associated with worse cardiovascular outcome within three years. These results reinforce that endothelial dysfunction and vascular inflammation affect the long-term prognosis after stroke. EMV and LMV might play a potential role in risk prediction for stroke patients.</p>
