Data for: Effects of long-term ethanol storage of blood samples on the estimation of telomere length

<p>Telomeres, DNA structures located at the end of eukaryotic chromosomes, shorten with each cellular cycle. The shortening rate is affected by factors associated with stress, and, thus telomere length has been used as a biomarker of ageing, disease, and different life history trade-offs. Telomere research has received much attention in the last decades; however, there is still a wide variety of factors that may affect telomere measurements and to date no study has thoroughly evaluated the possible long-term effect of a storage medium on telomere measurements. In this study we evaluated the long-term effects of ethanol on relative telomere length (RTL) measured by qPCR, using blood samples of magpies collected over twelve years and stored in absolute ethanol at room temperature. We firstly tested whether storage time had an effect on RTL and secondly we modelled the effect of time of storage (from 1 to 12 years) in differences in RTL from DNA extracted twice in consecutive years from the same blood sample. We also tested whether individual amplification efficiencies were influenced by storage time, and whether this could affect our results. Our study provides evidence of an effect of storage time on telomere length measurements. Importantly, this effect shows a pattern of decreasing loss of telomere sequence with storage time that stops after approximate 4 years of storage, which suggests that telomeres may degrade in blood samples stored in ethanol. Our method to quantify the effect of storage time could be used to evaluate other storage buffers and methods. Our results highlight the need to evaluate the long-term effects of storage on telomere measurements, particularly in long-term studies.</p>

Alien plant fitness is limited by functional trade-offs rather than a long-term increase in competitive effects of native communities

<p><span>Alien plants experience novel abiotic conditions and interactions with native communities in the introduced area. Intra- and interspecific selection on functional traits in the new environment may lead to increased population growth with time since introduction (residence time). However, selection regimes might differ depending on the invaded habitat. Additionally, in high-competition habitats, a build-up of biotic resistance of native species due to accumulation of eco-evolutionary experience to aliens over time may limit invasion success. We tested if the effect of functional traits and the population dynamics of aliens depends on interspecific competition with native plant communities. We conducted a multi-species experiment with 40 annual Asteraceae that differ in residence time in Germany. We followed their population growth in monocultures and in interspecific competition with an experienced native community (varying co-existence times between focals and community). To more robustly test our findings, we used a naïve community that never co-existed with the focals. We found that high seed mass decreased population growth in monocultures but tended to increase population growth under high interspecific competition. We found no evidence for a build-up of competition-mediated biotic resistance by the experienced community over time. Instead, population growth of the focal species was similarly inhibited by the experienced and naïve community. By</span><span> comparing the effect of experienced and naïve communities on population dynamics over two years across a large set of species with a high variation in functional traits and residence time, </span><span>this study advances the understanding of the long-term dynamics of plant invasions. I</span><span>n our study system, population growth of alien species was not limited by</span> <span>an increase of competitive effects by native communities (one aspect of biotic resistance) over time. </span><span>Instead, invasion success of alien plants may be limited because initial spread in low-competition habitats requires different traits than establishment in high-competition habitats. </span></p>

Stage-mediated priority effects and season lengths shape long-term competition dynamics

<p>The relative arrival time of species can affect their interactions and thus determine which species persist in a community. Although this phenomenon, called priority effect, is widespread in natural communities, it is unclear how it depends on the length of growing season. Using a seasonal stage-structured model, we show that differences in stages of interacting species could generate priority effects by altering the strength of stabilizing and equalizing coexistence mechanisms, changing outcomes between exclusion, coexistence, and positive frequency dependence. However, these priority effects are strongest in systems with just one or a few generations per season and diminish in systems where many overlapping generations per season dilute the importance of stage-specific interactions. Our model reveals a novel link between the number of generations in a season and the consequences of priority effects, suggesting that consequences of phenological shifts driven by climate change should depend on specific life histories of organisms.</p>

SGS-LTER Disturbance intensity and above- and belowground herbivory effects on long-term recovery of shortgrass steppe on the Central Plains Experimental Range, Nunn, Colorado, USA 1977-1990

This data package was produced by researchers working on the Shortgrass Steppe Long Term Ecological Research (SGS-LTER) Project, administered at Colorado State University. Long-term datasets and background information (proposals, reports, photographs, etc.) on the SGS-LTER project are contained in a comprehensive project collection within the Digital Collections of Colorado (http://digitool.library.colostate.edu/R/?func=collections&collection_id=3429). The data table and associated metadata document, which is generated in Ecological Metadata Language, may be available through other repositories serving the ecological research community and represent components of the larger SGS-LTER project collection. Additional information and referenced materials can be found: http://hdl.handle.net/10217/83444 The importance of disturbance intensity and herbivory by cattle and white grubs, or the larvae of June beetles to recovery of shortgrass steppe ecosystems in Colorado, USA were evaluated over a 14 year time period. Disturbance intensity was defined by survival of the dominant grass species (Bouteloua gracilis) after an outbreak of root feeding activity by white grubs. 16 patches of vegetation consisting of four pairs of adjacent ungrazed-grazed by cattle locations with 2 replicates that were recently affected by white grubs were selected in 1977. Disturbance intensity was determined in 1977 by the area in each patch that contained live tillers of B. gracilis. Permanent plots were lcoated both within and outside of each patch. Plant basal cover and density by species were estimated at time of peak aboveground biomass in 6 different years on each plot. Successional dynamics on patches was similar to areas affected by other types of disturbances, however, rate of recovery was faster for patches affected by grubs. Grazing by cattle was infrequently important to plant recovery, a result similar to effects of grazing on other aspects of shortgrass steppe. Disturbance intensity wa
