Inflammatory responses induced by the monophasic variant of Salmonella typhimurium in pigs play a role in the high shedder phenotype and fecal microbiota composition
<p>Pigs infected with <em><span>Salmonella</span></em> may excrete large amounts of <em><span>Salmonella</span></em>, increasing the risk of spread of this pathogen in the food chain. Identifying <em><span>Salmonella</span></em> high shedder pigs is therefore required to mitigate this risk. We analyzed immune-associated markers and composition of the gut microbiota in specific-pathogen-free pigs presenting different shedding levels after an oral infection with <span>Salmonella</span>. Immune response was studied through total blood cell counts, production of anti-<em><span>Salmonella</span></em> antibodies and cytokines, and gene expression quantification. Total <em><span>Salmonella</span></em> shedding for each pig was estimated and hierarchical clustering was used to cluster pigs into high, intermediate, and low shedders. Gut microbiota compositions were assessed using 16S rRNA microbial community profiling. Comparisons were made between control and inoculated pigs, then between high and low shedders pigs. Prior to infection, high shedders had similar immunological profiles compared to low shedders. As soon as 1 day postinoculation (dpi), significant differences in the cytokine production level and on the expression level of several host genes related to a proinflammatory response were observed between high and low shedders. Infection with <em><span>Salmonella</span></em> induced an early and profound remodeling of the immune response in all pigs, but the intensity of the response was stronger in high shedders. In contrast, low shedders seroconverted earlier than high shedders. Just after induction of the proinflammatory response (at 2 dpi), some taxa of the fecal microbiota were specific to the shedding phenotypes. This was related to the enrichment of several functional pathways related to anaerobic respiration in high shedders. In conclusion, our data show that the immune response to <em><span>Salmonella</span></em> modifies the fecal microbiota and subsequently could be responsible for shedding phenotypes. Influencing the gut microbiota and reducing intestinal inflammation could be a strategy for preventing <em><span>Salmonella</span></em> high shedding in livestock.</p>
Upconversion FRET quantitation: the role of donor photoexcitation mode and compositional architecture on the decay and intensity based responses
<p>Abstract</p> <p>Lanthanide-doped colloidal nanoparticles capable of photon upconversion (UC) offer long luminescence lifetimes, narrowband absorption and emission spectra, and efficient anti-Stokes emission. These features are highly advantageous for Förster Resonance Energy Transfer (FRET) based detection. Upconverting nanoparticles (UCNPs) as donors may solve the existing problems of molecular FRET systems, such as photobleaching and limitations in quantitative analysis, but these new labels also bring new challenges. Here we have studied the impact of the core-shell compositional architecture of upconverting nanoparticle donors and the mode of photoexcitation on the performance of UC-FRET from UCNPs to Rose Bengal (RB) molecular acceptor. We have quantitatively compared luminescence rise and decay kinetics of Er<sup>3+</sup> emission using core-only NaYF<sub>4</sub>: 20% Yb, 2% Er and core-shell NaYF<sub>4</sub>: 20% Yb @ NaYF<sub>4</sub>: 20% Yb, 5% Er donor UCNPs under three photoexcitation schemes: (1) direct short-pulse photoexcitation of Er<sup>3+</sup> at 520 nm; indirect photoexcitation of Er<sup>3+</sup> through Yb<sup>3+</sup> sensitizer with (2) 980 nm short (5–7 ns) or (3) 980 nm long (4 ms) laser pulses. The donor luminescence kinetics and steady-state emission spectra differed between the UCNP architectures and excitation schemes. Aiming for highly sensitive kinetic upconversion FRET-based biomolecular assays, the experimental results underline the complexity of the excitation and energy-migration mechanisms affecting the Er<sup>3+</sup> donor responses and suggest ways to optimize the photoexcitation scheme and the architecture of the UCNPs used as luminescent donors.</p>

Expanded response data from Perrin et al. - Contrasts in Public Perception of the Role of Alien Species in Climate Driven Species Turnover
<p>Responses given by 30 respondents from 3 different interest groups on their attitudes to climate change driven species turnover in Norwegian freshwater lakes.</p> <p>The groups are a) recreational fishers, b) individuals responsible for management decisions regarding freshwater lakes or rivers and c) academics involved in research regarding freshwater lakes or rivers.</p> <p>Use of square brackets indicates details have been altered so as to preserve anonymity of respondents.</p> <p>A total of 30 interviews were conducted between August of 2019 and April of 2020. Interviews lasted anywhere from 10 to 50 minutes. Of the 30 interviews, 16 were conducted in person, with the remaining 14 conducted via web meeting. Interview respondents were chosen using the snowball method, as described by Miles and Huberman <a href="https://paperpile.com/c/6PEtIR/dSUU/?noauthor=1">(1994)</a>. This requires an initial pool of contacts, who subsequently nominate other respondents that are suitable for the study. </p> <p>In compliance with requirements of the Norwegian National Research Ethics Committee, all respondents were given an overview of the topic beforehand, assured that their responses would be anonymous, and informed of the intended use of their responses. All interviews were anonymously recorded and subsequently transcribed verbatim. Any details which might have allowed the individuals to be identified based on descriptions of their roles or locations were removed. As interviews were anonymous, ethics approval from the Norwegian National Research Ethics Committee was not required.</p>
