Anaerobic Methane Oxidation is Quantitatively Important in Deeper Peat Layers of Boreal Peatlands: Evidence from in situ Stable Isotopes Depth Profiles, Anaerobic Incubations, and Microbial Communities

<p>Dataset contains complete result of laboratory anaerobic incubations with peat samples from 3 West Siberian peatlands.</p> <p>Before the incubation, the peat samples were thoroughly mixed to ensure homogeneity. Aliquots (140 &plusmn; 1 g) were mixed with distilled water at a ratio of 1:2 by weight, and then placed into sterile 500 ml glass bottles, flushed with pure argon (99.9999%, Voessen, Russia) for 5 min to remove any oxygen and sealed with butyl rubber septa to maintain anaerobic conditions. The bottles were kept at +5&deg;C for 1 day to allow the equilibration between the peat and the headspace. Then bottles were again thoroughly flushed with argon for 5 min, sealed, and an additional 5 ml of argon was added to prevent air diffusion into the bottle headspace. The incubation was performed in two different ways: i) unamended control, and ii) amended with 10 ml of CH3F to inhibit acetotrophic methanogenesis. Peat samples from the depths of 15-20 and 40-50 cm were incubated at 15&deg; and 10&deg;C, respectively, for a period of 60 days, whereas deeper peat samples were incubated at 5&deg;С, representing the temperature of the deeper peat layers of Mukhrino bog during the snow-free period for 150 days. Gas (1 mL for Н2, CH4 and CO2 concentration, 1 mL for stable isotope compositions) and liquid (1 mL for organic acids) samples were taken for analysis every two weeks after manually shaking the bottles for approximately 5 s to equilibrate the gaseous and aqueous phases. At the end of the incubation, methane headspace concentrations ranged from 1 to 3 % for deeper samples. The incubations with and without CH3F addition were carried out in three replicates for samples from Mukhrino bog and in two replicates for Chistoe and Lempino bogs. Net methane and CO2 production were calculated from the gas concentrations, the volume of the gas space, and the water volume using the ideal gas law. Gas solubility was calculated using Henry&rsquo;s law. The reported net methane and CO2 production are the averages of 2-3 replicates.</p> <p>See further details in a paper with the same title and the first author.</p>

Effects of genotype and host environment on the cuticular hydrocarbon profiles of Lysiphlebus parasitoids and aggression by aphid-tending ants

Parasitoids in the genus Lysiphlebus specialize on ant-tended aphids and have previously been reported to mimic the CHC profiles of their aphid hosts to avoid detection by ants. However, the precise mechanisms that mediate reduced ant aggression toward Lysiphlebus spp. are not known, nor is it clear whether such mechanisms are broadly effective or specialized to particular aphid hosts. Here we explore the effects of wasp genotype and host environment on Lysiphlebus cuticular hydrocarbon (CHC) profiles and ant aggression. Rearing asexual Lysiphlebus lines in different host aphid environments revealed effects of both wasp line and aphid host on wasp CHCs. However, variation in genotype and host affected different features of the CHC profile, with wasp genotype explaining most variation in linear and long-chain methyl alkanes, while aphid host environment primarily influenced short-chain methyl alkanes. Subsequent behavioral experiments revealed no effects of host environment on ant aggression, but stronger evidence for genotypic effects. The presence of genotypic variation in experienced ant aggression and relevant chemical traits is particularly relevant in light of recent evidence for genetic divergence among Lysiphlebus parasitoids collected from different aphid hosts.

Metabolomic Profile of Cerebral Tissue After Blood-Brain Barrier Opening using Microbubble-Assisted Ultrasound: A Focus on Contralateral Side.

<p>Microbubble (MB)-assisted ultrasound (US) is an innovative modality for the non-invasive, targeted and efficient delivery of the therapeutic molecules into the brain. Previously, we reported the first metabolomic signature of blood-brain barrier opening (BBBO) induced by MB-assisted US. In the present study, the neurometabolic consequences of acoustically mediated BBBO on cerebral tissue using multimodal metabolomics approaches. Sinusoid US waves (1 MHz, peak negative pressure 0.6 MPa, burst length 10 ms, total treatment time 30 s, MB bolus dose 0.7 105 MBs/g) were applied on the right striatum (ipsilateral side). Brain was collected and both striata were then dissected 3 h, 2 days and 1 week after BBBO. &nbsp; After tissue preparation, the samples were analyzed using nuclear magnetic resonance spectrometry (NMRS) and high-performance liquid chromatography coupled to mass spectrometry (HPLC-MS). Our findings showed a slight disruption of metabolic pathways in contralateral striata of animals. Analyses of metabolic pathways indicated change of amino acid metabolisms. In addition, tryptophan derivate dosages revealed the perturbation of a central metabolite of the kynurenin pathway (<em>i.e.</em>, 3-hydroxykynurenin). In conclusion, the acoustically mediated BBBO of the ipsilateral cerebral hemisphere induced significant change in metabolism of contralateral one.</p>

mTAGs: taxonomic profiling using degenerate consensus reference sequences of ribosomal RNA gene

<p>mTAGs is a tool for the taxonomic profiling of metagenomes. It detects sequencing reads belonging to the small subunit of the ribosomal RNA (SSU-rRNA) gene and annotates them through the alignment to full-length degenerate consensus SSU-rRNA reference sequences. The tool is capable of processing single-end and pair-end metagenomic reads, takes advantage of the information contained in any region of the SSU-rRNA gene and provides relative abundance profiles at multiple taxonomic ranks (Domain, Phylum, Class, Order, Family, Genus and OTUs defined at a 97% sequence identity cutoff).</p>
