Effects of the treatments by synthetic strigolactone (SL) and SL-enriched biostimulant on W/NUE under water and phosphate deprived conditions

<p>The aim of this study is to investigate on the physiological and agronomic effects caused from the exogenous SL treatment on tomato plants (<em>Solanum lycopersicum </em>L) grown under water and phosphate stress conditions. The experimental proofs, began in 2018, were performed in greenhouses sited in Grugliasco at University of Turin, DISAFA (Italy). The analysis of the data obtained (only partially completed for the second experimental test) allowed to estimate the W/NUE of each treatment in the different water and nutrient conditions. These preliminary results seem to confirm an active role of the strigolactones as key signal mediator in response to the abiotic stresses. Analogously also the innovative biostimulant SL-enriched proposed by StrigoLab was shown similar positive activity by proposing itself as promising commercial product.</p>

<p>This record contains raw data related to the article &ldquo;Nicotinic Acid Adenine Dinucleotide Phosphate Induces Intracellular Ca<sup>2+</sup>&nbsp;Signalling and Stimulates Proliferation in Human Cardiac Mesenchymal Stromal Cells&quot;</p> <p>Abstract</p> <p>Nicotinic acid adenine dinucleotide phosphate (NAADP) is a newly discovered second messenger that gates two pore channels 1 (TPC1) and 2 (TPC2) to elicit endo-lysosomal (EL) Ca<sup>2+</sup>&nbsp;release. NAADP-induced lysosomal Ca<sup>2+</sup>&nbsp;release may be amplified by the endoplasmic reticulum (ER) through the Ca<sup>2+</sup>-induced Ca<sup>2+</sup>&nbsp;release (CICR) mechanism. NAADP-induced intracellular Ca<sup>2+</sup>&nbsp;signals were shown to modulate a growing number of functions in the cardiovascular system, but their occurrence and role in cardiac mesenchymal stromal cells (C-MSCs) is still unknown. Herein, we found that exogenous delivery of NAADP-AM induced a robust Ca<sup>2+</sup>&nbsp;signal that was abolished by disrupting the lysosomal Ca<sup>2+</sup>&nbsp;store with Gly-Phe &beta;-naphthylamide, nigericin, and bafilomycin A1, and blocking TPC1 and TPC2, that are both expressed at protein level in C-MSCs. Furthermore, NAADP-induced EL Ca<sup>2+</sup>&nbsp;release resulted in the Ca<sup>2+</sup>-dependent recruitment of ER-embedded InsP<sub>3</sub>Rs and SOCE activation. Transmission electron microscopy revealed clearly visible membrane contact sites between lysosome and ER membranes, which are predicted to provide the sub-cellular framework for lysosomal Ca<sup>2+</sup>&nbsp;to recruit ER-embedded InsP<sub>3</sub>Rs through CICR. NAADP-induced EL Ca<sup>2+</sup>&nbsp;mobilization via EL TPC was found to trigger the intracellular Ca<sup>2+</sup>&nbsp;signals whereby Fetal Bovine Serum (FBS) induces C-MSC proliferation. Furthermore, NAADP-evoked Ca<sup>2+</sup>&nbsp;release was required to mediate FBS-induced extracellular signal-regulated kinase (ERK), but not Akt, phosphorylation in C-MSCs. These finding support the notion that NAADP-induced TPC activation could be targeted to boost proliferation in C-MSCs and pave the way for future studies assessing whether aberrant NAADP signaling in C-MSCs could be involved in cardiac disorders.</p>

Malignant transformation-induced alterations in bis(monoacylglycerol)phosphate (BMP)

<p>Bis(monoacylglycero)phosphates (BMP), a lipid class highly enriched within endolysosomal organelles, fulfills critical roles in lysosome function and stability. BMPs are required for proper formation of intraluminal lysosomal structures, preserving membrane integrity by facilitating the activation of lipid metabolizing enzymes. While BMPs are largely understudied in comparison to other phospholipid classes, recent reports associate BMP dysregulation with a variety of pathological states including neurodegenerative diseases and lysosomal storage disorders. Considering that the lysosome is extensively remodeled in the process of malignant transformation, we sought to investigate BMP alterations in cancer. In this study, we identified and quantified BMP species across <em>in vitro</em> and <em>in vivo</em> models of breast cancer and comparative non-transformed cells and tissues and report striking cancer-selective changes in overall BMP abundance and molecular composition. Specifically, we observed a consistent enrichment of 22:6 fatty acyl chain-containing BMP species in both human- and mouse-derived mammary tumorigenesis models, suggesting a distinct biological role for BMP in transformation. Indeed, functional analyses uncovered a potential role for 22:6 BMPs as reactive oxygen species scavengers in tumor cells, which exhibit elevated basal levels of oxidant stress relative to non-transformed cells. Our findings reveal the molecular diversity of BMP lipids in breast tumor and non-transformed tissue models, and contribute to a growing body of evidence implicating BMPs in physiological and disease processes.</p>
