Critical PO2 as a diagnostic biomarker for the effects of low-oxygen modified and controlled atmospheres on phytosanitary irradiation treatments in the Cabbage Looper Trichoplusia ni (Hübner)

BACKGROUND: Phytosanitary irradiation is a sustainable alternative to chemical fumigants for disinfesting fresh commodities from insect pests. However, irradiating insects in modified atmospheres with very low oxygen (<1 kPa O2) has repeatedly been shown to increase radioprotective response. Thus, there is a concern that modified atmosphere packaging could reduce the efficacy of phytosanitary irradiation. One hurdle slowing the widespread application of phytosanitary irradiation is a lack of knowledge about how moderate levels of hypoxia relevant to modified atmosphere packaging of most fresh commodities (3-10 kPa O2) may affect phytosanitary irradiation treatments. Therefore, we hypothesize that critical PO2 (Pcrit), the level of oxygen at which an insect's metabolism becomes impaired, can be used as a diagnostic biomarker to predict the induction of a radioprotective response. RESULTS: Using the cabbage looper Trichoplusia ni (Hübner), we show that there is a substantial increase in radiation resistance when larvae are irradiated in atmospheres more hypoxic than their Pcrit (3.3 kPa O2). These data are consistent with our hypothesis that Pcrit could be used as a diagnostic biomarker for what levels of hypoxia may induce radioprotective effects that could impact phytosanitary irradiation treatments. CONCLUSION: We propose that the relationship between Pcrit and radioprotective effects could allow us to build a framework for predicting the effects of low-oxygen atmospheres on the efficacy of phytosanitary irradiation. However, more widespread studies across pest species are still needed to test the generality of this idea.

WRF-Solar AOD550 & Clear-Sky Irradiance Forecasts & Verifying Observations over CONUS

This dataset is fully described in and is a companion of Lee et al. (2022):

Lee, J. A., P. A. Jiménez, J. Dudhia, and Y.-M. Saint-Drenan, 2023: Impacts of the aerosol representation in WRF-Solar clear-sky irradiance forecasts over CONUS. J. Appl. Meteor. Climatol., 62, 227–250, https://doi.org/10.1175/JAMC-D-22-0059.1.

Aerosol optical depth (AOD) is a primary source of solar irradiance forecast error in clear-sky conditions. Improving the accuracy of AOD in NWP models like WRF will thus reduce error in both direct normal irradiance (DNI) and global horizontal irradiance (GHI), which should improve solar power forecast errors, at least in cloud-free conditions. In this study clear-sky GHI and DNI was analyzed from four configurations of the WRF-Solar model with different aerosol representations: 1) The default Tegen climatology; 2) Imposing AOD forecasts from the GEOS-5 model; 3) Imposing AOD forecasts from the Copernicus Atmosphere Monitoring Service (CAMS) model; and 4) The Thompson-Eidhammer aerosol-aware water/ice-friendly aerosol climatology. Over eight months of these 15-min output forecasts are compared against high-quality irradiance observations at NOAA SURFRAD and Solar Radiation (SOLRAD) stations located across CONUS. In general, WRF-Solar with GEOS-5 AOD had the lowest errors in clear-sky DNI, while WRF-Solar with CAMS AOD had the highest errors, higher even than the two aerosol climatologies, which is consistent with validation of the four AOD550 datasets against AERONET stations. For clear-sky GHI, the statistics differed little between the four models, as expected due to the lesser sensitivity of GHI to aerosol loading. Hourly-average clear-sky DNI and GHI was also analyzed, and additionally compared with CAMS model output directly. CAMS irradiance performed competitively with the best WRF-Solar configuration (with GEOS-5 AOD). The markedly different performance of CAMS versus WRF-Solar with CAMS AOD indicates that CAMS is apparently less sensitive to AOD550 than WRF-Solar is.

wrf_aeronet_aod550_by_cycle_20191119-20200730.nc:

NetCDF, 317 MB

WRF-Solar AOD550 spatially interpolated to AERONET stations and AERONET observed AOD550. PAK3 downregulation induces cognitive impairment following cranial irradiation

Cranial irradiation is used for prophylactic brain radiotherapy as well as treatment of primary brain tumors. Despite its high efficiency, it often induces unexpected side effects, including cognitive dysfunction. Herein, we observed that mice exposed to cranial irradiation exhibited cognitive dysfunction, including altered spontaneous behavior, decreased spatial memory, and reduced novel object recognition. Analysis of actin cytoskeleton revealed that ionizing radiation (IR) disrupted the filamentous/globular actin (F/G-actin) ratio and downregulated the actin turnover signaling pathway p21-activated kinase 3 (PAK3)-LIM kinase 1 (LIMK1)-cofilin. Furthermore, we found that IR could upregulate microRNA-206-3p (miR-206-3p) targeting PAK3. As the inhibition of miR-206-3p through antagonist (antagomiR), IR-induced disruption of PAK3 signaling is restored. In addition, intranasal administration of antagomiR-206-3p recovered IR-induced cognitive impairment in mice. Our results suggest that cranial irradiation-induced cognitive impairment could be ameliorated by regulating PAK3 through antagomiR-206-3p, thereby affording a promising strategy for protecting cognitive function during cranial irradiation, and promoting quality of life in patients with radiation therapy.
