Supplemental Material: High serological barriers may contribute to restricted Influenza-A-virus transmission between pigs and humans

Human-to-swine (reverse zoonotic) transmission of seasonal and pandemic human influenza A viruses (IAV) to pigs primarily replenishes the vast reservoir of genetically and antigenically heterogeneous swine (sw) IAV maintained in domestic pigs worldwide. Sporadic but regularly observed cases of pig-to-human (zoonotic) infections with swIAV tend to be discovered by chance, with children being affected disproportionately often.

Here, a total of 3070 porcine and 333 human nasal swab samples from 135 swine farms in Germany were investigated for IAV by real time RT-PCR and full genome sequencing. In addition, swIAV sequences generated in the frame of this study were analyzed to determine potential mutations for human MxA and BTN3A3 escape.

01_Table S1: Summary of information of swine holdings and RT-qPCR results. 1 indicates applicable; 0 indicates not applicable; n.d. indicates not determined.

02_Table S2: Summary of information about human samples and. 1 indicates applicable; 0 indicates not applicable; n.d. indicates not determined.

03_Table S3: A. Comparison of relevant mutations in the genome of MWP/21, swine-MWP/21, and NRW/22 generated by Flusurver (http://flusurver.bii.a-star.edu.sg). 1 indicates the presence of mutation(s), 0 indicates the absence of those mutation(s). B. Visualization of the AA differences of affected segments of the zoonotic case MWP/21 and the corresponding sequence (sw-MWP/21) generated from pigs of the related herd.

04_Table S4: Accession number (EPI_ISL) of sequences analyzed in the frame of this study. Global dissemination of Influenza A virus is driven by wild bird migration through arctic and subarctic zones

Influenza A viruses (IAV) circulate endemically among many wild aquatic bird populations that seasonally migrate between wintering grounds in southern latitudes to breeding ranges along the perimeter of the circumpolar arctic. Arctic and subarctic zones are hypothesized to serve as ecologic drivers of the intercontinental movement and reassortment of IAVs due to high densities of disparate populations of long distance migratory and native bird species present during breeding seasons. Iceland is a staging ground that connects the East Atlantic and North Atlantic American flyways, providing a unique study system for characterizing viral flow between eastern and western hemispheres. Using Bayesian phylodynamic analyses, we sought to evaluate the viral connectivity of Iceland to proximal regions and how inter-species transmission and reassortment dynamics in this region influence the geographic spread of low and highly pathogenic IAVs. Findings demonstrate that IAV movement in the arctic and subarctic follows seabird migration around the perimeter of the circumpolar north, favoring short-distance flights between proximal regions rather than long distance flights over the polar interior. Iceland connects virus movement between mainland Europe and North America, particularly due to the westward migration of wild birds from mainland Europe to Northeastern Canada and Greenland. Though virus diffusion rates were similar among avian taxonomic groups in Iceland, gulls act as recipients and not sources of IAVs to other avian hosts prior to onward migration. These data identify patterns of virus movement in northern latitudes and inform future surveillance strategies related to seasonal and emergent IAVs with pandemic potential.

Antigen-specific memory NK cell responses against HIV and influenza utilize the NKG2/HLA-E axis

For over a decade, multiple studies have disputed the notion of natural killer (NK) cells as purely innate lymphocytes by demonstrating that they are capable of putative antigen-specific immunological memory against multiple infectious agents including two critical global health priorities – human immunodeficiency virus (HIV) and influenza. However, the mechanisms underlying antigen specificity remain unknown. Herein, we demonstrate that antigen-specific human NK cell memory develops upon exposure to both HIV and influenza, unified by a conserved and epitope-specific targetable mechanism largely dependent on the activating CD94/NKG2C receptor and its ligand HLA-E, and confirm these findings by three rigorous assays. We validated the permanent acquisition of antigen-specificity by individual memory NK cells by single-cell cloning. We identified biomarkers of antigen-specific NK cell memory through complex immunophenotyping by 30-parameter flow cytometry showing elevated expression of KLRG1, a4b7, and NKG2C. Finally, we show individual HLA-E-restricted peptides that may constitute the dominant NK cell response in HIV-1- and influenza-infected persons in vivo. Our findings clarify the mechanisms behind antigen-specific memory NK cell responses, and suggest they could be targeted for future vaccines, cure strategies, or other therapeutic interventions.

Excel mapping tool for 2019 avian influenza data reporting

Data collection is an important task of the European Food Safety Authority (EFSA) and a fundamental component of risk assessment (Articles 22 and 23 of Regulation (EC) No 178/2002). EFSA receives a large volume of data from Member States (MSs) that is used in support of its risk assessment mission.

Council Directive 2005/94/EC and Commission Decision 2010/367/EU lay down the guidelines for monitoring and reporting avian influenza surveillance data on poultry and wild birds by European Union (EU) MSs. EFSA has been assigned the task of collating, validating, analysing, and summarising in an annual report the results from the avian influenza surveillance programmes in poultry and wild birds. For the reporting of data, EFSA provides a Data Collection Framework that allows data providers to submit data in eXtensible Markup Language (XML) format through a web interface or a web service. Here, a data model describing the format, and the content requested when submitting data through the DCF, is provided.

The data model supports the reporting of laboratory testing results for avian influenza in both wild birds, and poultry. The use of this reporting standard ensures that reported results are comparable between reporting countries. Data reported in this format will be used to generate epidemiological updates on the avian influenza surveillance testing carry out in Europe, and to provide scientific advice to the European Commission.

The Efficacy of Azithromycin in the Treatment of Influenza A infection: A Systematic Review

Background:

The WHO recommendation against the use of macrolides reflects the little low-quality evidence about the effect of azithromycin (AZ) on influenza A infection. Considering recent reports about AZ use despite the WHO's unfavorable recommendations, we aimed to test the evidence for the usage of AZ in the treatment of influenza A infection.

Methods:

Various online databases were searched till August 2023 and thorough text and evidence evaluation resulted in selecting 7 studies.

Results:

The selected reports included randomized control trials (two), cohort (three), case/control (one) studies, and one case report. Pooling of the results was unattainable due to the variability among studies in patient criteria, study outcomes, and study designs. Based on the limited reports with insufficient evidence, a recommendation for the use of AZ in the treatment regimen was not possible despite the associated reduction in inflammatory cytokines by AZ administration and the absence of significant adverse effects or mortality in hospital-admitted or ICU patients. While these findings support the current guidelines for the treatment of influenza A infection, the evidence behind these guidelines and treatment characteristics were illustrated.

Conclusion:

AZ shows a potential symptom improvement and pro-inflammatory laboratory indices in influenza A patients. Another positive aspect is the absence of complications associated with AZ administration to critically ill patients and acceptable prognosis. However, it is crucial to have more quality research on this matter that focuses on the indication of the use of AZ, and the timing of initiation, dosage, and duration of treatment while monitoring adverse events.
