Time-lapse 3D confocal microscopy videos of mitochondrial dynamics in human alveolar epithelial cells (A549-DsRed) infected with Mycobacterium marinum (Mmar) strains

The dataset consists of time-lapse, 3D confocal images of mitochondrial dynamics in human alveolar epithelial cells (A549-DsRed) infected with Mycobacterium marinum (Mmar) strains. Images were captured at 60X magnification in an environmental chamber at 35°C for live-cell imaging. Host cell mitochondria were labeled with red fluorescent protein (RFP) and infected with both wildtype (wt) and ESAT-6 operon knockout mutant labeled with green fluorescent protein (GFP) at MOI of 100 for 24 hours at 35°C. Infected cells were identified and analyzed to explore the effect of pathogenic mycobacteria on mitochondrial morphology over time.

More details available in this preprint: https://doi.org/10.48550/arXiv.2411.06035

Single Particle Investigation of Triolein Digestion using Optical Manipulation, Polarized Video Microscopy, and SAXS

Hypothesis: Understanding how soft colloids, such as food emulsion droplets, transform based on their environment is critical for various applications, including drug and nutrient delivery and biotechnology. However, the mechanisms behind colloidal transformations within individual oil droplets still need to be better understood.

Experiments: This study employs optical micromanipulation with microfluidics and polarized optical video microscopy to investigate the pancreatic lipase- and pH-triggered colloidal transformations in a single triolein droplet. Small-angle X-ray scattering (SAXS) provides complementary statistical insights and allows for detailed structural assignment.

Findings: Optical video microscopy recorded the transformation of individual triolein emulsion droplets, with the smooth surface of these spherical particles becoming rough and the entire volume eventually being affected. The polarized microscopy revealed the coexistence of at least two distinct structures in a single particle during digestion, with their ratio and distribution altered by pH. The SAXS analysis assigned the optical anisotropy to emulsified inverse hexagonal- and multilamellar phases, coexisting with isotropic structures such as the micellar cubic phase. These results can help understand the liquid-liquid crystalline phase transformations inside an emulsion droplet and guide the design of advanced food emulsions.

In-vivo video microscopy of the rupturing process of thin blood vessels in transparent fish during contact with a blunt indenter

To clarify the mechanism of bruise injuries caused by blunt impact, in vivo microscopy was performed. A flat ended indenter made of transparent acrylic was loaded onto the lateral side of the tail region of anesthetized fish. The process of the rupture of thin blood vessels was recorded in two specimens.

This data set includes two types of files; (1) mp4 files of the in vivo microscopy; (2) pdf files to explain the mp4 files.

The mp4 files are the original data of “Fujikawa, T., Yamada, Y. High-speed video microscopy analysis of cilia before and after airway cell culture

High-speed video microscopy analysis (HSVA) is a diagnostic tool used within the UK Primary Ciliary Dyskinesia (PCD) Service to access airway ciliary function on nasal brushing biopsies. The Southampton PCD group is based at the University of Southampton, Faculty of Medicine and the University Hospital Southampton NHS Foundation Trust and is led by Professor Jane Lucas. We also use air-liquid interface (ALI) culture to differentiate airway epithelial cells to regrow healthy cilia to repeat standard PCD tests (including HSVA, immunofluoresence labelling of cilia proteins, transmission electron microscopy and functional genomics) and provide PCD research samples, which also allow us to develop new diagnostic approaches. ALI-culture can restore normal ciliary movement when secondary damage (due to infection or poor cell health) temporarily causes of abnormal cilia movement or a lack of cilia. ALI-culture can also re-confirm when ciliary defects and abnormal ciliary function are permanent and cause by inherited PCD (a ciliopathy).

Airfuge Ultracentrifugation of Suspensions on Electron Microscopy Grids for Particle Enrichment - Video

The video shows the ultracentrifugation of suspensions to enrich particles on electron microscopy grids for negative staining electron microscopy. For this purpose, an air-driven desktop ultracentrifuge, the Airfuge, is used with a fixed-angle rotor. Electron microscopy grids are placed in particular adaptors and the suspension is directly centrifuged on the grids. The method is used for diagnostic electron microscopy of infectious pathogens in clinical samples. The protocol of the procedure is available at https://doi.org/10.5281/zenodo.12731515.
