Nano MOFs as targeted drug delivery agents to combat antibiotic resistant bacterial infections

<p>The drug resistance of bacteria is a significant threat to human civilization while the action of antibiotics against drug-resistant bacteria is severely limited due to the hydrophobic nature of drug molecules, which unquestionably inhibit its permanency for clinical applications. The antibacterial action of nanomaterials offers major modalities to combat drug resistance of bacteria. The current work reports, the use of nano MOFs encapsulating drug molecules to enhance its antibacterial activity against model drug-resistant free living bacteria and biofilm of the bacteria. We have attached rifampicin (RF), a well-documented antituberculosis drug with tremendous pharmacological significance, into the pore surface of zeolitic imidazolate framework 8 (ZIF8) by a <span><span>simple synthetic procedure</span></span><span>.</span> The synthesized ZIF8 has been characterized using X-ray diffraction (XRD) method before and after drug encapsulation. The electron microscopic strategies such as scanning electron microscope (SEM) and transmission electron microscope (TEM) methods was performed to characterize the binding between ZIF8 and RF. We have also performed picosecond resolved fluorescence spectroscopy to validate the formation of the ZIF8-RF nanohybrids (NHs). The drug release profile experiment demonstrates that ZIF8-RF depicts pH-responsive drug delivery and ideal for targeting bacterial disease corresponding to its inherent acidic nature. Most remarkably, ZIF8-RF gives enhanced antibacterial activity against methicillin-resistant <i>S. aureus</i> (MRSA) bacteria and also prompts entire damage of structurally robust bacterial biofilms. Overall, the present study depicts a detailed physical insight for manufactured antibiotic-encapsulated NHs presenting tremendous antimicrobial activity that can be beneficial for manifold practical applications.</p>

TRATAMENTO DO MELASMA COM MICROAGULHAMENTO ASSOCIADO AO DRUG DELIVERY DE ÁCIDO TRANEXÂMICO: RELATO DE CASO

<p>Introdução: Clinicamente caracterizado por máculas amarronzadas de bordas irregulares e nítidas, o melasma é um distúrbio pigmentar cutâneo muito comum, afetando principlamente áreas fotoexpostas como face e pescoço. É comumente identificado em mulheres em idade reprodutiva e frequentemente descrita em tipos de pele Fitzpatrick IV a VI. Por ser um problema inestético, o melasma pode ter impacto na qualidade de vida como fator de estresse psicossocial e constrangimento. Objetivo: Descrever o uso do microagulhamento associado ao drug delivery de ácido tranexâmico para tratamento do melasma facial. Material e métodos: Relato de caso, paciente do sexo feminino, fototipo IV, 41 anos, portadora de melasma, a qual foi selecionada para ser tratada durante dois meses com duas sessões mensais de microagulhamento associado ao drug delivery de 1,0ml de ácido tranexâmico 8mg/2ml. A paciente foi avaliada antes e após o tratamento com os seguintes parâmetros: evolução fotográfica, autoavaliação com questionário capaz de avaliar objetivamente a qualidade de vida dos paciente acometidos pelo melasma e melhora do MASI. Resultados e discussão: Em relação ao MASI, houve uma melhora significativa, com redução de 54,4 ± 7,53 para 29,9 ± 7,60 (p = 0,007) (45%) e redução não significativa de 13,1 ± 3,79 para 10,8 ± 3,79 (p=0,022) (17%) no MELASQol. Conclusão: O percentual médio de melhora após&nbsp;tratamento&nbsp;por MASI foi de 45% e por MELASQol, de 17%. Não foi encontrada associação significativa entre os percentuais de melhora do MASI e do MELASQol (p=0,3641) refletindo que mesmo obtendo boa resposta na melhora do melasma, a persistência da mancha, ainda era fator de incômodo para a paciente.&nbsp;A associação proposta apresentou resultados satisfatórios, constituindo uma modalidade terapêutica inovadora, replicável e segura para o tratamento do melasma.</p>

DNA mechanocapsules for programmable piconewton responsive drug delivery

<p>The mechanical dysregulation of cells is associated with a number of disease states, that spans from fibrosis to tumorigenesis. Hence, it is highly desirable to develop strategies to deliver drugs based on the "mechanical phenotype" of a cell. To achieve this goal, we outline and synthesize DNA mechanocapsules (DMC) comprised of DNA tetrahedrons that are force responsive. Modeling shows&nbsp;the trajectory of force-induced DMC rupture and predicts how applied force spatial position and orientation tunes the force-response threshold. DMCs functionalized with adhesion ligands mechanically denature <i>in vitro</i> as a result of cell receptor forces. &nbsp;DMCs are&nbsp;designed to encapsulate macromolecular cargos such as dextran and oligonucleotide drugs with minimal cargo leakage and high nuclease resistance. Force-induced release and uptake of DMC cargo is&nbsp;validated using&nbsp;flow cytometry. Finally, we demonstrate force-induced mRNA knockdown of HIF-1α in a manner that is dependent on the magnitude of cellular traction forces. These results show that DMCs can be effectively used to target biophysical phenotypes which may find useful applications in immunology and cancer biology.&nbsp;</p>

Resveratrol loaded chitosan-pectin core-shell nanoparticles as novel drug delivery vehicle for sustained release and improved antioxidant activities

<p><span>Resveratrol, chemically known as 3, 5, 4'-trihydroxy-trans-stilbene, is a natural polyphenol with promising multi-targeted health benefits. The optimal therapeutic uses of resveratrol are limited due to its poor solubility, rapid metabolism and low bioavailability. To address the issues, we have encapsulated resveratrol inside the nanosized core made of chitosan and coated this core with pectin - shell in order to fabricate a drug delivery vehicle which can entrap resveratrol for a longer period of time. The core - shell nanoparticles fabricated in this way were characterized with the help of Fourier Transform Infrared Spectrometer (FTIR), Field Emission Scanning Electron Microscope (FESEM), Field Emission Transmission electron microscopy/Selected Area Electron Diffraction (FETEM/SAED), High-Resolution Transmission Electron Microscope (HRTEM)), Dynamic Light Scattering (DLS) and Zeta Potential measurements. In vitro drug release study showed the ability of the core-shell nanoparticles to provide sustained release of resveratrol for almost 30 hours. The release efficiency of the drug was found to be pH dependent and a sequential control over drug release can be obtained varying the shell thickness. The resveratrol encapsulated in a nanocarrier was found to have a better in vitro antioxidant activity than free resveratrol as determined by DPPH radical scavenging method. This work finally offers a novel nano-based drug delivery system.</span></p>
