Factorial design assisted RP-HPLC method for simultaneous determination of fluconazole, itraconazole, and terbinafine

A 23 full factorial design model was utilized for development of a new HPLC method with UV detection to estimate three antifungal drugs simultaneously. Fluconazole (FLU), itraconazole (ITR) and terbinafine (TRH), which are co-administered for severe fungal infections have been determined using MOS-1 Hypersil C18 column and an isocratic eluent; methanol 95% and phosphate buffer 5% with 0.001% triethylamine. The pH adjusted 7, and flow rate of 0.7 mL/min. The three drugs were separated within less than 7 minutes at 210 nm. The developed method gave linear response over 5-80, 5-50 and 1-50 μg/mL for FLU, ITR and TRH, respectively. It showed detection limits of 0.8761, 0.293 and 0.1965 μg/mL and quantification limits of 2.655, 0.8788 and 0.5956 μg/mL for the three drugs, respectively. The design of experiment facilitated the optimization of different variables affecting the separation of the three drugs. The designed method sensitivity permitted the simultaneous estimation of ITR and TRH in spiked human plasma successfully.

Human-centred design methods to empower 'teachers as designers'

The present datasets have been used for the paper entitled "Human-centred design methods to empower ‘teachers as designers’".

Abstract

Teachers are learning designers, often unwittingly. To facilitate this role of ‘teachers as designers’, educators of all sectors need to adopt a design mindset and acquire the skills needed to address the design challenges they encounter in their everyday practice. Human-centred design (HCD) provides professional designers with the methods needed to address complex problems. It emphasises the human perspective throughout the design lifecycle and provides a practice-oriented, context-aware, empathetic and incremental approach which naturally fits educators realities.

This research reports on a MOOC designed to ‘walk’ educators through the design of an ICT-based learning activity following an HCD process and its techniques. A mixed methods approach is used to gauge how participants experienced the MOOC and the various design tasks it comprises. Although the perceived level of difficulty and value on the different methods varied - and, significant differences were seen between the experience of novice and expert educators-, the participants felt the overall approach constituted a powerful means for them to design technology-enhanced learning activities. The results support the idea of HCD as a valuable framework for educators and inform ongoing international efforts to shape a science and practice of learning design for teaching.

Data from: The relative power of genome scans to detect local adaptation depends on sampling design and statistical method
Although genome scans have become a popular approach towards understanding the genetic basis of local adaptation, the field still does not have a firm grasp on how sampling design and demographic history affect the performance of genome scans on complex landscapes. To explore these issues, we compared 20 different sampling designs in equilibrium (i.e. island model and isolation by distance) and nonequilibrium (i.e. range expansion from one or two refugia) demographic histories in spatially heterogeneous environments. We simulated spatially complex landscapes, which allowed us to exploit local maxima and minima in the environment in 'pair' and 'transect' sampling strategies. We compared FST outlier and genetic–environment association (GEA) methods for each of two approaches that control for population structure: with a covariance matrix or with latent factors. We show that while the relative power of two methods in the same category (FST or GEA) depended largely on the number of individuals sampled, overall GEA tests had higher power in the island model and FST had higher power under isolation by distance. In the refugia models, however, these methods varied in their power to detect local adaptation at weakly selected loci. At weakly selected loci, paired sampling designs had equal or higher power than transect or random designs to detect local adaptation. Our results can inform sampling designs for studies of local adaptation and have important implications for the interpretation of genome scans based on landscape data.
