<p>A tail of 3dpf transgenic zebrafish larvae (<em>tg(mpeg1:Gal4/UAS:Kaede</em>)) was imaged from 1h to 6h after the tail fin amputation by ANDOR CSU-W1 confocal spinning disk on an inverted NIKON microscope (Ti Eclipse) with ANDOR Neo sCMOS camera (20x air/NA 0.75 objective, mosaic imaging 6x1 with 10% overlap between windows). Laser excitation/emission wavelength: 488/521 nm. Time step is 4min (75 repeats). Z-stack is 132μm with 4μm step (33 steps). Maximum projection is shown as well. Pixel size is 0.326μm.</p> <p>Macrophages expressing green fluorescent protein Kaede in the cytoplasm are moving toward the wound made at the tail fin.</p>

The Effect of 10% ointment concentration of ethyl acetate subfraction of Meniran (Phyllanthus niruri L.) leaves on excision wound healing in white male rats
<p><strong>Abstract</strong></p> <p><strong>Background: </strong>Wounds are skin problems that are often experienced by humans. Effective wound healing requires a complex arrangement of various healing processes that occur continuously. The plant that has been studied to have a role in the wound healing process is the herbal extract of meniran (<em>Phyllanthus niruri</em> L). This plant can be found in almost all parts of Indonesia. However, biomedical evidence related to the effect of giving meniran (<em>Phyllanthus niruri</em> L) leaves ethyl acetate subfraction ointment preparations is still not revealed yet.</p> <p><strong>Methods: </strong>This study was carried out using experimental animals, a total of 27 rats were divided into 3 large groups, each group consisted of 9 rats, where group 1 was the control based ointment, group 2 was the comparison group (ointment T®) and group 3 was treated with 10% concentration of meniran leaf ethyl acetate subfraction ointment. Each group was observed and measured for three parameters, namely, percentage of wound healing, epithelialization time, and hydroxyproline levels.</p> <p><strong>Results: </strong>From the results of data analysis using one-way (ANOVA) followed by the duncan test (SPSS 23.0) for epithelialization time and hydroxyproline levels, the results showed that 10% concentration of meniran leaf ethyl acetate subfraction ointment, the comparison group (T® ointment) and the control based group with the treatment group on the parameters of epithelialization time and hydroxyproline have levels significant of (p <0.05)</p> <p><strong>Conclusions: </strong>It can be concluded that the ethyl acetate subfraction ointment of meniran leaves with a concentration of 10% is effective in the healing process of the excision wound.</p> <p><strong>Keywords</strong></p> <p><em>Phyllanthus niruri </em>L, wound healing , epithelialization, hidroksiprolin</p>
Morphological changes of zebrafish macrophages during wound healing
<p>Tg(mpeg1:gal4/UAS:Kaede) larvae with mosaic expression of Kaede protein in macrophages were amputated at 3 dpf and imaged using high resolution Spinning Disk microscopy between 8 to 13 h,every 2.5 min (C) Quantification of perimeter (upper graph) and circularity value (lower graph) for individual macrophages present at the wound, during a time lapse sequence from 8 hpA to 13 hpA every 2.5 min. For quantification, we used automated image analysis, the image segmentation was carried out from 8 to 13 hpA. At first, the images were filtered by using space-time filtering that keeps the temporal coherence of moving macrophages (Sarti et al., 1999). Then, the filtered images were segmented by a combination of the local Otsu method (Otsu, 1979; Saddami et al., 2019) and (Park et al., unpublished) and the subjective surface segmentation (SUBSURF) method (Sarti et al., 2000) where the binarized images from the local Otsu method were considered as an initial condition of the SUBSURF equation (Park et al., 2023). Trajectories of macrophages in segmented images were reconstructed based on automatic cell tracking (Park et al., 2023), and shape descriptors (perimeter and circularity) were measured. Demonstrator of a rotor-fed asynchronous start for a Salient Pole Wound Field Synchronous Machine
<p>The video shows the asynchronous run-up (up to ca. 620 rpm) of a four pole, 60 kVA, 400V, 50 Hz salient pole synchronous generator. The run-up is obtained by AC-supplying a special rotor winding arrangement, capable of both exciting the machine at synchronism and providing a multiphase rotating MMF during the run-up. The armature phases, which have two parallel current paths, are conveniently short-circuited during the rotor acceleration. </p>
WHiM-BC Dataset: Evolution of CSCs and non CSCs MCF-7 Migration in Wound Healing Assay

Observational Signatures of Tightly Wound Spirals Driven by Buoyancy Resonances in Protoplanetary Disks
<p>These are simulated CO data cubes presented in "Observational Signatures of Tightly Wound Spirals Driven by Buoyancy Resonances in Protoplanetary Disks" by Bae, Teague and Zhu. <p>This record contains data related to article "Evaluation of cell metabolic adaptation in wound and tumour by fluorescence Lifetime imaging Microscopy"</p> <p>Abstract</p> <p>Acidic pH occurs in acute wounds progressing to healing as consequence of a cell metabolic adaptation in response to injury-induced tissue hypoperfusion. In tumours, high metabolic rate leads to acidosis affecting cancer progression. Acidic pH affects activities of remodelling cells in vitro. The pH measurement predicts healing in pathological wounds and success of surgical treatment of burns and chronic ulcers. However, current methods are limited to skin surface or based on detection of fluorescence intensity of specific sensitive probes that suffer of microenvironment factors. Herein, we ascertained relevance in vivo of cell metabolic adaptation in skin repair by interfering with anaerobic glycolysis. Moreover, a custom-designed skin imaging chamber, 2-Photon microscopy (2PM), fluorescence lifetime imaging (FLIM) and data mapping analyses were used to correlate maps of glycolytic activity in vivo as measurement of NADH intrinsic lifetime with areas of hypoxia and acidification in models of skin injury and cancer. The method was challenged by measuring the NADH profile by interfering with anaerobic glycolysis and oxidative phosphorylation in the mitochondrial respiratory chain. Therefore, intravital NADH FLIM represents a tool for investigating cell metabolic adaptation occurring in wounds, as well as the relationship between cell metabolism and cancer.</p>
