Antibody features towards VAR2CSA and CSA binding infected erythrocytes in a cohort of pregnant women from PNG

Plasmodium falciparum causes placental malaria, which results in adverse outcomes for mother and child. P. falciparum infected erythrocytes that express the parasite protein VAR2CSA on their surface can bind to placental chondroitin sulfate-A. It has been hypothesized that naturally acquired antibodies towards VAR2CSA protect against placental infection, but it has proven difficult to use measures of antibody to identify individuals protected from disease. We used a systems serology approach to identify naturally acquired antibody features mid pregnancy that were associated with protection from placental malaria at delivery. Machine learning techniques selected six out of 169 measured antibody features towards VAR2CSA that could predict (with 86% accuracy) whether a woman would subsequently have active placental malaria infection at delivery. Selected features were associated with inhibition of placental binding and/or opsonic phagocytosis of infected erythrocytes, and network analysis indicated that there are not one but multiple pathways to protection from placental malaria.

Analysis of antibody signal loss for elongating RNA polymerase II upon transcription inhibition

This data set assesses the loss of antibody signal for elongating RNA polymerase II in zebrafish embryos upon transcription inhibition with flavopiridol. Polymerase was labeled by immunofluorescence, microscopy images were acquired by instant-SIM micrscopy, and analyzed using MatLab scripts and the bioformats importer. This data set contains the raw image data as well as all further analysis scripts.

Code related to article "The antibody response to SARS-CoV-2 infection persists over at least 8 months in symptomatic patients"

This code is related to article "The antibody response to SARS-CoV-2 infection persists over at least 8 months in symptomatic patients"

Abstract

The factors involved in the persistence of antibodies to SARS-CoV-2 are unknown. We evaluated the antibody response to SARS-CoV-2 in personnel from 10 healthcare facilities and its association with individuals’ characteristics and COVID-19 symptoms in an observational study. We enrolled 4735 subjects (corresponding to 80% of all personnel) over a period of 5 months when the spreading of the virus was drastically reduced. For each participant, we determined the rate of antibody increase or decrease over time in relation to 93 features analyzed in univariate and multivariate analyses through a machine learning approach. In individuals positive for IgG ( ≥ 12 AU/mL) at the beginning of th study, we found an increase [p= 0.0002] in antibody response in symptomatic subjects, particularly with anosmia/dysgeusia (OR 2.75, 95% CI 1.753 – 4.301), in a multivariate logistic regression analysis. This may be linked to the persistence of SARS-CoV-2 in the olfactory bulb.

Fig. 2 in Triterpene glycosides from Blighia welwitschii and evaluation of their antibody recognition capacity in multiple sclerosis

Fig. 2. Data distribution of IgM antibody responses to saponins 2, 6, 7, hederagenin monodesmoside and giganteaside J from B. welwitschii, in MS patients' sera, determined by ELISA. CSF114(Glc) and its unglucosylated analogue, CSF114, were tested as positive and negative controls, respectively. Glc oleanolate from O. obtusifolia, whose glycoside part is significantly different from B. welwitschii saponins, was tested as a reference compound. Data are reported as absorbance at 405 nm of sera diluted 1:100. Horizontal lines represent mean values with their SD. Mann-Witney U test showed that the CSF114(Glc) distribution significantly differs from this of CSF114 (p value <0.0001, two-tailed). Kruskal-Wallis test including multiple comparison of natural glycosides distributions showed statistical differences between compound 2 and 6 (p value <0.05), 2 and 7 (p value <0.0001), 2 and hederagenin monodesmoside (p value <0.01), Glc oleanolate and hederagenin monodesmoside (p value <0.05), Glc oleanolate and 7 (p value <0.0001) and Glc oleanolate and 6 (p value <0.05).

Fig. 3 in Triterpene glycosides from Blighia welwitschii and evaluation of their antibody recognition capacity in multiple sclerosis

Fig. 3. Data distribution of IgM antibody responses to saponins 2, 6, 7, hederagenin monodesmoside and giganteaside J from B. welwitschii as well as Glc oleanolate from O. obtusifolia, in the sera of multiple sclerosis (MS) and ctrl patients, determined by ELISA. CSF114(Glc) and the unglucosylated peptide CSF114 were tested as controls. Data are reported as absorbance at 405 nm of sera diluted 1:100. Horizontal lines represent mean values with their SD. MannWitney U test showed that MS and ctrl distributions significantly differ for CSF114(Glc), CSF114 (p value <0.0001, two-tailed) and for Glc oleanolate (p value <0.05, two-tailed). In contrast, saponins 2, 6, 7, hederagenin monodesmoside and giganteaside J did not show any statistical differences between the two sera distributions (p value> 0.05, two-tailed).
